The isomerism of glucaric acids and the complexity of the composition of Leonurus japonicus Houtt. increased the difficulty of the separation of glucaric acids from the herb. In the present study, three glucaric acids were isolated from Leonurus japonicus Houtt. by using high-speed countercurrent chromatography combined with semi-preparative high-performance liquid chromatography. Cation exchange resin chromatography was applied to remove the alkaloids and enrich the glucaric acid fractions. Preliminary separation of the glucaric acid extract by high-speed countercurrent chromatography was carried out at 45℃ by using an optimized solvent system of ethyl acetate/n-butanol/formic acid/water (1:1:0.01:2, v/v/v/v) with satisfied stationary phase retention and separation factor. The semi-preparative high-performance liquid chromatography was used for further separation and purification of the target fractions, and three monomeric compounds were obtained with purities of 90.0, 91.0, and 95.3%. UV spectroscopy, NMR spectroscopy, and mass spectrometry were employed to identify their structures, which were assigned as 2-syringyl glucaric acid, 2,4-disyringyl glucaric acid, and 3,4-disyringyl glucaric acid, respectively, and 2,4-disyringyl glucaric acid was reported for the first time. 相似文献
Ultrasound has been recognized as an exciting tool to enhance the therapeutic efficacy in tumor chemotherapy owing to the triggered drug release, facilitated intracellular drug delivery, and improved spatial precision. Aiming for a precise localized drug delivery, novel dendritic polyurethane-based prodrug (DOX-DPU-PEG) was fabricated with a drug content of 18.9% here by conjugating DOX onto the end groups of the functionalized dendritic polyurethane via acid-labile imine bonds. It could easily form unimolecular micelles around 38 nm. Compared with the non-covalently drug-loaded unimolecular micelles (DOX@Ph-DPU-PEG), they showed excellent pH/ultrasound dual-triggered drug release performance, with drug leakage of only 4% at pH 7.4, but cumulative release of 14% and 88% at pH 5.0 without and with ultrasound, respectively. The ultrasound responsiveness was attributed to the unique strawberry-shaped topological structure of the DOX-DPU-PEG, in which DOX was embedded in the skin layer of the hydrophobic DPU cores. With ultrasound, the DOX-DPU-PEG unimolecular micelles possessed enhanced tumor growth inhibition than free DOX but showed no obvious cytotoxicity on the tumor cells without ultrasound. Saxifraga atrata is an important traditional Tibetan medicine used to treat cough and pneumonia, and has tremendous medicinal potential. In this study, we devised a technique to separate 1,1-diphenyl-2-picrylhydrazyl inhibitors from a methanol extract of S. atrata. The material was first processed using MCI GEL CHP20P medium-pressure liquid chromatography, yielding 1.1 g of the target fraction Fr2. Subsequently, online hydrophilic interaction liquid chromatography-1,1-diphenyl-2-picrylhydrazyl assay was used to identify prospective 1,1-diphenyl-2-picrylhydrazyl inhibitors, and two 1,1-diphenyl-2-picrylhydrazyl inhibitor fractions (Fr24 and Fr25) were identified from Fr2. Then, medium-pressure preparation was continued using an XIon column to separate two 1,1-diphenyl-2-picrylhydrazyl inhibitor fractions (Fr24 and Fr25). The target compound was concentrated in fractions Fr24 and Fr25 using reverse-phase liquid chromatography during further separation procedures. Finally, the purity, structure, and 1,1-diphenyl-2-picrylhydrazyl inhibitory activity of the isolated 1,1-diphenyl-2-picrylhydrazyl inhibitors were determined. Two 1,1-diphenyl-2-picrylhydrazyl inhibitors (adenosine with the half maximal inhibitory concentration of 66.87 ± 14.33 μM and (-)-4-O-(E)-caffeoyl-l -threonic acid with the half maximal inhibitory concentration of 59.06 ± 5.02 μM) were isolated with purities exceeding 95%. Nanocarbon black (CB) was introduced into ethylene-vinyl acetate/brucite (EM) composites to investigate the synergistic effect of CB and metal hydroxide on improving the flame retardancy of EVA. Flammability properties of the as-prepared EVA composites were investigated by thermogravimetric analysis, limiting oxygen index (LOI), UL-94 test and cone calorimetry test. The results indicated that the optimum mass ratio of CB/brucite was 1/54, at which the EVA composites displayed dramatic improvement on thermal stability and flame retardancy. The LOI value was as high as 35.3%, the UL-94 passed the V-0 rating, and the peak heat release rate reduced 79% in comparison with pure EVA. Based on the morphology and structure analysis for residue chars, the flame-retardant mechanism was attributed mainly to the positive synergistic effect of CB and brucite on promoting the formation of better carbon protective layer during combustion.
This work deals with the prediction and experimental measurements of the (solid + liquid) equilibrium (SLE) in acid medium for industrial purposes. Specific systems including KCl–ethanol–water–HCl and K2SO4–water–H2SO4 were analyzed. At first, a critical discussion of SLE calculations was given, based on the well-known UNIQUAC extended and LIQUAC models. Two new proposals were derived, considering the explicit necessity of a new reference state for SLE calculations for the studied (solvents + acid) mixtures. The solubility of KCl in water–ethanol–HCl mixed solvents was measured in the temperature range of 300.15 to 315.15 K using an analytical gravimetric method. These results combined with some other experimental data reported in the open literature let us to propose a set of parameters for the new models. They included the interaction parameters between ethanol and the H+ ion. The prediction capability of the new models, for calculations in acid medium, was illustrated. Experimentally, it was observed that the (K2SO4 + water + H2SO4) system presented the unusual behavior of increasing K2SO4 solubility with an increase in the sulfuric acid concentration. To solve the problem that the magnetic resonance (MR) image has weak boundaries, large amount of information, and low signal-to-noise ratio, we propose an image segmentation method based on the multi-resolution Markov random field (MRMRF) model. The algorithm uses undecimated dual-tree complex wavelet transformation to transform the image into multiple scales. The transformed low-frequency scale histogram is used to improve the initial clustering center of the K-means algorithm, and then other cluster centers are selected according to the maximum distance rule to obtain the coarse-scale segmentation. The results are then segmented by the improved MRMRF model. In order to solve the problem of fuzzy edge segmentation caused by the gray level inhomogeneity of MR image segmentation under the MRMRF model, it is proposed to introduce variable weight parameters in the segmentation process of each scale. Furthermore, the final segmentation results are optimized. We name this algorithm the variable-weight multi-resolution Markov random field (VWMRMRF). The simulation and clinical MR image segmentation verification show that the VWMRMRF algorithm has high segmentation accuracy and robustness, and can accurately and stably achieve low signal-to-noise ratio, weak boundary MR image segmentation. 相似文献
