Rapid identification of unknown microorganisms of clinical and agricultural importance is not only critical for accurate diagnosis of infections but also essential for appropriate and prompt treatment. We describe here a rapid method for microorganisms typing based on quantitative analysis of fatty acids by iFAT approach (Isotope-coded Fatty Acid Transmethylation). In this work, lyophilized cell lysates were directly mixed with 0.5M NaOH solution in d3-methanol and n-hexane. After 1 min of ultrasonication, the top n-hexane layer was combined with a mixture of standard d0-methanol derived fatty acid methylesters with known concentration. Measurement of intensity ratios of d3/d0 labeled fragment ion and molecular ion pairs at the corresponding target fatty acids provides a quantitative basis for hierarchical clustering. In the resultant dendrogram, the Euclidean distance between unknown species and known species quantitatively reveals their differences or shared similarities in fatty acid related pathways. It is of particular interest to apply this method for typing fungal species because fungi has distinguished lipid biosynthetic pathways that have been targeted for lots of drugs or fungicides compared with bacteria and animals. The proposed method has no dependence on the availability of genome or proteome databases. Therefore, it is can be applicable for a broad range of unknown microorganisms or mutant species. 相似文献

Theranostics provides opportunities for precision cancer therapy. However, theranostic probes that simultaneously turn on their diagnostic signal and pharmacological action only in respond to a targeted biomarker have been less exploited. We herein report the synthesis of a macrotheranostic probe that specifically activates its near‐infrared fluorescence (NIRF), photoacoustic (PA), and photothermal signals in the presence of a cancer‐overexpressed enzyme for imaging‐guided cancer therapy. Superior to the small‐molecule counterpart probe, the macrotheranostic probe has ideal biodistribution and renal clearance, permitting passive targeting of tumors, in situ activation of multimodal signals, and effective photothermal ablation. Our study thus provides a macromolecular approach towards activatable multimodal phototheranostics. 相似文献

The emerging metal single‐atom catalyst has aroused extensive attention in multiple fields, such as clean energy, environmental protection, and biomedicine. Unfortunately, though it has been shown to be highly active, the origins of the activity of the single‐atom sites remain unrevealed to date owing to the lack of deep insight on electronic level. Now, partially oxidized Ni single‐atom sites were constructed in polymeric carbon nitride (CN), which elevates the photocatalytic performance by over 30‐fold. The 3d orbital of the partially oxidized Ni single‐atom sites is filled with unpaired d‐electrons, which are ready to be excited under irradiation. Such an electron configuration results in elevated light response, conductivity, charge separation, and mobility of the photocatalyst concurrently, thus largely augmenting the photocatalytic performance. 相似文献

通过一步电沉积技术制备了普鲁士蓝/氧化锆修饰玻碳电极。采用电化学阻抗技术表征修饰电极。采用循环伏安法研究了pH值和扫描速率对该修饰电极的电化学行为的影响。结果表明:普鲁士蓝的峰电流与其扫描速率的一次方在一定范围内呈良好的线性关系。此外,该修饰电极在含有KCl(1.0mol/L)的磷酸盐缓冲溶液(0.1mol/L,pH=7.0)中,对H2O2具有明显的电催化作用,在无酶检测H2O2领域具有潜在的应用价值。 相似文献

Fe0.95S1.05 with high reactivity and stability was incorporated into WS2 nanosheets via a one-step solvothermal method for the first time. The resulted hybrid catalyst has much higher catalytic activity than WS2 and Fe0.95S1.05 alone, and the optimal WS2/Fe0.95S1.05 hybrid catalyst was found by adjusting the feed ratio. The addition of Fe0.95S1.05 was proven to be able to enhance the hydrogen evolution reaction (HER) activity of WS2, and vice versa. At the same time, it was found that the catalytic effect of the hybrid catalyst was the best when the feed ratio was W : Fe=2 : 1. In other words, we confirmed that there is a synergistic effect between W- and Fe-based sulfide hybrid catalysts, and validated that the reason for the improved HER performance is the strong interaction between the two in the middle sulfur. WS2/Fe0.95S1.05-2 hybrid catalyst leads to enhanced HER activity, which shows a low overpotential of ∼0.172 V at 10 mA cm−2, low Tafel slope of ∼53.47 mV/decade. This study supplies innovative synthesis of a highly active WS2/Fe0.95S1.05 hybrid catalyst for HER. 相似文献

以NO3-插层类水滑石(NiAl-NO3-LDH)为前驱体制备了一系列不同Ni/Al摩尔比的Ni-Al2O3催化剂,考察了其催化乙酰丙酸液相加氢性能.表征结果表明,随着Ni/Al摩尔比的增加,类水滑石层板结构中存在的游离态γ-AlOOH物种逐步演变为游离的Ni(OH)2物种,制备的催化剂中金属-载体相互作用逐渐减弱,Ni物种分散度逐渐降低,表面酸性中心数量先增多后减少;当Ni/Al摩尔比为3时,所制备的催化剂表面具有最丰富的酸性中心和加氢中心.在酸性中心和加氢中心的协同作用下,该催化剂表现出优异的催化乙酰丙酸加氢合成γ-戊内酯性能及良好的使用稳定性.在160℃,4 MPa氢气条件下反应5 h时,γ-戊内酯的收率最高可达92.7%. 相似文献
