The structural properties and Angiotensin-I converting enzyme (ACE) inhibition activities of a polysaccharide (PGE) extracted from Gastrodia elata Blume were investigated. PGE was extracted using hot water and purified by Sephadex G-200 followed by ultra-filtration. The structural characterisation of PGE was analysed by FT–IR, NMR spectroscopy, specific rotation determination, periodate oxidation-smith degradation, methylation analysis, GC–MS and Congo red test. The results revealed that PGE was composed by glucose, with an average molecular weight of 1.54 × 103 kDa. The structure of PGE was 1→3 and 1→4,6-branched-glucopyranose that had a linear backbone of (1 → 4)-linked-d-glucopyranose (Glcp). ACE-inhibitory activity results showed that PGE was efficient to inhibit ACE and the IC50 value was 0.66 mg/mL. 相似文献

Four new gastrodin derivatives containing a trans-cinnamoyl unit (1–4) and nine known compounds (5–13) were isolated from the rhizomes of Gastrodia elata Blume. All these compounds were evaluated for their neuroprotective effects against 6-hydroxydopamine-induced cell death, and compounds 7 and 12 showed potent activities with EC50 values of 10.5 and 10.2 μM, respectively. 相似文献

Gastrodia elata (G. elata), as a natural plant with nourishing and edible value, plays a vital role in food and pharmaceutical production. In the present study, a novel approach for the simultaneous determination of six components based on high‐performance liquid chromatography (HPLC) coupled with photodiode array detection (PDA) was developed for the quality evaluation of G. elata from different regions. Meanwhile, antioxidant and α‐glucosidase inhibitory activities were estimated and compared. The results indicated that the total contents of the six compounds in G. elata from Guizhou and Zhejiang provinces obtained by boiling were higher than those obtained by steaming in Anhui and Yunnan provinces. In addition, samples from Guizhou, Yunnan and Shanxi provinces contained more p‐hydroxybenzyl alcohol, and always possessed higher antioxidant activity, while samples collected from Anhui province showed the highest α‐glucosidase inhibitory activity with the highest gastrodin content. The results revealed that the quality of G. elata was affected by different regions and different initial processing technologies, which provided a reference for the selection and application of G. elata. 相似文献

Gastrodia rhizome, a dried and steamed tuber of Gastrodia elata Blume (Orchidaceae), has been traditionally used in Korea, China and Japan for the treatment of neurological and nervous disorders such as headaches, dizziness, vertigo and convulsive illnesses. The ethyl acetate and water extracts of G. elata stimulated plasmin activity. The active ethyl acetate fraction was subjected to centrifugal partition chromatography (CPC) with a two‐phase solvent system, composed of n‐hexane–ethyl acetate–methanol–water (3:7:4:6, v/v) followed by semi‐preparative HPLC purification to separate active compounds and the water fraction was purified by Diaion HP‐20 resin and semi‐preparative HPLC. In ethyl acetate extract, 4‐hydroxybenzyl alcohol (1), 4‐hydroxybenzoic acid (2), 4‐hydroxybenzaldehyde (3), 4‐ethoxymethylphenol (4), 4,4′‐oxybis(methylene)diphenol (5) and 4,4′‐methylenediphenol (6) were obtained with high purities. Parishin (7) and parishin B (8) were isolated from water extract. Among isolated compounds, 4‐hydroxybenzyl alcohol (1), 4‐hydroxybenzaldehyde (3) and 4‐ethoxymethylphenol (4) significantly stimulated plasmin activity. Copyright © 2015 John Wiley & Sons, Ltd. 相似文献

Phytochemical investigation of the rhizomes of Gastrodia elata Blume (Orchidaceae) led to the isolation and identification of twelve compounds, including two new phenolic compounds, 4‐hydroxybenzyl vanillyl ether (=4‐{[(4‐hydroxyphenyl)methoxy]methyl}‐2‐methoxyphenol; 1 ) and 4‐{{4‐[4‐(methoxymethyl)phenoxy]benzyl}oxy}benzyl methyl ether (=1‐[4‐(methoxymethyl)phenoxy]‐4‐{[4‐(methoxymethyl)phenoxy]methyl}benzene; 2 ). Gastrodia Rhizoma is a Traditional Chinese Medicine applied in the treatment of stroke, numbness of limb, headache and dizziness. However, its clinical effect is threatened by sulfur‐fumigation used in the process of storage. This article employs content determination coupled with high‐performance liquid chromatography fingerprint to investigate the effect of sulfur‐fumigation on Gastrodia Rhizoma so as to evaluate the quality of Gastrodia Rhizoma. The result was that most active ingredient in Gastrodia Rhizoma decreased after sulfur‐fumigation and the fingerprints analyzed by mathematical statistics between sulfur‐fumigated Gastrodia Rhizoma and unfumigated Gastrodia Rhizoma have substantial differences, which reveals that sulfur‐fumigation has a significant influence on the quality of Gastrodia Rhizoma. The conclusion of hierarchical clustering analysis, principal component analysis and partial least squares could validate each other, which implies that the method of mathematical statistics applied for assessing the quality of Gastrodia Rhizoma is effective and stable. The method not only affords a viable strategy for distinguishing Gastrodia Rhizoma whether sulfur‐fumigated or not and assessment of the quality of Gastrodia Rhizoma, but also provides a reference for other herbal medicine that suffers from sulfur‐fumigation. 相似文献

A novel homogeneous polysaccharide named GEP-1 was isolated and purified from Gastrodia elata (G. elata) by hot-water extraction, ethanol precipitation, and membrane separator. GEP-1, which has a molecular weight of 20.1 kDa, contains a polysaccharide framework comprised of only glucose. Methylation and NMR analysis showed that GEP-1 contained 1,3,6-linked-α-Glcp, 1,4-linked-α-Glcp, 1,4-linked-β-Glcp and 1,4,6-linked-α-Glcp. Interestingly, GEP-1 contained citric acid and repeating p-hydroxybenzyl alcohol as one branch. Furthermore, a bioactivity test showed that GEP-1 could significantly promote the growth of Akkermansia muciniphila (A. muciniphila) and Lacticaseibacillus paracasei (L. paracasei) strains. These results implied that GEP-1 might be useful for human by modulating gut microbiota. 相似文献

为建立测定天麻粉中硒的方法,采用微波消解技术溶解天麻粉,利用原子荧光法在优化条件下测定了其中硒的含量。结果表明,测定天麻中硒的回收率为93.1%~103.5%,RSD为4.5%。该法具有检出限低、灵敏度高、简便、准确、快速、线性范围宽等特点,能够达到测定天麻中微量硒的要求。 相似文献

Gastrolatathioneine (1), an unusual natural product derived from ergothioneine, a fungal amino acid containing an imidazole-2-thione moiety, was isolated from an aqueous extract of "tian ma" (the Gastrodia elata rhizomes). The structure of 1 including the absolute configuration was determined by extensive spectroscopic data analysis, combined with comparison of an experimental circular dichroism spectrum and calculated electronic circular dichroism spectra of stereoisomers, and confirmed by X-ray crystallography. The natural origin of 1 was proved by HPLC-ESIMS analysis of the crude extract. A biogenetic pathway of 1 is proposed on the basis of metabolic post-modification of ergothioneine that is biosynthesized by a symbiotic fungus.
