Laser desorption postionization mass spectrometry (LDPI-MS) imaging is demonstrated with a 10.5 eV photon energy source for analysis and imaging of small endogenous molecules within intact biofilms. Biofilm consortia comprised of a synthetic Escherichia coli K12 coculture engineered for syntrophic metabolite exchange are grown on membranes and then used to test LDPI-MS analysis and imaging. Both E. coli strains displayed many similar peaks in LDPI-MS up to m/z 650, although some observed differences in peak intensities were consistent with the appearance of byproducts preferentially expressed by one strain. The relatively low mass resolution and accuracy of this specific LDPI-MS instrument prevented definitive assignment of species to peaks, but strategies are discussed to overcome this shortcoming. The results are also discussed in terms of desorption and ionization issues related to the use of 10.5 eV single-photon ionization, with control experiments providing additional mechanistic information. Finally, 10.5 eV LDPI-MS was able to collect ion images from intact, electrically insulating biofilms at ~100 μm spatial resolution. Spatial resolution of ~20 μm was possible, although a relatively long acquisition time resulted from the 10 Hz repetition rate of the single-photon ionization source.

Bacterial nanocellulose (BNC) is an indomitable biomaterial of utmost usage in different technological areas. Previously, the BNC production has been reported in the simplified bioreactors. Thus, pioneering bioreactor-assisted strategies are desirable for the commendable BNC production. Advanced bioreactors must be corroborated along with different bacterial strains to obtain creditable BNC yield. This study deals with BNC production in rotary disc bioreactor (RDBR) using Gluconacetobacter xylinus NCIM 2526 strain. RDBR-based production of BNC provided 189?±?14 gL?1 of wet BNC, i.e., equivalent to 6.6?±?0.3 gL?1 dry BNC yield in 10 days. However, in static cultivation mode, 56?±?12 gL?1 wet weight of BNC, corresponding to 2.4?±?0.4 gL?1 dry weight, was produced. Thus, BNC production was approximately 2.75 folds higher in RDBR than statically produced BNC from the same volume of the media. The sugar to BNC conversion yield (12.2?±?0.8%) was doubled in RDBR-based production as compared to static BNC production (6.2?±?1.4%) with efficient sugar consumption (90.0?±?3.3%). The maximum amount of BNC was produced at 7 RPM and pH 6. RDBR-based BNC showed a more hydrophilic nature than statically produced BNC. The RDBR might be appropriate for large-scale BNC production, especially for wet-end applications, as an ample amount of BNC can be produced from a single fermentation process. These BNC pellicles might have the potential for biomedical applications like wound dressings, biofacial masks, hydrogels, and tissue engineering scaffolds.

Correlation between the dynamic viscosity (η) and the microviscosity of a hybrid green medium constituted of water and poly(ethylene glycol) (PEG) of average molar mass (200, 400, and 600) g · mol?1, respectively, is explored over the temperatures range (10 to 90) °C across the complete composition regime. The microviscosity is obtained using a fluorescence probe 1,3-bis-(1-pyrenyl)propane (BPP), which is manifested through the ratio of the monomer-to-intramolecular excimer intensities (IM/IE). Aqueous PEG mixtures are observed to behave similar to Newtonian fluids as the temperature dependence of dynamic viscosity follows Arrhenius-type behavior. Surprisingly, a simple and convenient linear dependence of ln η with wt% PEG of the mixture is established. The BPP IM/IE is observed, in general, to increase with the bulk dynamic viscosity of the mixture having >10 wt% PEG suggesting a good correlation between the bulk dynamic viscosity and BPP-reported microviscosity when the viscosity of the aqueous PEG mixture is relatively high.  相似文献

An activated concrete sample was counted at different source to detector distances with CdZnTe and HPGe detectors. The experimental count rates for different radionuclides were converted to dose rate using Monte Carlo code and compared with the Measured dose rates obtained using digital survey meter. The results agreed well for both the detectors. This indicates that CdZnTe detector having a better portability but poorer resolution than HPGe detector can be effectively used for online monitoring of radioactivity as well as dose rate calculations.  相似文献

