Design,synthesis and evaluation of novel dehydroabietic acid-dithiocarbamate hybrids as potential multi-targeted compounds for tumor cytotoxicity

In the present study, novel representatives of the important group of biologically-active, dehydroabietic acid-bearing dithiocarbamate moiety, were synthesized and characterized by ¹H NMR, ¹³C NMR, HR-MS. The in vitro antiproliferative activity evaluation (MTT) indicated that these compounds exhibited potent inhibitory activities in various cancer cell lines (HepG-2, MCF-7, HeLa, T-24, MGC-803). Particularly, compound III-b possessed extraordinary cytotoxicity with low micromolar IC₅₀ values ranging from 4.07 to 38.84 µM against tested cancer cell lines, while displayed weak cytotoxicity on two normal cell lines (LO-2 and HEK 293 T). Subsequently, the potential mechanisms of representative compound III-b were elementarily investigated by Transwell experiment, which showed III-b can inhibit cancer cells migration. Annexin-V/PI dual staining showed that the compound can induce HepG-2 cells apoptosis in a dose-dependent manner. Meanwhile this apoptosis may be related to the upregulated protein expression of cleaved-caspase 3, cleaved-caspase 9, Bax and downregulated of Bcl-2 indicated by Western Blot. Later study further confirmed that ROS levels in HepG-2 cells increased significantly with the rise of concentrations. In addition, through the network pharmacology data analyzing, the core targets and signaling pathways of compound III-b for treatment of liver neoplasms were forecasted. Molecular docking model showed that compound III-b had high affinity with hub targets (CASP3, EGFR, HSP90AA1, MAPK1, ERBB2, MDM2), suggesting that compound III-b might target the hub protein to modulate signaling activity. Taken together, these data indicated that dehydroabietic acid structural modification following the “Molecular hybridization” principle is a feasible way to discover the potential multi-targeted antitumor compounds.     

Quantitative determination of potential genotoxic impurity 3-aminopyridine in linagliptin active pharmaceutical ingredient using HILIC–UV

This study aimed to develop an analytical method to determine the quantity of the impurity 3-aminopyridine (3AP). 3-Aminopyridine is a reactive reagent in the synthesis of linagliptin. The method was sensitive at level of 30.0 ppm of 3AP relative to linagliptin. The analysis was carried out using hydrophilic interaction liquid chromatography. The analytical column was Tracer Extrasil Silica (150 × 4.0 mm, 3 μm). A mobile phase of water–acetonitrile (10:90, v/v) containing 10.0 mM ammonium acetate was prepared and adjusted to pH 6.0. A UV detector was used to detect the amount of 3AP at a wavelength of 298 nm. Validation of the method was performed as per the International Council for Harmonization of Technical Requirements for Pharmaceuticals for Human Use in terms of detection limit, quantitation limit, linearity, accuracy, precision, specificity and robustness. The calibration curve was linear (r² = 0.999) for 3AP concentration in the range of 30.0–450.0 ppm. This method showed a good sensitivity with a detection limit and a quantitation limit of 7.5 and 25.0 ppm, respectively.     

