Leaf samples of Lawsonia inermis (Li) were examined for their antimicrobial potential. Broth extracts in different concentrations were prepared and bioassayed in vitro for the growth of Escherichia coli. The growth of E. coli pathogen was inhibited to various degrees by increasing the concentration of the herbal powder. In addition to the observed alterations which were detected electrophoretically in the protein pattern, were activities of the amylase enzyme and glycoprotein fractions. The protein pattern has one common band of Rf 0.47 and two characteristic bands of Rf 0.36 and Rf 0.42 for E. coli sample. The quantitative mutation was observed in the bacteria with different concentrations of L. inermis compared with the control. Some types of proteins in E. coli completely disappeared upon being S.I affected. The amylase pattern showed one common band with Rf 0.037 and two characteristic bands with Rf 0.18 and Rf 0.37 for E. coli sample. The obvious quantitative mutation observed in bacteria with different concentrations of L. inermis compared with E. coli. The glycoprotein pattern recorded one common band at R1 with Rf 0.94 for E. coli sample and bacteria inoculated with different concentrations of L. inermis. These results confirmed the antibacterial activity of henna leaves and supported the traditional use of the plant in therapy of bacterial infections and disturbances that occurred at the biochemical level. The broth extract of the L. inermis leaves showed obvious antibacterial activity against E. coli.  相似文献

Surface imprinting over nanosized support materials is particularly suitable for protein templates, considering the problems with mass transfer limitation and low binding capacity. Previously we have demonstrated a strategy for surface protein imprinting over vinyl-modified silica nanopartiles with lysozyme as a model template by polymerization in high-dilution monomer solution to prevent macrogelation. Herein, the synthesis process was further studied toward enhancement of the imprinting performance by examining the effect of several synthesis conditions. Interestingly, the feed crosslinking degree was found to have a great impact on the thickness of the formed imprinting polymer layers and the recognition properties of the resulting imprinted materials. The imprinted particles with a crosslinking degree up to 50% showed the best imprinting effect. The imprinting factor achieved 2.89 and the specific binding reached 23.3 mg g−1, which are greatly increased compared to those of the lowly crosslinked imprinted materials reported previously. Moreover, the relatively high crosslinking degree led to no significant retarding of the binding kinetics to the imprinted particles, and the saturated adsorption was reached within 10 min. Therefore, this may be a promising method for protein imprinting.  相似文献

Analyte–wall interaction is a significant problem in capillary electrophoresis (CE) as it may compromise separation efficiencies and migration time repeatability. In CE, self-assembled polyelectrolyte multilayer films of Polybrene (PB) and dextran sulfate (DS) or poly(vinylsulfonic acid) (PVS) have been used to coat the capillary inner wall and thereby prevent analyte adsorption. In this study, atomic force microscopy (AFM) was employed to investigate the layer thickness and surface morphology of monolayer (PB), bilayer, (PB-DS and PB-PVS), and trilayer (PB-DS-PB and PB-PVS-PB) coatings on glass surfaces. AFM nanoshaving experiments providing height distributions demonstrated that the coating procedures led to average layer thicknesses between 1 nm (PB) and 5 nm (PB-DS-PB), suggesting the individual polyelectrolytes adhere flat on the silica surface. Investigation of the surface morphology of the different coatings by AFM revealed that the PB coating does not completely cover the silica surface, whereas full coverage was observed for the trilayer coatings. The DS-containing coatings appeared on average 1 nm thicker than the corresponding PVS-containing coatings, which could be attributed to the molecular structure of the anionic polymers applied. Upon exposure to the basic protein cytochrome c, AFM measurements showed an increase of the layer thickness for bare (3.1 nm) and PB-DS-coated (4.6 nm) silica, indicating substantial protein adsorption. In contrast, a very small or no increase of the layer thickness was observed for the PB and PB-DS-PB coatings, demonstrating their effectiveness against protein adsorption. The AFM results are consistent with earlier obtained CE data obtained for proteins using the same polyelectrolyte coatings.  相似文献

The typical concentration of protein loaded varies from 0.13 to 1.40 μg/μL for a classical silver staining method in 2DE gel. Here, we present a simple modified classical silver staining method by modifying the silver impregnation and development reaction steps. This modified method detects the protein spots at extremely low loaded concentrations, ranging from 0.0048 to 0.0480 μg/μL. We recommend this modified silver staining as an excellent method for the limited biological samples used for silver‐stained 2DE analysis. Altogether, the protocol takes close to two days from first dimension separation to second dimension separation, followed by silver staining, scanning, and analysis.  相似文献

A simple and economical CE method has been developed for the analysis of four model basic proteins by employing N‐methyl‐2‐pyrrolidonium methyl sulfonate ionic liquid (IL) as the dynamic coating material based on the interaction of both between electrostatic attraction and hydrogen bond, and between the organic cations of IL and the inner surface of bare fused‐silica capillary. The N‐methyl‐2‐pyrrolidonium‐based IL modified capillary not only generated a stable suppressed electroosmotic flow, but also effectively eliminated the wall adsorption of proteins. Several important parameters such as the IL concentration, pH values, and concentrations of the background electrolyte were optimized to improve the separation of basic proteins. Consequently, under the optimum separation conditions, a satisfied separation of basic proteins including lysozyme, cytochrome c, ribonuclease A, and α‐chymotrypsinogen A with theoretical plates ranging from 2.09 × 105 to 4.48 × 105 plates/m had been accomplished within 15 min. The proposed method first illustrated the effect of hydrogen bond between coating material and inner capillary surface on the coating, which should be a new strategy to design and select more effective coating materials to form more stable coatings in CE.  相似文献

As a noncovalent fluorescence probe, in this study, salicylaldehyde azine (SA) was introduced as a sensitive fluorescence‐based dye for detecting proteins both in 1D and 2D polyacrylamide electrophoresis gels. Down to 0.2 ng of single protein band could be detected within 1 h, which is similar to that of glutaraldehyde‐silver stain, but approximately four times higher than that of SYPRO Ruby fluorescent stain. Furthermore, comparative analysis of the MS compatibility of SA stain with SYPRO Ruby stain indicated that SA stain is compatible with the downstream of protein identification by LC‐MS/MS. Additionally, the probable mechanism of the SA stain was investigated by molecular docking. The results demonstrated that the interaction between SA and protein was mainly contributed by hydrogen bonding and hydrophobic forces.  相似文献

以牛血清蛋白(BSA)为模型蛋白,N-异丙基丙烯酰胺(NIPAM)为单体,N,N'-亚甲基双丙烯酰胺(BIS)为交联剂,采用原位沉淀聚合法制备了单分散的温度响应型蛋白纳米胶囊(nBSA).通过调整单体与蛋白的比例制备了粒径大小不同的含有单个蛋白分子的BSA纳米胶囊.采用基质辅助激光解吸电离飞行时间质谱仪(MALDI-TOF MS)、透射电镜(TEM)和动态光散射仪(DLS)等对BSA蛋白的修饰度,nBSA的形貌和结构,以及温度响应性能进行了表征,并用HeLa细胞对nBSA的体外安全性进行了初步评价.结果表明,在一定范围内,随着单体和蛋白比例的升高,蛋白纳米胶囊的粒径也逐渐增大,且在7.4~17 nm之间可控,而nBSA的响应温度则逐渐减小在33~41℃之间可控;制备的nBSA单分散性较好;nBSA具有温度响应性能,当环境温度高于其响应温度时,nBSA的粒径可显著增大16~33倍,且这种变化随温度呈现可逆性,并通过对nBSA细胞毒性的初步考察,评价将其用于生物领域的潜力.  相似文献
