A new series of quinolotacrine hybrids including cyclopenta- and cyclohexa-quinolotacrine derivatives were designed, synthesized, and assessed as anti-cholinesterase (ChE) agents. The designed derivatives indicated higher inhibitory effect on the acetylcholinesterase (AChE) with IC50 values of 0.285–100 µM compared to butyrylcholinesterase (BChE) with IC50 values of?>?100 µM. Of these compounds, cyclohexa-quinolotacrine hybrids displayed a little better anti-AChE activity than cyclopenta-quinolotacrine hybrids. Compound 8-amino-7-(3-hydroxyphenyl)-5,7,9,10,11,12-hexahydro-6H-pyrano[2,3-b:5,6-c'] diquinolin-6-one (6m) including 3-hydroxyphenyl and cyclohexane ring moieties exhibited the best AChE inhibitory activity with IC50 value of 0.285 µM. The kinetic and molecular docking studies indicated that compound 6m occupied both the catalytic anionic site (CAS) and peripheral anionic site (PAS) of AChE as a mixed inhibitor. Using neuroprotective assay against H2O2-induced cell death in PC12 cells, the compound 6h illustrated significant protection among the assessed compounds. In silico ADME studies estimated good drug-likeness for the designed compounds. As a result, these quinolotacrine hybrids can be very encouraging AChE inhibitors to treat Alzheimer’s disease.

A novel series of quinolotacrine hybrids were designed, synthesized, and evaluated against AChE and BChE enzymes as potential agents for the treatment of AD. The present study evaluates the binding of cetylpyridinium chloride (CPC) with cellulase in various experimental conditions using potentiometric, fluorescence spectroscopy and turbidimetric techniques. The analysis of binding curves revealed the existence of two sets of binding sets for CPC. The binding parameters were estimated and interpreted in terms of structural viewpoints of cellulase. The observation of turbidity suggests that CPC molecules individually nucleate around cellulase/CMC complex to form micelle-like structures. Fluorescence spectroscopy analysis of cellulase/CMC-surfactant system showed that these complexes could be compact to elucidate the mechanism of binding cellulase/CMC complex to CPC. The differential response of the enzyme/CMC to surfactant, indicates that the interaction on the complex surface is strongly ionic and hydrophobic(cooperative) in nature. A sequencing analysis was also conducted on β-1, 4-endoglucanase from A. niger (EglA) and others from family 12 in order to examine the nature of interaction involved in binding process and structure of carbohydrate-protein complexes. The results suggest that the conserved residues are located in a more hydrophobic microenvironment and apolar area energy is more than polar within enzyme structure.  相似文献

The Fourier coefficient map is considered as an operator from a weighted Lorentz space on the circle to a weighted Lorentz sequence space. For a large range of Lorentz indices, necessary and sufficient conditions on the weights are given for the map to be bounded. In addition, new direct analogues are given for known weighted Lorentz space inequalities for the Fourier transform. Applications are given that involve Fourier coefficients of functions in LogL and more general Lorentz–Zygmund spaces.  相似文献

The method of using rearrangements to give sufficient conditions for Fourier inequalities between weighted Lebesgue spaces is revisited, a comparison between two known sufficient conditions is completed, and the method is extended to provide sufficient conditions for a new range of indices. When \(1<q<p<\infty \), simple conditions on weights ensure that the Fourier transform will map a weighted \(L^p\) space into a weighted \(L^q\) space. These are established in Theorems 1 and 4 of Benedetto and Heinig (J Fourier Anal Appl 9(1):1–37, 2003). The proofs apply when \(2<q<p\) and \(1<q<p<2\) but not in the remaining case, \(1<q<2<p\). Here, counterexamples are given to show that these simple conditions are no longer sufficient when \(1<q<2<p\). Also, various additional conditions are presented, any of which will restore sufficiency in that case.  相似文献
