Dehydropeptides containing dehydroamino acid (ΔAA) are frequently found in natural resources with important biological activity. Herein, we report the selective synthesis of Z- and E-ΔAbu from l- and l-allo-threonine as starting materials through selenation and oxidative elimination. BACKGROUND: 1alpha,25-Dihydroxyvitamin D(3) (1alpha,25(OH)(2)D(3)) has been shown to modulate not only proliferation and differentiation, but also apoptosis in malignant cells, indicating that it could be useful for treating cancer. Little information is available concerning the structural motifs of the 1alpha, 25(OH)(2)D(3) molecule responsible for modulation of differentiation and apoptosis, however. We set out to synthesize singly dehydroxylated A-ring analogs of 19-nor-1alpha,25(OH)(2)D(3) in a catalytic asymmetric fashion, and to investigate their biological activities in leukemia HL-60 cells. RESULTS: A series of singly dehydroxylated 19-nor-1alpha,25-dihydroxyvitamin D(3) A-ring analogs were synthesized using a combinatiorial sequence of regioselective propiolate-ene reaction and catalytic asymmetric carbonyl-ene cyclization. Surprisingly, the analogs could be clearly divided into two categories; one group, bearing 1alpha-hydroxy or 3beta-hydroxy groups in the A-ring, were potent differentiators and the second group, bearing 1beta-hydroxy or 3alpha-hydroxy groups, were potent stimulators of apoptosis. CONCLUSIONS: We have clearly identified the structural motifs of 19-nor-1alpha,25(OH)(2)D(3) analogs responsible for differentiation and apoptosis in HL-60 cells. These findings will provide useful information not only for development of therapeutic agents for treatment of leukemia and other cancers, but also for structure-function studies of 1alpha,25(OH)(2)D(3).  相似文献

Abstract

Electrochemical evolution of hydrogen gas, oxygen gas and chlorine at the electrodes is the usual reaction in conventional electrolysis of aqueous salt solution. However, here we demonstrate that the electrolysis governing reaction in hydrothermal solutions is different from the electrolysis performed at atmospheric pressure and temperatures up to 100°C. Experimental electrolytic reaction of aqueous salt solution carried out inside a sealed 300-mL batch autoclave showed that; accumulation of hydrogen gas, oxygen gas and chlorine is highly suppressed under hydrothermal (250°C and 7 MPa) conditions. We have also observed that, when organics are present in the aqueous salt solution being hydrothermally electrolyzed, an effective oxidation of organics is accomplished. Furthermore, for hydrothermal electrolytic oxidation (HEO) with oxygen gas added, experimentally observed TOC removal profile demonstrates apparent anodic oxidation electrical current efficiency of almost 200% for highconcentrated acetic acid solutions.  相似文献

Effect of ultrasound sonication was examined on the electroplating of iridium in aqueous hexabromoiridate(III) solution. The electrodeposits were evaluated by observing the defects of the iridium deposits by means of voltammetry, in which the current-potential curves of the iridium deposits on copper were measured. Applying ultrasound sonication to the electroplating of iridium decreased the defects including the cracks in the deposit whenever the glycerol as the additives was contained or not in the electrolyte.  相似文献

Conoideoxime A (1), a prenyl-tryptophan dimer possessing bis-oxime functionality, was isolated from cultures of the whitefly pathogenic fungus Conoideocrella luteorostrata BCC 76664. The structure was elucidated by spectroscopic analyses. It displayed antibacterial activity against Gram-positive bacteria, Bacillus cereus, Enterococcus faecium, and Staphylococcus aureus, with MIC values of 3.13, 6.25, and 6.25?μg/mL, respectively.  相似文献

Hypocrellutins A–C (13), 2- and 4-pyridone alkaloids, were isolated from cultures of the scale-insect pathogenic fungus Hypocrella discoidea BCC 71382. The structures were elucidated on the basis of NMR spectroscopic and mass spectrometry data. The absolute configuration of hypocrellutin B (2) was determined by ECD calculation. Their plausibly biosynthetic pathways are proposed. All isolated compounds were inactive in the antifungal, antimalarial, antitubercular, and cytotoxic activity assays.  相似文献

A stir foam composed of graphene oxide, poly(ethylene glycol) and natural latex (GO-PEG-NL) was prepared for use in micro-solid phase extraction sorbent of preservative agents and antioxidants from cosmetic products. The extracted analytes were quantified by GC-MS. Under the optimized conditions, the calibration plots are linear in the concentration ranges between 5.0 μg·L?1 to 1.0 mg·L?1 for benzoic acid, of 10.0 μg·L?1 to 1.0 mg·L?1 for 2-methyl-3-isothiazolinone (MI), and between 1.0 μg·L?1 and 1.0 mg·L?1 for both 3-tert-butyl-4-hydroxyanisole (BHA) and 2,6-di-tert-butyl-p-hydroxytoluene (BHT). The LODs are 1.0 μg·L?1 for benzoic acid, 5.0 μg·L?1 for MI and 0.5 μg·L?1 for both BHA and BHT. The stir-foam can be easily prepared, is inexpensive and well reproducible (RSDs <3%, for n?=?6). It can be re-used for up to 12 times after which extraction efficiency has dropped to 90%. The method was successfully applied to the determination of preservatives and antioxidants in cosmetic samples. Recoveries from spiked samples ranged between 94.5?±?2.1% and 99.8?±?1.8%.

A new lanostane triterpene (1), together with three known compounds (24), were isolated from cultivated fruiting bodies of the basidiomycete Ganoderma australe. The structure was elucidated on the basis of NMR spectroscopic and mass spectrometry data. The olefinic geometry of methyl australate (2) was revised from 20(22)Z to 20(22)E. These compounds (14) were different from the lanostanes isolated from mycelial cultures of the same strain source.  相似文献
