New optically active organoantimony (BINASb) and bismuth (BINABi) compounds comprising a 1,1′-binaphthyl core: synthesis and their use in transition metal-catalyzed asymmetric hydrosilylation of ketones

Racemic 2,2′-bis[diarylstibano]-1,1′-binaphthyls [(±)-BINASbs] and 2,2′-bis[di(p-tolyl)bismuthano]-1,1′-binaphthyl [(±)-BINABi], which are the antimony and bismuth congeners of BINAP, have been prepared from 2,2′-dibromo-1,1′-binaphthyl (DBBN) via 2,2′-dilithio-1,1′-binaphthyl intermediate by treatment with the appropriate metal halides [(p-Tol)₂SbBr, Ph₂SbBr and (p-Tol)₂BiCl]. The optical resolution of the (±)-BINASbs could be achieved via the separation of a mixture of the diastereomeric Pd-complexes derived from the reaction of (±)-BINASbs with di-μ-chlorobis{(S)-2-[1-(dimethylamino)-ethyl]phenyl-C¹,N}dipalladium(II). Optically active (R)-BINASb and (R)-BINABi could be also obtained from optically active (R)-DBBN by the same procedure. The enantiopure BINASbs have been shown to be effective chiral ligands for the rhodium-catalyzed asymmetric hydrosilylation of ketones.     

Highly diastereoselective chemoenzymatic synthesis of (2′R)- and (2′S)-2′-deoxy[2′-H]guanosines

In an effort to develop an efficient synthetic method of highly diastereoselective (2′R)- and (2′S)-2′-deoxy[2′-²H]guanosines, chemoenzymatic conversion was investigated. The synthesis of (2′R > 98% de)-2′-deoxy[2′-²H]guanosine was achieved by biological transdeoxyribosylation using (2′R > 98% de)-2′-deoxy[2′-²H]uridine, 2,6-diaminopurine, and Enterobacter aerogenes AJ-11125, followed by treatment with adenosine deaminase. (2′S > 98% de)-2′-Deoxy[2′-²H]guanosine was synthesized from (2′S > 98% de)-2′-deoxy[2′-²H]uridine and 2,6-diaminopurine using thymidine phosphorylase and purine nucleoside phosphorylase instead of E. aerogenes AJ-11125.     

Synthesis of furan 4′-thio-C-nucleosides, their methylsulfonium and sulfoxide derivatives. Evaluation as glycosidase inhibitors

A series of furan C-nucleosides having a sulfur atom in the sugar ring were synthesised. The α and β anomers of 3-ethoxycarbonyl-2-methyl-5-(4′-thio-d-erythrofuranosyl)furans 10 and 11 were obtained by acid treatment of (4′-S-acetyl-4′-thio-d-arabino-tetritol-1-yl)furan 9. Oxidation of 10 with m-chloroperbenzoic acid gave sulfoxide 12 as one epimer at the sulfur atom whereas 11 was transformed into sulfoxide 13 as an epimeric mixture. S-Methylation of 10 and 11 with methyl triflate led to sulfonium salts 14 and 15. The prepared compounds were found to be moderate inhibitors of α-l-fucosidase.     

Synthesis of pyridine and 2,2′-bipyridine derivatives from the aza Diels-Alder reaction of substituted 1,2,4-triazines

Amidrazone 1a and the tricarbonyl derivatives 2b-d reacted in boiling ethanol in the presence of 2,5-norbornadiene 5 giving the pyridine derivatives 6b-d respectively (59-72%) and in the presence of 2,3-dihydrofuran 7 yielding the lactones 10b-d (39-44%). The 2,2′-bipyridine derivatives 6e-g were similarly obtained in good yield (81-87%) from the reaction of amidrazone 1b and tricarbonyl derivatives 2b-d in the presence of 2,5-norbornadiene 5.     

Deprotonated and non-deprotonated cobalt(II) complexes of the bis-amide tetradentate ligand N,N′-(dipicolyl)-1,8-naphthylene diamine. Three different modes of coordination by the ligand

The preparation and some properties of the cobalt(II) complexes Co(LH₂)Cl₂·2H₂O, Co(LH₂(NCS)₂ and CoL·H₂O (whereLH₂=N,N-(dipicolyl)-1,8-naphthylenediamine) are reported. On the basis of magnetic moments, visible reflectance and IR data, the structure is proposed to be pseudo-octahedral for Co(LH₂)Cl₂·2 H₂O, pseudo-tetrahedral for Co(LH₂)(NCS)₂ and square planar for CoL·H₂O.

