以3-脲丙基三甲氧基硅烷为偶联剂,制备了一种新型极性脲丙基-C30(TPU-C30)反相色谱固定相。采用扫描电子显微镜、元素分析、红外光谱和热分析等对该固定相进行表征。结果表明,TPU-C30固定相已成功制备,连续制备3次固定相,其元素含量的相对偏差均小于5%,说明该合成工艺重复性良好。以不同极性、位置异构、碱性化合物为溶质探针,以传统的C18色谱柱与C30色谱柱为参比,对制备的固定相的色谱性能进行了研究。研究结果表明,TPU-C30固定相具有不同于传统C18柱、C30柱的选择性和更优的择形性,明显改善了碱性物质的峰形,其具有广阔的应用空间。 相似文献

Citric acid is finding new areas of use each year and the demand for the acid is constantly increasing. Being a bulk chemical,
the continuous production of citric acid would be advantageous. The paper presents the results from ammonia limited batch
and continuous fermentations using the yeast strainSaccharomycopsis (Candida) lipolytica (NRRL Y-7576). Mathematical models were developed for growth and glucose utilization in batch and continuous culture. Cell
and acid yields appeared to be almost the same in batch and continuous culture. The specific production rates were found to
be constant, equal to 0.053 g/g h, in the batch fermentations but varied in the continuous experiments from 0 to 0.11 g/g

Mg-Al spinel is synthesized by using industrial waste-residue and basic magnesium carbonate in the aluminum factory as the main raw materials. The influence of V2O5 and Fe2O3 mineralizers on the structure, cell parameters and microscopic morphology of synthesized Mg-Al spinel has been discussed. The Mg-Al spinel is characterized by using XRD, SEM and relevant analytical software, such as Philips plus, Rietveld quantification and so on. The experimental results show that certain amounts of V2O5 and Fe2O3 are beneficial to the formation of Mg-Al spinel, and the optimum addition of V2O5 and Fe2O3 is respectively 2wt% and 3wt%. V2O5 has few effects on cell parameters of Mg-Al spinel; but Fe2O3 can form solid solution with Mg-Al spinel in which Mg-Fe spinel can also dissolve to form continuous solid solution. Therefore, with the increase of Fe2O3 content, the cell parameters of Mg-Al approximately present linear increase. 相似文献

The effect of ammonium nitrate concentration in the citric acid biosynthesis by Aspergillus niger NC-12 in single-stage continuous cultures with biomass retention was investigated. Experiments were carried out in a BIOMER
laboratory fermenter with 5 dm3 working volume. At the initial stage of each cultivation, the substrate in the bioreactor contained 1.5 g NH4NO3 dm−3. After 120 h onwards, the bioreactor was fed continuously at a constant dilution rate of 0.009 h−1. NH4NO3 concentration in the feed was varied from one culture to another, ranging between 0.5 g dm−3 and 2.5 g dm−3. Promising results were obtained when NH4NO3 concentration of 1.5 g dm−3 was used. The observed concentration of citric acid (c
P) and yield of citric acid with respect to the introduced sucrose (Y
P/S) were 117.88 g dm−3 and 78.59 %, respectively. The efficiency coefficient of citric acid biosynthesis (K
ef) was very high, amounting to 83.38.

In DNA aptamer selection, existing methods do not discriminate aptamer sequences based on their binding affinity and function and the reproducibility of the selection is often poor, even for the selection of well-known aptamers like those that bind the commonly used model protein thrombin. In the present study, a novel single-round selection method (SR-CE selection) was developed by combining capillary electrophoresis (CE) with next generation sequencing. Using SR-CE selection, a successful semi-quantitative and semi-comprehensive aptamer selection for thrombin was demonstrated with high reproducibility for the first time. Selection rules based on dissociation equilibria and kinetics were devised to obtain families of analogous sequences. Selected sequences of the same family were shown to bind thrombin with high affinity. Furthermore, data acquired from SR-CE selection was mined by creating sub-libraries that were categorized by the functionality of the aptamers (e. g., pre-organized aptamers versus structure-induced aptamers). Using this approach, a novel fluorescent molecular recognition sensor for thrombin with nanomolar detection limits was discovered. Thus, in this proof-of-concept report, we have demonstrated the potential of a “DNA Aptaomics” approach to systematically design functional aptamers as well as to obtain high affinity aptamers. 相似文献

Herein, the photocatalytic hydrosilylation of alkynes and alkenes under continuous flow conditions is described. By using 0.2 mol % of the developed [Cu(dmp)(XantphosTEPD)]PF6 under blue LEDs irradiation, a large panel of alkenes and alkynes was hydrosilylated in good to excellent yields with a large functional group tolerance.
