Bacterial peptidoglycan is composed of a network of beta-[1,4]-linked glyan strands that are cross-linked through pendant peptide chains. The final product, the murein sacculus, is a single, covalently closed macromolecule that precisely defines the size and shape of the bacterial cell. The recent increase in bacterial resistance to cell wall active agents has led to a resurgence of activity directed toward improving our understanding of the resistance mechanisms at the molecular level. The biosynthetic enzymes and their natural substrates can be invaluable tools in this endeavor. While modern experimental techniques have led to isolation and purification of the biosynthetic enzymes utilized in peptidoglycan biosynthesis, securing useful quantities of their requisite substrates from natural substrates has remained problematic. In an effort to address this issue, we report the first total synthesis of lipid II (4), the final monomeric intermediate utilized by Gram positive bacteria for peptidoglycan biosynthesis. 相似文献

Background
We have recorded responses from single neurons in murine visual cortex to determine the effectiveness of the input from the two murine cone photoreceptor mechanisms and whether there is any unique selectivity for cone inputs at this higher region of the visual system that would support the possibility of colour vision in mice. Each eye was stimulated by diffuse light, either 370 (strong stimulus for the ultra-violet (UV) cone opsin) or 505 nm (exclusively stimulating the middle wavelength sensitive (M) cone opsin), obtained from light emitting diodes (LEDs) in the presence of a strong adapting light that suppressed the responses of rods.Results
Single cells responded to these diffuse stimuli in all areas of striate cortex. Two types of responsive cells were encountered. One type (135/323 – 42%) had little to no spontaneous activity and responded at either the on and/or the off phase of the light stimulus with a few impulses often of relatively large amplitude. A second type (166/323 – 51%) had spontaneous activity and responded tonically to light stimuli with impulses often of small amplitude. Most of the cells responded similarly to both spectral stimuli. A few (18/323 – 6%) responded strongly or exclusively to one or the other spectral stimulus and rarely in a spectrally opponent manner.Conclusion
Most cells in murine striate cortex receive excitatory inputs from both UV- and M-cones. A small fraction shows either strong selectivity for one or the other cone mechanism and occasionally cone opponent responses. Cells that could underlie chromatic contrast detection are present but extremely rare in murine striate cortex. 相似文献5.

The dynamic processivity of individual T4 lysozyme molecules was monitored in the presence of either linear or cross-linked peptidoglycan substrates. Single-molecule monitoring was accomplished using a novel electronic technique in which lysozyme molecules were tethered to single-walled carbon nanotube field-effect transistors through pyrene linker molecules. The substrate-driven hinge-bending motions of lysozyme induced dynamic electronic signals in the underlying transistor, allowing long-term monitoring of the same molecule without the limitations of optical quenching or bleaching. For both substrates, lysozyme exhibited processive low turnover rates of 20-50 s(-1) and rapid (200-400 s(-1)) nonproductive motions. The latter nonproductive binding events occupied 43% of the enzyme's time in the presence of the cross-linked peptidoglycan but only 7% with the linear substrate. Furthermore, lysozyme catalyzed the hydrolysis of glycosidic bonds to the end of the linear substrate but appeared to sidestep the peptide cross-links to zigzag through the wild-type substrate. 相似文献

Aqueous solutions of poly(ethylene glycol) (PEG) of mean molecular mass of 600 g/mol (PEG600) are investigated by Brillouin scattering technique. At high PEG content, a relaxation phenomenon is observed, which is related to a local rearrangement of the polymer structure where the interaction, via hydrogen bonding, with the solvent molecules plays a role. The obtained values of the relaxation times match the literature data very well for a fast relaxation time revealed by dielectric relaxation measurements in very similar mixtures. The calculated concentration behaviors of the excess adiabatic compressibility turns out in good agreement with the previous findings from ultrasonic measurements at 3 MHz. The observed minimum in the adiabatic compressibility is interpreted as the result of the interaction between water and the EO units of the PEG chain, which results in a structure tighter then that typical of bulk water and of pure PEG600. Such a hypothesis is supported by the observation that volume fraction value of about 0.3 coincides with the concentration value at which full hydration of EO units takes place. The observation that at the same concentration, the polymer coils start to overlap each other further supports the idea that the adiabatic compressibility behavior is monitoring the structural evolution of the mixture. However, similar results are obtained for largely different binary mixture which suggests caution in taking this conclusion too literally. In particular, the hypothesis that the occurrence of an extreme in the excess adiabatic compressibility could be simply originated by statistical effects and that further work is required for disentangling entropic contribution from effects of hetero-association and self-aggregation of one or both the components. 相似文献