Deprotonierte und Nicht-deprotonierte Co(II)-Komplexe des vierzähnigen Bisamid-Liganden N,N-(Dipicolyl)-1,8-naphthyldiamin. Drei verschiedene Koordinationstypen

Zusammenfassung Es werden die Darstellung und einige Eigenschaften der Kobalt(II)-Komplexe Co(LH₂)Cl₂·2 H₂O, Co(LH₂)(NCS)₂ und CoL·H₂O [LH₂=N,N-(dipicolyl)-1,8-naphthylendiamin] diskutiert. Auf der Grundlage von magnetischen Momenten, von Daten der sichtbaren Reflektions-und IR-Spektren wird eine pseudooctaedrische Struktur für Co(LH₂)Cl₂·2H₂O, eine pseudotetraedrische für Co(LH₂)(NCS)₂ und eine planar-quadratische für CoL·H₂O vorgeschlagen.

     

Synthesis of hapten-phosphoramidites from 2′-deoxyuridine

A total of 11 novel phosphoramidites, 3a-d, 4a-d and 14a-c were prepared from 2′-deoxyuridine functionalized at 5 and 6-position of the pyrimidine ring with hapten reporter groups, e.g. adamantane, carbazole, dansyl and dabsyl, suitable for use in immunodetection nucleic acid testing assays.     

TheSemmler-Wolff aromatization andSchmidt reaction applied to some pyrido[2′,3′:3,4]pyrazolo[5,1-c][1,2,4]benzotriazines

Pyrido[2,3:3,4]pyrazolo[5,1-c][1,2,4]benzotriazin-4(1H)ones were transformed via their oximes in aSemmler-Wolff aromatization process in the tetracyclic heteroaromatic amines4 or bySchmidt reaction into a mixture of the same amine4 and a ring enlarged lactam3. Syntheses of some halo pyrazolo[3,4-b]pyridines and a photochemical transformation of 3-azidopyrazolo[3,4-b]pyridine are also described.

Über dieSemmler-Wolff- undSchmidt-Reaktion einiger Pyrido[2,3:3,4]pyrazolo[5,1-c][1,2,4]benzotriazine

Zusammenfassung Pyrido[23:3,4]pyrazolo[5,1-c][1,2,4]benzotriazin-4(1H)one werden über Oxime in einerSemmler-Wolff-Reaktion in die tetracyklischen aromatischen Amine4 umgewandelt. In einerSchmidt-Reaktion wurden dieselben Ketone in ein Gemisch aus Amin4 und Lactam3 übergeführt. Synthesen von halogensubstituierten Pyrazolo[3,4-b]pyridinen und photochemische Umwandlung von 3-Azidopyrazolo[3,4-b]pyridin werden beschrieben.

     

Iron(III) chloride catalysed three-component Grieco condensation: Synthesis of tetrahydropyrido[2′,3′:3,4]pyrazolo[1,5-a]pyrimidines/quinazolines

Three-component Grieco condensation reaction of 3-aminopyrazolo[3,4-b]pyridine, formaldehyde/benzaldehyde and electron rich alkenes in presence of iron(III) chloride gave tetrahydropyrido[2′,3′:3,4]pyrazolo[1,5-a]pyrimidines/quinazolines in single pot. The sequence of reactions is formation of di-azadiene in situ and the subsequent regioselective addition of alkenes in aza-Diels Alder type reaction. The structure and stereochemistry of the products were confirmed by spectral data and single crystal X-ray crystallography.     

Diastereoselective addition of dimethyl phosphite to 3,3′,4,4′-tetramethyl-1,1′-diphosphaferrocene-2-carboxaldehyde

Addition of dimethyl phosphite to racemic 3,3′,4,4′-tetramethyl-1,1′-diphosphaferrocene-2-carboxaldehyde gives almost exclusively one diastereomer of the corresponding α-hydroxyphosphonate (d.r. ?96:4). Its absolute configuration (S, R_p)-(R, S_p) was established by X-ray diffraction.     

Quantum-mechanical exploration of the properties of the sugar rings

Theab initio SCF method is used for computing the main electronic properties of the ribose unit of the nucleic acids. The present study is devoted to the ribose in the C₃-endo,gg conformation. The properties investigated include the distribution of the electronic charges, the electrostatic molecular potential around the four oxygens of the unit, the hydration and the Na⁺ binding schemes studied in the supermolecule approximation. The possibilities of through-water binding of the cation to the sugar are also explored. The predictions of the computation in particular with regard to cation binding to the ribose ring are correlated with recent experimental results.