基于混合空穴注入层的湿法制备高效蓝光热激活延迟有机发光二极管

在溶液法制备有机电致发光器件(OLEDs)的研究中, PEDOT∶PSS由于具有较好的成膜性与高透光性而常被用作器件的空穴注入层。但相关研究表明, PEDOT∶PSS本身稳定性较差以及功函数较低,这使得溶液法制备OLEDs的性能差且不稳定。蓝色作为全彩色的三基色之一,制备高效的蓝光器件对于实现高质量显示器件和固态照明装置必不可少。而目前溶液法制备蓝光OLEDs的器件效率普遍较差,针对此问题,本文利用传统的蓝光热激活延迟荧光发光(TADF)材料DMAC-DPS作为发光层,用溶液法制备了蓝光TADF OLEDs,通过在PEDOT∶PSS中掺杂PSS-Na制备混合空穴注入层(mix-HIL)来提高空穴注入层的功函数,研究其对于蓝光TADF OLEDs器件性能的影响。首先在PEDOT∶PSS水溶液中掺入不同体积的PSS-Na溶液,在相同条件下旋涂制膜,进行器件制备。通过观测各个实验组器件的电致发光(EL)光谱,发现掺入PSS-Na后器件EL谱存在光谱蓝移的现象,这是由于掺入PSS-Na水溶液后, mix-HIL层的厚度有所降低,使得在微腔效应作用下, EL光谱发生蓝移。通过对比各组器件的电流密度-电压-亮度(J-V-L)曲线及其计算所得器件的电流效率,结果显示随着PSS-Na的掺入,器件的亮度和电流都有所增大,器件的电流效率也得到了提升,当掺杂比例为0.5∶0.5(PEDOT∶PSS/PSS-Na)时提升幅度最大(亮度提升86.7%,电流效率提升34.3%)。通过在瞬态电致发光测试过程中施加或撤去驱动电压观测了器件EL强度的变化,分析了在混合空穴注入层/发光层(mix-HIL/EML)界面处的电荷积累情况。实验证明,通过在PEDOT∶PSS中掺杂PSS-Na制备mix-HIL获得了蓝光TADF OLEDs器件性能的提升,这是一个获得高效率溶液法制备OLEDs的可行方法。     

Phase transition in the one-dimensional pair-hopping model with unusual one-electron hopping

By modifying the conventional one-electron hopping behavior, we study effects of an occupation-dependent hopping on the ground state of the half-filled one-dimensional pair-hopping model. At weak coupling, the use of bosonization and renormalization-group analysis techniques helps to derive the phase diagram. Such unusual hopping is shown to drive a spin-gap transition and to introduce a new region where the triplet superconducting instability dominates for positively small pair-hopping interaction.     

新疆两处遗址出土绿松石文物的成分分析和产源判别

绿松石是一种产地有限,却在世界范围内被广泛使用的珍稀宝石资源,研究绿松石文物的矿料来源可为了解古代不同地区间珍稀资源的获取与交流模式、文化传播途径、早期贸易网络等学术问题提供帮助。新疆绿松石文物的矿料来源是近年来科技考古关注的热点问题之一,目前存在中原说、波斯说、新疆说和多元说等观点。研究新疆绿松石文物的产源,其难点在于既要保证文物的安全,又要确保分析数据的准确性,同时要避免样品表面污染对数据结果带来的干扰。鉴于此,采用等离子体原子发射光谱LA-ICP-AES对新疆加依、西沟两处墓地出土的绿松石进行化学成分分析,并结合秦岭东部五处产地的绿松石成分数据,经主成分分析绘制散点图——建立产地区分模型。主成分分析结果显示加依墓地样品中有五枚是罕见的锌绿松石,其余同西沟墓地绿松石的成分相近,均含有相对较高比例的Fe和Sr。结合中原地区的绿松石成分数据对比分析,发现新疆两遗址绿松石的微量元素特征相近,以高B2O低BaO为特征区别于中原地区的绿松石样品。在产地区分模型中,代表加依墓地和西沟墓地出土绿松石的散点聚集成团,且明显区分于白河、郧县、洛南、竹山、淅川五地的散点分布区域。综上结果表明新疆东部两处遗址的绿松石制品与中原东秦岭五处绿松石矿区所产矿料的成分差异较大,鉴于近年来新疆哈密发现与两处遗址同时代的绿松石采矿遗址,推测加依墓地、西沟墓地的绿松石制品其矿料来自中原地区的可能性较小。     

Vertex-disjoint cycles in local tournaments

Let $r$ be a positive integer. The Bermond–Thomassen conjecture states that, a digraph of minimum out-degree at least $2r-1$ contains $r$ vertex-disjoint directed cycles. A digraph $D$ is called a local tournament if for every vertex $x$ of $D$, both the out-neighbours and the in-neighbours of $x$ induce tournaments. Note that tournaments form the subclass of local tournaments. In this paper, we verify that the Bermond–Thomassen conjecture holds for local tournaments. In particular, we prove that every local tournament $D$ with ${\delta }^{+}\left(D\right)\ge 2r-1$ contains $r$ disjoint cycles $C_1,C_2,\dots ,C_r$, satisfying that either $C_i$ has the length at most 4 or is a shortest cycle of the original digraph of $D-C_1-\dots -C_{i-1}$ for $1\le i\le r$.
