Methyl 5-amino-1H-[1,2,4]triazole-3-carboxylate hydrochloride (1). and free ester (2). were obtained and 2 was reacted with Ac(2)O to give the acetylated products 3-6. Compounds 1-6 were studied using HPLC, GC-MS, FTIR and multinuclear NMR spectroscopy, including the cross-polarisation magic angle spinning (CPMAS) technique. The results of the acetylation of 2 were compared to those of the acetylation of 5-amino-1H-[1,2,4]triazole, and for 2 a significant decrease in the susceptibility to acetylation was found. The reaction of 2 with Ac(2)O at 20 degrees C, regardless of the amount and the concentration of the latter, including neat Ac(2)O, proceeds fully regioselectively and leads to one product: methyl 1-acetyl-5-amino-1H-[1,2,4]triazole-3-carboxylate (3). In sharp contrast to 5-amino-1H-[1,2,4]triazole, neither an additional monoacetylated isomer, whether annular or exocyclic, nor any diacetylated derivative could be detected. The diacetylation of 2 requires the process to be carried out in neat boiling Ac(2)O and, as in the case of 5-amino-1H-[1,2,4]triazole, gives two diacetylated isomers. These are methyl 1-acetyl-3-(acetylamino)-1H-[1,2,4]triazole-5-carboxylate (4) and 1-acetyl-5-(acetylamino)-1H-[1,2,4]triazole-3-carboxylate (5). Hypothetical pathways of their formation have been suggested. A mixture of 4 and 5 upon hydrolysis of the ring acetyl group gives the monoacetylated derivative methyl 5-(acetylamino)-1H-[1,2,4]triazole-3-carboxylate (6). The spectroscopic, structural and conformational characteristics of compounds 1-6 have been given and methods for their preparation have been provided.  相似文献

The angucycline antibiotic family of the landomycins displays potent antitumor activity. To elucidate early post polyketide synthase (PKS) tailoring steps of the landomycin E biosynthetic pathway in Streptomyces globisporus 1912, the mutant S. globisporus M12 was prepared through gene replacement experiment of lndM2. It encodes an enzyme with putative oxygenase and reductase domains, according to sequencing of the gene and its counterpart lanM2 from S. cyanogenus S136 landomycin A biosynthetic gene cluster. The isolation of the novel shunt products 11-hydroxytetrangomycin and 4-hydroxytetrangomycin along with the well-known angucyclines tetrangomycin and tetrangulol from the culture of S. globisporus M12 provides evidence for the involvement of lndM2 in the early biosynthetic pathway of the landomycins, in particular in the formation of the alicyclic 6-hydroxy function of the landomycin aglycon. We therefore propose LndM2 to be responsible for both hydroxylation of the 6-position and its subsequent reduction. These reactions are necessary before the glycosylation reactions can occur. The results are in agreement with the originally published structure of landomycin but do not support the recently suggested revised structure.  相似文献

A 3 kb DNA fragment from the Streptomyces globisporus 1912 landomycin E (LaE) biosynthetic gene cluster (lnd) was completely sequenced. Three open reading frames were identified, lndGT4, lndZ4, and lndZ5, whose probable translation products resemble a glycosyltransferase, a reductase, and a hydroxylase, respectively. Studies of generated mutants from disruption and complementation experiments involving the lndGT4 gene allowed us to determine that LndGT4 controls the terminal L-rhodinose sugar attachment during LaE biosynthesis and that LndZ4/LndZ5 are responsible for the unique C11-hydroxylation of the landomycins. Generation of the novel landomycins F, G, and H in the course of these studies provided evidence for the flexibility of lnd glycosyltransferases toward their acceptor substrates and a basis for initial structure-activity relationships within the landomycin family of antibiotics.  相似文献

A facile procedure for the synthesis of benzannelated diazacycloalkanes 3 is elaborated on the basis of reductive cleavage of 1,2-polymethylenebenzimidazoles 2 with DIBAL-H. The reduction of the corresponding salts prepared by treatment of 1,2-polymethylenebenzimidazoles 2 with methyl iodide proceeded differently affording exclusively o-phenylenediamines 10. The limitations and scope of the method were established, and a tentative mechanism proposed.  相似文献

