A simple stereoselective high performance liquid chromatographic method was developed for the determination of the in vitro transport of the enantiomers of nateglinide (N-(trans-4-isopropylcyclohexyl-carbonyl)-phenylalanine) in the rat intestine using a Chiralcel OJ-RH column (150 x 4.0 mm, 5 microm). The effects of the mobile phase composition, pH, the flow rate, and the temperature on the chromatographic separation were investigated. The enantioseparation was achieved at 33 degrees C using a mobile phase containing 100 mM potassium dihydrogen phosphate, pH 2.5, and ACN (32:68 v/v) delivered at a flow rate of 1 mL/min. The analytes were monitored at 210 nm and linearity (r >0.99) was obtained for a concentration range of 0.5-50 microg/mL. The LOD and LOQ were 0.2 and 0.5 microg/mL for the R-enantiomer and 0.2 and 0.8 microg/mL for the S-enantiomer, respectively. Both, the intra- and interday accuracy and precision of the calibration curves were determined. The method was successfully applied to estimate the in vitro passage of the enantiomers and the racemate of nateglinide in duodenum, jejunum, and ileum of rats. Generally, higher concentrations of nateglinide and the S-enantiomer were observed when the racemate was administered compared to administration of the individual enantiomers of nateglinide.  相似文献

In this study, a fluorescent material, 2‐naphthyl‐4‐amino benzoate, is synthesized by the esterification of 4‐aminobenzoic acid with 2‐naphthol. This molecule is used in the bulk polymerization of aniline, which results in the formation of poly(aniline‐2‐naphthyl‐4‐aminobenzoate). For comparison, polyaniline and also poly(aniline‐4‐aminobenzoic acid) salts are prepared via bulk polymerization. Formation and properties of these polymeric materials are evaluated by Fourier‐transform infrared (FT‐IR), 13C nuclear magnetic resonance, matrix‐assisted laser desorption ionization, UV‐Vis, Fluorescence, X‐ray diffraction (XRD), Field emission‐scanning electron microscopy (FE‐SEM), Differential scanning calorimetry (DSC), thermogravimetric analysis, electrical resistance and electrochemical techniques. P(ANI‐2NA4ABA) is obtained in nanofiber morphology in 106 wt% yield with respect to the amount of aniline used with comparable conductivity of conventional polyaniline salts. This polymer salt is stable up to 220°C and indicates melting at 146°C on heating and crystal formation at 128°C on cooling. This polymer shows higher wavelength fluorescence compared to the conventional polyaniline salts. This polymer is used as an electrode material without binder, which shows a specific capacitance of 360 F g?1 at 0.25 A g?1.  相似文献

Unbiased identification of organisms by PCR reactions using universal primers followed by DNA sequencing assumes positive amplification. We used six universal loci spanning 48 plant species and quantified the bias at each step of the identification process from end point PCR to next-generation sequencing. End point amplification was significantly different for single loci and between species. Quantitative PCR revealed that Cq threshold for various loci, even within a single DNA extraction, showed 2,000-fold differences in DNA quantity after amplification. Next-generation sequencing (NGS) experiments in nine species showed significant biases towards species and specific loci using adaptor-specific primers. NGS sequencing bias may be predicted to some extent by the Cq values of qPCR amplification.  相似文献

The ternary blends of acrylate rubber (ACM), poly(ethyleneterephalate) (PET), and liquid crystalline polymer (LCP) were prepared by varying the amount of LCP, but fixing the ratio of ACM and PET using melt mixing procedure. The compatibility behavior of these blends was investigated with infrared spectroscopy (IR), differential scanning calorimetry (DSC), and dynamic mechanical analyzer (DMA). The IR results revealed the significant interaction between the blend components. Glass transition temperature (Tg) and melting temperature (Tm) of the blends were affected depending on the LCP weight percent in the ACM/PET, respectively. This further suggests the strong interfacial interactions between the blend components. In the presence of ACM, the nucleating effect of LCP was more pronounced for the PET. The thermogravimetric (TGA) study shows the improved thermal stability of the blends.  相似文献
