Summary The design of new dynamic, axial-compression columns with a system for continous packed bed adjustment and monitoring of the
floating adapter position is described. The columns are meant for liquid chromatography at low pressures (up to 8 bar) in
aqueous and organic media with stationary phases of all types. The columns have adapter position pickups for continuous automatic
monitoring of the bed height (original “swellographic” monitoring). The column described with gas pressurisation was tested
with soft Sephadex G-10 and G-25. In spite of the reduction in external porosity there was no dramatic increase in back-pressure.
The column proved to provide long-term stability of the packed bed and improvement in resolution.

In this paper, superhigh reproductive rate strains of MEV with titre more than HA8192* or TCID50 log9.7 10 have been achieved both by cultivation in cell lines with different susceptibility to MEV and by isolating and identifying in field by the author. The systematic tests proved that S18 and L12 strains of MEV are the best strains for vaccine preparation. In this study, the best means for the tissue cultivation of MEV and the most advanced technological process for the production and detection of serum-free cell-cultured MEV fluids with super-high HA titre in batches in large quantities have been established for the first time. Optimum conditions for MEV inactivation were determined, and safe and effective inactivated vaccines with mineral oil or A1(OH)3 gel adjuvant were successfully prepared with serum-free cell-cultured MEV fluids. Copper oxide was incorporated into MCM‐41 by a one‐pot synthesis under acidic conditions to prepare a new mesoporous nitrosamines trap for protection of the environment. The resulting composites were characterized by XRD, N2 adsorption–desorption, and H2 temperature‐programmed reduction techniques, and their adsorption capabilities were assessed in the gaseous adsorption of N‐nitrosopyrrolidine (NPYR). The adsorption isotherms were consistent with the Freundlich equation. The copper salt was deposited onto MCM‐41 during the evaporation stage and was fixed on the host in the calcination process that followed. MCM‐41 was able to capture NPYR in air below 373 K but not at 453 K. Loading of copper oxide on MCM‐41 greatly improved its adsorption capability at elevated temperatures. The influence of the incorporation of copper into MCM‐41 samples and the adsorption behavior of these samples are discussed in detail. 相似文献

A micellar liquid chromatographic (MLC) procedure was developed for the clinical monitoring of imipramine and its active metabolite, desipramine. The determination of these highly hydrophobic substances was carried out after direct injection of the serum samples using a mobile phase composed of 0.15 m SDS--6% (v/v) pentanol buffered at pH 7, pumped at 1.5 mL/min into a C(18) column (250 x 4.6 mm), and electrochemical detection at 650 mV. Using this MLC method, calibration was linear (r > 0.995) and the limits of detection (ng/mL) were 0.34 and 0.24 for imipramine and desipramine, respectively. Repeatabilities and intermediate precision were tested at three different concentrations in the calibration range and a CV (%) below 2.2 was obtained. In this MLC procedure, the serum is determined without treatment, thus allowing repeated serial injections without changes in retention factors, and reducing the time and consumables required to carry out the pretreatment process. The assay method can be applied to the routine determination of serum imipramine and its metabolite in therapeutic drug monitoring. 相似文献

A new method has been developed for monitoring the degradation of paintings. Two inorganic pigments (ultramarine blue and red ochre) were blended with linseed oil and spread on canvas. Each canvas was subjected to simulated accelerated ageing in the presence of typical degradation agents (UV radiation and acidic solution). Periodically the painted surfaces were analysed by FT-Raman, to investigate the status of the surface. The data obtained were analysed by principal component analysis (PCA). Finally the Shewhart and cumulative sum control charts based on the relevant principal components (PC) and the so called scores monitoring and residuals tracking (SMART) charts were built. 7-Ethyl-10-hydroxycamptothecin (SN-38) is an active metabolite of Irinotecan (CPT-11), an anticancer pro-drug. To support clinical pharmacokinetic studies for liposome based formulation of SN-38 (LE-SN38) in cancer patients, a rapid, simple and sensitive liquid chromatography tandem mass spectrometry (LC-MS/MS) method has been developed and validated for the quantification of total SN-38 in human plasma. Sample preparation was carried out by one-step protein precipitation using cold acetonitrile with 0.5% acetic acid (v/v). Camptothecin was used as an internal standard (IS). Chromatographic separation of SN-38 and IS was achieved using a Synergi Hydro-RP column (C(18), 50 x 2 mm, 4 micro m), with a gradient elution of acetonitrile and 0.1% acetic acid. After ionization in electrospray source (positive ions), the acquisition was performed in the multiple reactions monitoring mode. Quantitation was accomplished using the precursor-->product ion combinations of m/z 393.1-->349.2 for SN-38 and 349.1-->305.1 for IS. The quantification limit of 0.05 ng/mL was achieved by using much lower volume (0.2 mL) of plasma and in the presence of LE-SN38. The method was validated over the concentration range of 0.05-400 ng/mL. Accuracy was within +/-12% of nominal at all concentration levels. Inter-day and intra-day precisions expressed as percentage coefficient of variation (%CVs) for quality control (QC) samples were less than 14 and 5%, respectively. 相似文献

A self‐sampling‐and‐flow biosensor was fabricated by sandwiching a nitrocellulose strip on the working electrode side of the double‐sided microporous gold electrodes and a wicking pad on the counter electrode side. The double‐sided microporous electrodes were formed by plasma sputtering of gold on a porous nylon substrate. Sample was taken up to the enzyme‐immobilized working electrode by the capillary action of the front nitrocellulose strip dipped into the sample solution, analyzed electrochemically at the enzyme‐immobilized electrode, and diffuses out to the backside wicking pad through the micropores of the electrodes, constituting a complete flow cell device with no mechanical liquid‐transporting device. Biosensor was formed by co‐immobilizing the glucose oxidase and electron transfer mediator (ferrocene acetic acid) on the thioctic acid self‐assembled monolayer‐modified working electrode. A typical response time of the biosensor was about 5 min with the sensitivity of 2.98 nA/mM glucose, providing linear response up to 22.5 mM. To demonstrate the use of self‐sampling‐and‐flow biosensor, the consumption rate of glucose in the presence of yeast was monitored for five days. 相似文献

for the separation/preconcentration of Mn, Cu, Cr, Zn, Ni and Co prior to their determination by inductively coupled plasma
optical emission spectrometer (ICP-OES) in environmental samples. The factors affecting the separation and preconcentration
of analytes such as pH, sample flow rate and volume, eluent concentration and volume were determined, interfering ions were
studied, and the optimal experimental conditions were established. The adsorption capacity of nanometer-sized ZrO2 for Mn, Cu, Cr, Zn, Ni and Co was found to be 1.3, 1.3, 1.7, 2.0, 3.9 and 1.5 mg g−1, respectively. The detection limits of the method were 12, 58, 24, 2, 7 and 36 ng L−1, respectively, with a preconcentration factor of 25. The precision of this method was 1.7% (Mn), 2.9% (Cu), 5.9% (Mn), 3.8%
(Mn), 6.2% (Mn) and 4.3% (Mn) with 9 determinations of 10 ng mL−1 of target analytes, respectively. The method was successfully applied to the determination of trace metals in lake water,