An application of the flow differential pulse voltammetry with tubular detector based on silver solid amalgam for determination of antineoplastic drug lomustine in pharmaceutical preparations is presented. The highest sensitivity was obtained in [0.10 mol dm?3 MES; 2.00 mol dm?3 NaCl; pH 6.0]:EtOH (9 : 1) with flow rate 0.50 mL min?1, and the magnitude of the modulation amplitude ?0.070 V. The calibration dependence was linear in the range 1×10?6–1 × 10?4 mol dm?3 (R2=0.999). The limit of detection was 1.5×10?7 mol dm?3. This method was successfully used for determination of lomustine in real samples of chemotherapy drug CeeNU Lomustine 40 mg.  相似文献

An enzymatic reactor based on silver solid amalgam powder was suggested as the main part of biosensors in flow systems for the first time. Biosensors were tested with following enzymes: ascorbate oxidase, glucose oxidase, catalase, tyrosinase and laccase. The current response of each biosensor was optimized with respect to the detection potential, flow rate, the injection and reactor volume. Relative standard deviation for detection with the studied enzymes was found to be in the range of 0.81–2.1 %. The biosensor with the ascorbate oxidase reactor was used for determination of ascorbic acid in the vitamin tablets Celaskon.  相似文献

Technologically important composites with enhanced thermal and mechanical properties rely on the reinforcement by the high specific strength ceramic nanofibers or nanowires (NWs) with high aspect ratios. However, conventional synthesis routes to produce such ceramic NWs have prohibitively high cost. Now, direct transformation of bulk Mg‐Li alloys into Mg alkoxide NWs is demonstrated without the use of catalysts, templates, expensive or toxic chemicals, or any external stimuli. This mechanism proceeds through the minimization of strain energy at the boundary of phase transformation front leading to the formation of ultra‐long NWs with tunable dimensions. Such alkoxide NWs can be easily converted in air into ceramic MgO NWs with similar dimensions. The impact of the alloy grain size and Li content, synthesis temperature, inductive and steric effects of alkoxide groups on the diameter, length, composition, ductility, and oxidation of the produced NWs is discussed.  相似文献

Anion…π interactions are newly recognized weak supramolecular forces which are relevant to many types of electron‐deficient aromatic substrates. Being less competitive with respect to conventional hydrogen bonding, anion…π interactions are only rarely considered as a crystal‐structure‐defining factor. Their significance dramatically increases for polyoxometalate (POM) species, which offer extended oxide surfaces for maintaining dense aromatic/inorganic stacks. The structures of tetrakis(caffeinium) μ12‐silicato‐tetracosa‐μ2‐oxido‐dodecaoxidododecatungsten trihydrate, (C8H11N4O2)4[SiW12O40]·3H2O, (1), and tris(theobrominium) μ12‐phosphato‐tetracosa‐μ2‐oxido‐dodecaoxidododecatungsten ethanol sesquisolvate, (C7H9N4O2)3[PW12O40]·1.5C2H5OH, (2), support the utility of anion…π interactions as a special kind of supramolecular synthon controlling the structures of ionic lattices. Both caffeinium [(HCaf)+ in (1)] and theobrominium cations [(HTbr)+ in (2)] reveal double stacking patterns at both axial sides of the aromatic frameworks, leading to the generation of anion…π…anion bridges. The latter provide the rare face‐to‐face linkage of the anions. In (1), every square face of the metal–oxide cuboctahedra accepts the interaction and the above bridges yield flat square nets, i.e. {(HCaf+)2[SiW12O40]4?}n. Two additional cations afford single stacks only and they terminate the connectivity. Salt (2) retains a two‐dimensional (2D) motif of square nets, with anion…π…anion bridges involving two of the three (HTbr)+ cations. The remaining cations complete a fivefold anion…π environment of [PW12O40]3?, acting as terminal groups. This single anion…π interaction is influenced by the specific pairing of (HTbr)+ cations by double amide‐to‐amide hydrogen bonding. Nevertheless, invariable 2D patterns in (1) and (2) suggest the dominant role of anion…π interactions as the structure‐governing factor, which is applicable to the construction of noncovalent linkages involving Keggin‐type oxometalates.  相似文献
