Estimation of the ionization pK(a) of pharmaceutical substances using the computer program Sparc

The computer program sparc was used to calculate the pK(a) values of some important pharmaceutical substances. The sparc models proved to be suitable for estimating the pK(a) values of beta-adrenergic blocking agents and benzodiazepine drugs. Ionization macroconstants, microconstants, zwitterionic equilibria, speciation curves as a function of the pH and the isoelectric points of a semi-essential amino acid, arginine, and an anti-inflammatory, niflumic acid, were calculated.     

First total synthesis of (+/-)-strychnofoline via a highly selective ring-expansion reaction

An efficient synthesis of the antitumor alkaloid (+/-)-strychnofoline is documented. Key to the development of the highly convergent strategy delineated is the coupling of a cyclic imine with spiro[cyclopropan-1,3'-oxindole], which takes place in a highly diastereoselective manner. The ability to conduct annulation reactions of spirocyclopropyloxindoles with functionalized cyclic imines provides new avenues for the preparation of this important class of biologically active structures.     

Fused pyrazolopyrimidines. I. Pyrazolo[4,3-e]-v-triazolo[1,5-a]pyrimidine. A new heterocyclic system

5-Azido-4-cyano-1-phenylpyrazole reacts with benzylcyanide in the presence of sodium methoxide to give the title ring system.     

IDENTIFICATION OF A LUMAZINE PROTEIN FROM PHOTOBACTERIUM LEIOGNATHI BY COHERENT ANTI-STOKES RAMAN SPECTROSCOPY

The vibrational fingerprint in the frequency region 1700-1100 cm^-1 has been obtained for a fluorescent protein purified from the bioluminescent bacterium Photobacterium leiognathi. using the technique of resonance coherent anti-Stokes Raman scattering. The frequencies of the 4 prominent modes are the same as for lumazine protein purified from Photobacterium phosphoreum , and for an authentic sample of 6,7-dimethyl-8-ribityllumazine. The fluorophore in the new protein, therefore, must have the same chemical structure. The absence of frequency shifts between the free and bound ligand, in contrast to observations of fiavoproteins, implies that the lumazine ring undergoes no changes in hydrogen bonding between the free and protein-bound ligand in its ground electronic state. However, changes in relative intensities of two modes are seen, arising from differences in interactions in the excited state, and this is also reflected by differences in fluorescence properties.     

Catalytic asymmetric synthesis with rh-diene complexes: 1,4-addition of arylboronic acids to unsaturated esters

A general route to enantioenriched tert-butyl 3,3-diarylpropanoates is presented. These useful building blocks are prepared via an asymmetric rhodium-catalyzed conjugate addition of arylboronic acids to unsaturated tert-butyl esters in the presence of chiral dienes as ligands. The addition of both electron-poor and electron-rich boronic acids proceeds smoothly with various enoates in 63-90% yield with high enantioselectivites (89-94% ee). [reaction: see text]     

Cobalt-catalyzed hydrohydrazination of dienes and enynes: access to allylic and propargylic hydrazides

[reaction: see text] The cobalt-catalyzed hydrohydrazination reaction of dienes and enynes is presented. Allylic and propargylic hydrazines were obtained in synthetically useful yields (allylic amines, 60-90%; propargylic amines, 47-83%) and good chemo- and regioselectivity.     

Ring-fused cyclobutanes via cycloisomerization of alkylidenecyclopropane acylsilanes

A novel Lewis acid-catalyzed cycloisomerization of alkylidenecyclopropane acylsilanes is disclosed. The readily available starting materials participate in tandem Prins addition/ring expansion/1,2-silyl shift to grant access to bicyclo[4.2.0]octanes and bicyclo[3.2.0]heptanes, which are common motifs in terpenoid natural products. Notably, the transformation relies on the ability of acylsilanes to act sequentially as acceptors and donors on the same carbon atom.

A novel Lewis acid-catalyzed cycloisomerization of alkylidenecyclopropane acylsilanes is disclosed that involves tandem Prins addition/ring expansion/1,2-silyl shift to grant access to bicyclo[4.2.0]octanes and bicyclo[3.2.0]heptanes.