Monte Carlo simulation is important to get efficiencies for cases where the experimental efficiencies are difficult to get such as for samples with nonstandard geometries and for large samples. In this paper, efficiency of the HPGe detector, routinely used in our lab for a variety of samples, has been computed for point source geometry and its parameters has been optimized to match MCNP and experimental efficiencies within 5% at different sample to detector distances. This optimized geometry was then validated by efficiency transfer to other geometries.  相似文献

The interaction of surface-active molecules with lipid bilayers is ubiquitous both in biological systems and also in several technological applications. Here we explore the interaction of ionic surfactants with liposomes whose composition mimics the ocular epithelia. In this study, liposomes with a composition mimicking ocular epithelia are loaded with calcein dye above the self-quenching concentration. The liposomes are then exposed to surfactants, and the rate of dye leaked from the liposomes due to the interaction of surfactants is measured. Both cationic and anionic surfactants at various concentrations and ionic strengths are explored. Results show that the liposome bilayer permeability to the dye increases on exposure to the surfactants, leading to the release of the dye trapped in the core. However, the dye release stops after a finite time, suggesting a transient increase in permeability followed by healing. The leakage profiles exhibit two different timescales for the cationic surfactant but only one timescale for the anionic surfactant. The total dye leakage increases with surfactant concentration, and at a given concentration, the dye leakage is significantly higher for the cationic surfactants. The timescale for the healing decreases with increasing surfactant concentration, and increasing ionic strength increases the dye leakage for the anionic surfactant. These results show that the surfactant binding to the lipid bilayer increases the permeability while the bilayers heal likely because of the surfactant jump from the outer to the inner leaflet and/or rearrangement into tighter aggregates.  相似文献

The effects of UV irradiation (λ=254 nm) on polyester nuclear track detector have been investigated employing bulk-etch technique, UV–visible spectrophotometry and infra-red spectrometry (FTIR). The activation energy values for bulk-etching were found to decrease with the UV-irradiation time indicating the scission of the polymer. Not much shift in the absorption edge due to UV irradiation was seen in the UV–visible spectra. FTIR studies also indicate the scission of the chemical bonds, thereby further validating the bulk-etch rate results.  相似文献

CdZnTe detector was used to monitor the radioactivity at several locations of primary heat transport (PHT) system of the Tarapur Atomic Power Station-III & IV reactor during shut down as well as operating condition of the reactor. Dose measurements at the same locations were also carried out using a teletector. MCNP code was used to simulate the detector efficiency for the required counting geometry. The efficiencies were then used to convert the experimental count rates (cps) to corresponding activity concentration (Bq/mL) of different radionuclides in coolant water. The total dose rates in these locations were also calculated by summing up the simulated dose rates of individual γ ray energy. The simulated and the measured total dose rates were found to be in reasonably good agreement.  相似文献

Densities of {poly(ethylene glycol) [PEG] + water} prepared with PEG average molar mass (200, 400, 600, and 1500) g · mol?1 have been measured over the entire composition range over the temperature range (283.15 to 363.15) K at 10 K intervals using a density meter based on electromagnetically-induced oscillations of a U-shaped glass tube and an inbuilt Peltier thermostat. The density versus temperature data of (PEG + water) at each composition for all PEGs were fit to a simple quadratic equation: ρ/(g · cm?3) = ρ0/(g · cm?3) + a(T/K) + b(T/K)2. Fits were observed to be satisfactory at each composition for all four (PEG + water). The excess molar volumes of (PEG + water) are observed to be negative and significant over the entire composition range for all four (PEG + water). Irrespective of the temperature, the maximum absolute excess molar volumes are observed in the water-rich region of the mixture and are found to decrease with increasing temperature. This is attributed to the presence of strong interactions within the (PEG + water). Specifically, it is proposed to be due to the H-bonding interactions between the PEG and the water molecules within the mixtures.  相似文献
