设计合成了一系列吲哚咔唑结构的抗肿瘤新化合物. 通过对反应溶剂进行选择和调整, 优化了吲哚咔唑母核合成的反应条件, 使后处理更为简便. 用溴化噻唑蓝四氮唑(MTT)法对所合成的9个目标化合物进行了体外细胞毒活性测试, 结果表明, 化合物4a, 4b, 4d, 4f和4i对人结肠癌HCT116和鼠白血病P388细胞的活性均强于阳性对照ED-571, 其中化合物4f对P388细胞的活性是阳性对照的10倍.  相似文献

A number of new functionalised bridged indolocarbazole systems have been prepared by ring-opening reactions of a key cyclic sulfate intermediate, prepared from the corresponding diol by the action of sulfuryl diimidazole and DBU. Pyrazole analogs of the staurosporine aglycon K252c were recently described as potent inhibitors of the three Pim protein kinase isoforms. To evaluate the impact of the introduction of a sugar moiety on the biological activities of this heterocyclic scaffold, four new glucosylated pyrazole analogs of K252c were synthesized. Their biological evaluation demonstrated that most active compounds 11 and 19 substituted by a β-d-glucosyl moiety at N12 or N13 positions exhibited antiproliferative activities toward HCT116 cells.  相似文献

Indole-3-pyruvic acid is transformed to prodeoxyviolacein by the novel enzyme VioE, which is involved in the violacein biosynthetic pathway in Chromobacterium violaceum ATCC12472. VioE catalyzes the decarboxylation and indole-ring rearrangement of a nascent compound that is produced from indole-3-pyruvic acid and by the action of chromopyrrolic acid synthase (VioB or StaD), and ultimately the reaction yields prodeoxyviolacein.  相似文献

在B3LPY/6-31G(d, p)基组水平上,利用密度泛函理论(DFT)优化了吲哚并咔唑五种同分异构体的几何和电子结构.基于这五种同分异构体的几何结构下,其吸收和发射光谱的研究使用相同的基组水平并采用极化连续介质模型(PCM)下用含时密度泛函理论(TD-DFT)计算.由于三种近似线性分子(吲哚并[2,3-a]咔唑、吲哚并[2,3-b]咔唑和吲哚并[3,2-b]咔唑)的电荷转移跃迁的振荡强度较大,这些异构体的发射光谱存在明显差异;基于吲哚并[2,3-c]咔唑和吲哚并[3,2-a]咔唑的构型特征,这两种同分异构体的发射谱具有高能量.比较计算结果表明,吲哚并[2,3-b]咔唑在这些分子中的振荡强度最大.这是因为当吲哚并咔唑的五种同分异构体的结构从基态变为激发态时,这些分子的分子轨道(MO)能级不同.由计算结果还得出,这五个分子适用于P型传输材料,并且每个分子的三个苯环均具有共轭效应.  相似文献

ZHD-0501, a novel naturally occurring staurosporine analog showing anticancer activity in vitro, was isolated from the fermentation broth of a marine-derived Actinomadura sp. 007 through a bioassay-guided separation procedure and its structure elucidated by spectroscopic methods. ZHD-0501 provided the first example of staurosporin analog carrying a heterocycle fused to the pyran ring adopting a boat conformation with C-2′ and C-5′ as the bow and stern, which inhibited the proliferation of mammalian cancer A549, BEL-7402, HL60, P388, and tsFT210 cells.  相似文献

分别采用密度泛函理论(DFT)和单激发组态相互作用(CIS),在6-31G(d,p)基组水平上优化了5个吲哚咔唑分子的基态和激发态结构.在此结构的基础上,用含时密度泛函理论(TD-DFT)在相同基组水平和极化连续介质模型(PCM)下计算了模型分子的吸收和发射光谱.这几个吲哚咔唑异构体的发射光谱有明显的差别,如异构体5([3,2-b]型)有较大的振子强度,但是相对于其他异构体,其发射能量最小;异构体4([3,2-a]型)的发射能量最大;异构体2([2,3-b]型)的最大振子强度在250-450 nm范围内,与其他几个分子相比为最小.这主要是由分子的激发态几何变化和轨道能级的不同导致的.本文还考察了这类分子的一阶超极化率,结果显示5个分子极化率在同一水平,但静态第一超极化率(β0)有明显差别,异构体2的β0值最大.  相似文献
