The novel NAD+-linked opine dehydrogenase from a soil isolate Arthrobacter sp. strain 1C belongs to an enzyme superfamily whose members exhibit quite diverse substrate specificites. Crystals of this opine dehydrogenase, obtained in the presence or absence of co-factor and substrates, have been shown to diffract to beyond 1.8 ? resolution. X-ray precession photographs have established that the crystals belong to space group P21212, with cell parameters a = 104.9, b = 80.0, c = 45.5 ? and a single subunit in the asymmetric unit. The elucidation of the three-dimensional structure of this enzyme will provide a structural framework for this novel class of dehydrogenases to enable a comparison to be made with other enzyme families and also as the basis for mutagenesis experiments directed towards the production of natural and synthetic opine-type compounds containing two chiral centres.  相似文献

The Bamberger-Ham condensation of 4-substituted nitrosobenzenes in concentrated sulfuric acid reported as a method of synthesis for phenazine N-oxides has been found to be limited to electron donating substituents. Methyl 4-nitrosobenzoale has been found to react under these conditions to give dimethyl 2-nitrodiphenylamine-4, 5-diearboxylate ( 2 ). Compounds of unknown structure previously reported to arise from acid treatment of 4-bromo- and 4-chloronilrosobenzene have been shown to be 4, 5-dibromo-2-nitrosophenylamine ( 10 ) and the analogous dichloro compound. Treatment under stronger acidic conditions (oleum) converted 10 but not 2 into the corresponding phenazine N-oxide. Mechanistic implications are discussed.  相似文献

The adverse health effects caused by increased exposure to ultraviolet radiation (UVR) due to deterioration of stratospheric ozone are of major concern. These health effects include sunburn, skin cancer, cataracts and immune suppression. Immune suppression has been associated with the release of cytokines, a defect in antigen presentation, induction of suppressor T cells and suppression of contact hypersensitivity (CH). CH is typically assessed by the mouse ear swelling test (MEST). Previous studies have demonstrated enhanced CH responses with vitamin A acetate (VAA) dietary supplementation assessed by MEST and the local lymph node assay (LLNA). To determine the effect that VAA has on UVR-induced immune suppression, we examined both the induction and elicitation phases of CH using murine models. The MEST was used to evaluate the interaction of UVR and VAA on CH elicitation. However, a positive MEST response requires that the induction phase as well as the elicitation phase of CH be functional. The LLNA was used to evaluate the interaction of UVR and VAA only on CH induction. We tested the hypothesis that mice maintained on a VAA-enriched diet are more resistant to UVR-induced immune suppression (CH) than those maintained on a control diet. Mice were maintained on a VAA-enriched or the control diet for 3 weeks and then exposed to UVR 3 days prior to sensitization with 2,4-dinitrofluorobenzene (DNFB). VAA enhanced the MEST response in both UVR-exposed and non-UVR-exposed mice. The VAA-enriched diet did not significantly alter the LLNA response in either UVR- or non-UVR-exposed mice. However, there was significant suppression in CH by UVR as measured by the LLNA. These results indicate that (1) the VAA-enriched diet does not restore the number of proliferating cells in the CH induction phase of UVR-induced immunosuppression; (2) the immunosuppressive effects of UVR affect the induction phase of CH; and (3) the LLNA should be examined as an alternative to the MEST for measurement of UVR-induced immunosuppression. The data indicate that the VAA-enriched diet enhanced the elicitation response (MEST) but not the earlier induction phase (LLNA). Further studies are necessary to define mechanisms of action, but modulation of cytokines and effects of specific lymphocyte subsets, as well as systemic effects and local modulation at the site of elicitation are possible. Additionally, future studies to evaluate the effect of the VAA-enriched diet when multiple doses of both UVR and DNFB are used would be of interest for both the LLNA and MEST end-points.  相似文献

A series of five 8-substituted α-[bis(2-hydroxyethyi)aminoethyl]naphtho[2,1-b]thiophenes 7 and a series of seven N-(3-dimethylaminopropyl)-8-substituted naphtho[2,1-b]thiophene-4-carboxamides 8 have been synthesized. The naphtho[2,1-b]thiophene-4-carboxylic acids 4 were prepared by photooxidative cyclization of α-(2-thienyi)-β-arylacrylic acids 3. The carboxylic acids 4 were converted by a conventional five step route involving α-bromoketone intermediates to the a-[bis(2-hydroxyethyl)aminomethyl]-8-substituted naphtho-[2,1-6]thiophene-4-methanols 7 and by a standard two-step amide preparation to the N-(3-dimethylaminopropyl)-8-substituted naphtho[2,1-6]thiophene-4-carboxamides 8 .  相似文献

Symmetrical and unsymmetrical bithiophene-substituted heterocycles bearing carbonitriles including imidazo[1,2-a]pyridine, benzimidazole, and pyridine derivatives have been synthesized via different synthetic protocols. The bithiophene bis-imidazo[1,2-a]pyridine derivatives 3a,b were achieved in three steps starting from 2-acetyl-5-bromothiophene. Suzuki coupling reaction of 2a with 5-formylthiophen-2-ylboronic acid forms the formyl derivative 5, which by condensation with 3,4-diaminobenzonitrile in the presence of sodium bisulfite furnishes the unsymmetrical bithiophene derivative6. The bis-benzimidazole derivative 8 was obtained via hexabutylditin-mediated homocoupling of 5-bromothiophene-2-carboxaldehyde, while the benzimidazole derivatives 12a,b were prepared via the formyl derivatives 11a,b, a product of Velsmier formylation reaction of 10a,b. Two synthetic protocols for the aryl/hetaryl-2,2′-bithiophene derivative 14 have also been presented. In addition, the guanyl hydrazones of bithiophenes, 16 and 17, were prepared from bis(tri-n-butylstannyl)-2,2′-bithiophene through a Stille coupling reaction followed by a condensation step.  相似文献

A class of feedback attitude-control systems which efficiently result in high-accuracy (10–4 radians), Earth-pointing motions of satellites of different configurations in elliptic orbits is developed. Gas jets provide the control torque. The state of the attitude motion is assumed to be completely known. Linear differential equations, which have time-varying coefficients and include terms for the gravity torque due to an oblate Earth and terms for the aerodynamic torque, are used to describe the attitude motion when the satellite is practically Earth-pointing. Pontryagin's maximum principle, the necessary conditions for exact solutions of optimal bounded-phase-coordinate problems, and guidelines obtained from the minimum-fuel attitude controls devised for single-axis systems are used in the development. The motions of somestable and someunstable satellites with the developed attitude-control systems are simulated on an analog computer. Carbon-13 NMR chemical shifts are reported for six angular and one linear dichloropyridoquinolines in CDCl3. The chemical shift assignments have been made using model compounds, fully coupled spectra, selective proton decoupling and results from lanthanide shift studies. Chlorine substituent effects are compared to those reported for chloroquinolines. The effect of the heteroaromatic nitrogen atoms on 13C? 1H coupling constants in these polycyclic systems are compared to those reported for pyridine systems.  相似文献
