Ziritaxestat is a first-in-class autotoxin inhibitor. The purpose of this study was to develop a liquid chromatography/electrospray ionization tandem mass spectrometric (LC–MS/MS) method for the determination of ziritaxestat in rat plasma. The plasma sample was deproteinated using acetonitrile and then separated on an Acquity BEH C18 column with water containing 0.1% formic acid and acetonitrile as mobile phase, which was delivered at 0.4 ml/min. Ziritaxestat and the internal standard (crizotinib) were quantitatively monitored with precursor-to-product transitions of m/z 589.3 > 262.2 and m/z 450.1 > 260.2, respectively. The total running time was 2.5 min. The method showed excellent linearity over the concentration range 0.5–2000 ng/ml, with correlation coefficient >0.9987. The extraction recovery was >82.09% and the matrix effect was not significant. Inter- and intra-day precisions (RSD) were <11.20% and accuracies were in the range of −8.50–7.45%. Ziritaxestat was demonstrated to be stable in rat plasma under the tested conditions. The validated LC–MS/MS method was successfully applied to study the pharmacokinetic profiles of ziritaxestat in rat plasma after intravenous and oral administration. Pharmacokinetic results demonstrated that ziritaxestat displayed a short half-life (~3 h) and low bioavailability (20.52%). 相似文献
Photothermal therapy (PTT) is an emerging noninvasive and precise localized therapeutic modality; however, it is deeply limited by its poor tumor accumulation, inadequate photothermal conversion efficiency, and the thermoresistance of cancer cells. Aimed at these shortcomings, tumor‐targeting nanoparticles (iRGD‐W18O49‐17AAG) comprising carboxyl‐group‐functionalized W18O49 nanoparticles, integrin‐targeting peptide iRGD, and HSP90‐inhibitor 17AAG are developed. The W18O49 nanoparticles act as excellent PTT carriers and computed tomography (CT) imaging contrast agents. The ring type polypeptide iRGD promotes the accumulation of nanoparticles in the tumour and further penetration into cancer cells. The introduction of 17AAG can inhibit the heat‐shock response and overcome the thermoresistance, thus increasing the curative effect of PTT and reducing the chance of tumor recurrence. The W18O49 nanoparticles can also be used to monitor and guide the phototherapeutic through CT and near‐infrared fluorescence imaging after modification with Cy5.5. In addition, superior biosafety is also indicated in both preliminary in vitro and in vivo assessments. The potential of iRGD‐W18O49‐17AAG in tumor targeting, dual modality imaging‐guided and remarkable enhanced PTT of gastric cancer with ignorable side effect both in vitro and in vivo, which may be further applied in clinic, is highlighted. 相似文献
With the development of nano-powder technology, polymeric nano-materials are widely used in various industries, while not much research on their thermal decomposition and dust explosion characteristics has been conducted. The thermal behaviors and explosion characteristic parameters of the nano-polystyrene (nano-PS) with a typical particle size of 90 nm were studied by employing thermogravimetric analysis (TG), MIE-D 1.2 minimum ignition energy (MIE) test device, and 20-L spherical dust explosion test equipment. The results showed that the thermal decomposition of the nano-PS occurred in a two-step process which was different from the single process for conventional PS. Meanwhile, the reaction rate of the thermal decomposition for nano-PS increased with the heating rate. The TG and DTG curves shifted to the higher-temperature zone when the heating rate increased, and the initial temperature, final temperature, temperature at the maximum rate, and the maximum rate also increased. The sensitivity parameter of the minimum ignition energy of nano-PS varied as the dust concentration altered, and the most sensitive explosive concentration was about 200 g m−3. Also, nano-PS was proved to be quite sensitive to the electrostatic spark, as its calculated MIE value was as low as 11 mJ. For the severity parameters, the explosion pressure and its rising rate of nano-PS tended to increase at first and then decrease with the increase in dust concentrations. According to the risk classification standard, the explosion risk class of nano-PS was St2. The results were further extensively compared to other previous works. The results demonstrated both the higher explosion possibility and severity of nano-PS. This study could provide guidance for the safety management of nano-PS in its manufacture, storage, and handling process.
