Metabolic thermogenic curves of mitochondria isolated from liver tissue of Carassius auratus and the effect of different concentration of NaAsO2 on it were investigated by TAM air isothermal microcalorimeter, ampoule method, at 28.00 ℃. From the thermogenic curves, activity recovery rate constants k, the maximum heat production rate (Pmax) and the total heat produced (Q) were obtained. The values of k and Pmax decline gradually with the increase of the concentration of NaAsO2, and both of the values of Pmax and k are highly correlated to the concentration of NaAsO2. When concentration of NaAsO2 reached 16.0 μg/mL, the maximum heat production rate dropped to 70.8% of the control group, and the corresponding percentage of k was 71.4% of the control group. This experimental result indicates that the addition of NaAsO2 has restrained the metabolic activities of mitochondria in vitro.  相似文献

在模拟人体生理条件下,利用荧光光谱法研究了黄芩素(Bcl)与人血清白蛋白(HSA)的相互作用。研究结果表明,黄芩素能够猝灭人血清白蛋白的内源荧光,其猝灭机理为静态猝灭。热力学参数ΔHθ(-70.8 kJ/mol),ΔSθ(-133 J·mol-1·K-1)的值均小于0,表明氢键和范德华力是维持基态复合物稳定性的主要作用力。位点竞争实验表明site I是黄芩素在HSA上的结合部位。三维荧光光谱实验结果表明黄芩素与人血清白蛋白相互作用后,改变了人血清白蛋白的构象。  相似文献

A microcalorimetric technique based on the metabolic heat-output was explored to evaluate the inhibition of cyanide on the mitochondrial metabolism of aquatic animal, Cyprinus carpio. The power-time curves could be divided into four parts: lag phase, active recovery phase, stationary phase, and decline phase, and the corresponding thermokinetic parameters were obtained. The maximum heat production rate Pmax decreased in a linear manner with the increase of concentration of cyanide, however, such mitochondria of aquatic animal were still metabolized actively even under the action of high concentration of cyanide. All the observations suggested that the mitochondria of this aquatic animal should exhibit considerable ability of cyanide-resistant respiration.  相似文献

在模拟人体生理条件下(pH=7.4),采用荧光光谱、位点竞争、三维荧光光谱研究了二苯甲酮(BP)与人血清白蛋白(HSA)之间的相互作用。根据修正的Stern-Volmer方程计算了不同温度下的结合常数,并结合Van't Hoff方程计算出相应的热力学参数。实验结果表明,BP对HSA的猝灭机制为静态猝灭过程,氢键和范德华力是维持BP-HSA复合物稳定的主要作用力;位点竞争实验揭示了BP在HSA上的结合位点位于亚域结构II A上的疏水腔中(site I位);三维荧光光谱分析表明BP使HSA发生了轻微地解旋,HSA的二级结构发生了改变。  相似文献

用TAM air等温微量热检测系统,测定鲫鱼(Carassius auratus)肝脏线粒体体外代谢热及不同浓度联苯胺(1.0,5.0,10.0,20.0,40.0mg/L)对其代谢活性的影响.结果表明。线粒体体外代谢活性恢复期速率常数k1,活性衰竭期速率常数k2以及最大放热功率Pmax均受到联苯胺的影响,且随着联苯胺浓度的升高,线粒体代谢总产热量Q依次下降。  相似文献
