Ultra high performance liquid chromatography coupled with mass spectrometry and combining a tissue‐smashing extraction technique was developed for the simultaneous quantitative analysis of 12 compounds in the roots of Pulsatilla chinensis . Among them, compound 6 was characterized and accurately quantified in this herb for the first time. The parameters of extraction condition were simultaneously optimized with a Box–Behnken design and Derringer's function. The optimized conditions were as follows: sample quantity of 0.5 g, ethanol concentration of 70%, and extraction time of 200 s. Multiple‐reaction monitoring scanning was employed for the quantification between positive and negative mode in a single run of 6 min. Full validation of the method was carried out, and the results indicated that the method was rapid, specific, and reliable. The developed method was successfully applied to quantify the 12 compounds in 33 batches of P. chinensis from different provinces. Moreover, the principal component analysis was performed to compare the P. chinensis collected from different provinces of China based on quantitative data and the results indicated that the content of compounds could be used to differentiate the origins of P. chinensis . These results demonstrated that this method is feasible and reliable for the quality control of P. chinensis .  相似文献

Abstract

Phellodendri Chinensis Cortex is widely used in the clinic of traditional Chinese medicine. In order to enlarge the range of application, it is necessary to processed with honey, salt-water, and rice-wine, respectively. We hope to elucidate the connotation of processing, an UPLC-QqQ-MS method was used for determination and comparison the tissue distribution of alkaloids and triterpenes after oral administration water-extracts of crude and processed products. The results showed that the berberine, phellodendrine, magnoflorine, limonin, and obacunone in crude and processed products were distributed in all tissues, especially in the small intestine and stomach. In this study, we can provide a scientific basis for explaining the processing connotation of Phellodendri Chinensis Cortex processed with salt-water and rice-wine, respectively.  相似文献

采用非水毛细管电泳(NACE)法,以pH5.8的醋酸盐-甲醇为背景电解质溶液,运行电压25kV,温度20℃,检测波长210nm,正极压力(5kPa×10s)进样,建立了黄柏药材的NACE指纹图谱。以氯化两面针碱为内标,确定黄柏的NACE指纹峰为13个。测定了10个产地黄柏的NACE指纹图谱,与对照NACE指纹图谱的相似度为0.718~0.982。本方法具有较好的精密度,各指纹峰相对迁移时间的相对标准偏差(RSD)<1.5%,相对峰面积的RSD≤5%。方法可用于黄柏药材的质量控制。  相似文献

Hydrodistilled leaf oils of Pistacia chinensis Bunge from five locations in China were analyzed using GC/MS. A total of 58 compounds was identified in the oils, and a relatively high variation in their contents was found. The major compounds include β-phellandrene (0.54–53.86%), α-pinene (4.74–54.44%), β-pinene (0.49–42.90%), caryophyllene (5.64–20.01%), cis-ocimene (tr−43.93%), eudesmadiene (0–15.06%), and camphene (tr−20.57%). Cluster analysis classified the leaf oils into two chemotypes: one rich in α-pinene and β-pinene, and the other rich in β-phellandrene. 为研究中药射干(Belamcanda chinensis)的化学成分,应用多种柱色谱方法对其乙醇提取物进行分离和纯化,并得到了11个异黄酮化合物,通过1H NMR、13C NMR及HMBC、HSQC、1H-1H COSY、NOESY等波谱手段解析化学结构,发现一个新化合物,命名为异野鸢尾苷. 对这些化合物的1H NMR和13C NMR进行了完全归属,并探讨了取代基对1H、13C化学位移的影响.  相似文献
