Two-step preparation for catalyst-free biodiesel fuel production   总被引:10,自引:0,他引:10  
Biodiesel fuel was prepared by a two-step reaction: hydrolysis and methyl esterification. Hydrolysis was carried out at a subcritical state of water to obtain fatty acids from triglycerides of rapeseed oil, while the methyl esterification of the hydrolyzed products of triglycerides was treated near the supercritical methanol condition to achieve fatty acid methyl esters. Consequently, the two-step preparation was found to convert rapessed oil to fatty acid methyl esters in considerably shorter reaction time and milder reaction condition than the direct supercritical methanol treatment. The optimum reaction condition in this two-step preparation was 270°C and 20 min for hydrolysis and methyl esterification, respectively. Variables affecting the yields in hydrolysis and methyl esterification are discussed.  相似文献

A simple solid phase extraction (SPE) method coupled with high performance liquid chromatography (HPLC) using UV detector and microemulsion electrokinetic chromatography (MEEKC) has been developed and compared for the quantitative determination of miconazole nitrate in pharmaceutical formulation. For HPLC method, two parameters were optimized, namely, the wavelength and the mobile phases. The optimized condition was at the 225 nm wavelength and the mobile phase of ACN:MeOH (90:10 v/v). There are seven MEEKC parameters that were optimized, in this research, which were applied to voltage, temperature, wavelength, sodium dodecyl sulfate (SDS) concentration, buffer pH, buffer concentration and butan-1-ol concentration. The optimum MEEKC condition was obtained using 86.35 % (w/w) 2.5 mM borate buffer pH 9, 0.25 % (w/w) SDS, 0.8 % (w/w) ethyl acetate, 6.6 % w/w butan-1-ol and 6.0 % (w/w) acetonitrile. The combination of SPE using a diol column with HPLC–UV and the MEEKC methods were successfully applied for the determination of miconazole nitrate in a pharmaceutical formulation with the recovery percentage of 98.35 and 92.50 %, respectively.  相似文献

The 3-dimensional structure of the Paucimonas lemoignei poly(3-hydroxybutyrate) (PHB) depolymerase PhaZ7 has significant similarity to Bacillus subtilis lipase LipA but differs from the latter by the presence of an additional domain. Analysis of this lid-like domain revealed the presence of many hydrophobic amino acid residues including Tyr105. In this study we constructed His-tag fusions of PhaZ7 for simplified purification and investigated the effect of amino acid exchange of eight tyrosine codons of the lid-like domain. Exchanges of Tyr103, Tyr172, Tyr173, Tyr203 or Tyr204 to alanine or serine had no phenotype but muteins with substitution of Tyr189, Tyr190 and Tyr105 to alanine showed a lag phase of the in vitro PHB depolymerase reaction. Replacement of Tyr105 by glutamate further increased the lag phase. Binding assays of the purified PHB depolymerase proteins with the natural substrate, native PHB granules, revealed a significantly reduced binding ability of the Tyr105Glu mutant compared to the wild type protein and confirmed that Tyr105 is involved in interaction with the polymeric substrate.  相似文献

A three‐phase hollow fiber liquid‐phase microextraction method coupled with CE was developed and used for the determination of partition coefficients and analysis of selected nitrophenols in water samples. The selected nitrophenols were extracted from 14 mL of aqueous solution (donor solution) with the pH adjusted to pH 3 into an organic phase (1‐octanol) immobilized in the pores of the hollow fiber and finally backextracted into 40.0 μL of the acceptor phase (NaOH) at pH 12.0 located inside the lumen of the hollow fiber. The extractions were carried out under the following optimum conditions: donor solution, 0.05 M H3PO4, pH 3.0; organic solvent, 1‐octanol; acceptor solution, 40 μL of 0.1 M NaOH, pH 12.0; agitation rate, 1050 rpm; extraction time, 15 min. Under optimized conditions, the calibration curves for the analytes were linear in the range of 0.05–0.30 mg/L with r2>0.9900 and LODs were in the range of 0.01–0.04 mg/L with RSDs of 1.25–2.32%. Excellent enrichment factors of up to 398‐folds were obtained. It was found that the partition coefficient (Ka/d) values were high for 2‐nitrophenol, 3‐nitrophenol, 4‐nitrophenol, 2,4‐dinitrophenol and 2,6‐dinitrophenol and that the individual partition coefficients (Korg/d and Ka/org) promoted efficient simultaneous extraction from the donor through the organic phase and further into the acceptor phase. The developed method was successfully applied for the analysis of water samples.  相似文献

A rapid estimation of octanol–water partition coefficient (log P ow) was developed for triazole fungicides by micellar electrokinetic chromatography (MEKC). Five standard compounds with known log P ow values from 2.9 to 4.3 (cyproconazole, bromuconazole, epoxiconazole, bitertanol and difenoconazole) were used for constructing the calibration curve of the log P ow against the MEKC retention factor, log k. A linear relationship was achieved between log P ow and log k, in the MEKC system containing 40 mM sodium deoxycholate (SDC) and 4 mM borate buffer at pH 9.3, with a correlation of determination, r 2 = 0.9905. The log P ow values of four test compounds of triazole fungicides (triadimenol, myclobutanil, propiconazole and penconazole) were calculated based on the log k values measured by MEKC and the slope and intercept of the calibration curve. This MEKC method can give good estimation of the log P ow of the four test compounds of triazole fungicides with the differences between the literature log P ow values and estimated log P ow from the MEKC method were from 0.15 to 0.23 log units.  相似文献

The measurement of soil reaction forces on a lug of a movable lug cage wheel was carried out in a soil bin. To elucidate the experimental results, a theoretical analysis of soil reaction forces on the lug of the movable lug cage wheel was made by using an analysis of the lug trajectory and a modified theory in soil–vehicle mechanics. The existing theory was modified and adjusted by considering the actual lug trajectory and the soil trench made by the preceding lug. The results showed that the theoretical analysis gave a good representation of the reaction forces measured experimentally. The higher pull and lift forces of the movable lug cage wheel compared with those of the fixed lug wheel was supported by the theoretical analysis. Although the theoretical representation of soil reaction forces should be improved by further works, it is sufficiently accurate to estimate the performance of the movable lug cage wheels by the proposed theory.  相似文献

Linearity assessment as required in method validation has always been subject to different interpretations and definitions by various guidelines and protocols. However, there are very limited applicable implementation procedures that can be followed by a laboratory chemist in assessing linearity. Thus, this work proposes a simple method for linearity assessment in method validation by a regression analysis that covers experimental design, estimation of the parameters, outlier treatment, and evaluation of the assumptions according to the International Union of Pure and Applied Chemistry guidelines. The suitability of this procedure was demonstrated by its application to an in-house validation for the determination of plasticizers in plastic food packaging by GC.  相似文献

A movable lug wheel using a rollers-sliding groove mechanism was designed, constructed and tested. Two types of lug moving patterns of the movable lug wheel were proposed and evaluated. Tests were conducted in a soil bin test apparatus to determine the traction performance of the wheel as affected by lug moving pattern, lug spacing, horizontal load and vertical load. Similar tests were also conducted using a fixed lug wheel. Generally, under the same level of vertical load, the fixed lug wheel sank more than the movable lug wheels did. However in general, under various horizontal loads, there was no significant difference of slip between the movable lug wheel and the fixed lug wheel. Among the test lug wheels, the movable lug wheel with lug moving pattern-2 required the smallest driving torque and developed the highest traction efficiency.  相似文献
