肠道病毒71型外壳蛋白VP1在大肠杆菌中的表达   总被引:4,自引:0,他引:4  
将扩增得到的肠道病毒71型外壳蛋白VP1基因克隆到测序载体pGEM-T,测序验证该序列为目的片段后,将目的基因克隆到原核表达载体pGEX-5x-1中,转化大肠杆菌BL21,IPTG诱导表达,产物经SDS-PAGE分析和Western blot验证。结果表明,在经IPTG诱导的BL21中检测到分子量与预期大小相符的大约60 kDa的融合蛋白。利用表达产物作为抗原,对EV71感染病人阳性血清的检测初步证实,重组蛋白VP1可以作为检测EV71感染的检测用抗原。  相似文献

The effective parameters of chiral composite are studied using a simple model, that is, randomly oriented non-interacting wire helices embedded in a nonchiral host medium. It is found that both the effective permittivity and permeability are independent on the handedness of the chiral objects while the effective chirality admittance is dependent. It is also found that when the ratio of the radius of the chiral helix to its pitch is about 0.23, maximum chirality admittance is achieved. The effective parameters of equichiral sample are also discussed.  相似文献

The scattering cross sections for arbitrarily shaped dielectric objects with rough surface are determined for optical and infrared frequencies using the Kirchhoff approximation. The formula of the coherent scattering cross section is derived, and numerical method of incoherent scattering cross section is given. As a specific example, the infrared laser scattering cross sections of rough spheres are calculated at 1.06 m.  相似文献

It is proved that the apparent transverse velocities β_(app) and the spectral powers at 10GHz P_(10) of the cores of known superluminal sources are correlated. An interpretation ofthis result within the framework of the relativistic jet model shows that the jet Lorentzfactor γ depends on the intrinsic luminosity of the sources. The probable existence of twosubpopulations of the superluminal sources is shown by a linear regression analysis. "Small"sources f i.e. the sources without extended emission or with a distance from the core tothe outer edge of the lobe<20 kpc, show lower β_(app) than the "large" objects; this can beinterpreted as an orientation effect. Since all BL Lac objects belong to the "small" class,this result sheds new light on such a question as why these sources have lower apparent ve-locities than the "classical" superluminal quasars.  相似文献

Repressors are proteins that interact with inducers and operators to regulate the synthesis of enzymes in the cell. Lac repressor ensures that sufficient β-galactosidase is available to the bacterium Escherichia coli, depending upon the amount of lactose present in the culture medium. It has proved possible to isolate lac repressor from E.coli. The present progress report describes how these discoveries were made, as well as the formation and mode of action of the repressor.  相似文献

 研究了客体模型FTO的闪光照相系统X光输运过程,给出了直穿照射量、散射照射量、直散比、直穿照射量能谱、散射照射量能谱、直穿X光通量能谱和散射X光通量能谱在记录平面的空间分布。结果表明:后锥是照射量散射成分的主要来源,后锥照射量占总散射量97%;后锥也是造成散射的空间分布不均匀的主要器件,这一不均匀性高达58%。照相系统的最小直散比非常小,表明锥造成的散射已经严重地淹没了直穿(轫致辐射)信号。计算中使用高空间分辨率记录法进行分点,合成图像对吸收系数的复原结果与国外报道的结果相符。  相似文献

A novel approach to locate, identify and refine positions and whole areas of cell structures based on elemental contents measured by X‐ray fluorescence microscopy is introduced. It is shown that, by initializing with only a handful of prototypical cell regions, this approach can obtain consistent identification of whole cells, even when cells are overlapping, without training by explicit annotation. It is robust both to different measurements on the same sample and to different initializations. This effort provides a versatile framework to identify targeted cellular structures from datasets too complex for manual analysis, like most X‐ray fluorescence microscopy data. Possible future extensions are also discussed.  相似文献

This paper focuses on coupling methods for hybrid Navier–Stokes/molecular dynamics (MD) simulations. The computational domain is split in a continuum flow region, where a finite‐volume discretisation of the Navier–Stokes equations is used, and one or more particle domains, where molecular level modelling of the flow is employed. The domains are defined with a partial overlap, in which the flow states are coupled through an exchange of the velocity components. For the steady flows considered, an under‐relaxed Newton iteration method is used to drive the coupled system to convergence. The main focus of the present work is on methods to impose nonperiodic boundary conditions on the particle domain(s). A particle forcing is applied in the direction normal to the particle domain boundary to impose the boundary normal velocity component. A novel aspect of the present work is the extension of this method to more general nonplanar particle domain boundaries. The main contribution of the paper is the development of a particle forcing method in the direction tangential to the domain boundary, which is based on the equivalent continuum‐flow boundary shear stresses along with an iterative forcing strength adjustment based on the extrapolated particle boundary velocity. Furthermore, an adaptation scheme is presented, which uses the finite‐volume flux residuals of the particle bin averaged velocity field as a truncation criterion for the iterative force‐update scheme. It is demonstrated that by comparing the residual reduction for the momentum equation in the nonhomogeneous directions during the molecular dynamics simulations with that for a homogeneous direction, the forcing iteration at which the statistical noise in the velocity field dominates the uncertainty in the forcing strength can be determined. At this point the iteration can be truncated. It is shown that with adaptive schemes of this type, the total number of MD evaluations required in a coupled Navier–Stokes/MD simulation can be reduced relative to a hybrid scheme with a fixed number of forcing‐strength updates.
