We report on the development of a bi-layer bi-enzyme biosensor architecture using different peroxidases and alcohol oxidase
from Hansenula polymorpha C-105 as biological recognition elements. The sensor architecture comprises a first layer containing either horseradish peroxidase
or royal palm tree peroxidase crosslinked with an Osmium complex-modified redox hydrogel. On top, a second layer was formed
by electrochemically induced precipitation of a cathodic electrodeposition paint simultaneously entrapping alcohol oxidase
isolated from a genetically modified strain of Hansenula polymorpha C-105. The sensor architecture was optimized with respect to effective electron transfer and stability of the enzyme. The
main characteristics of the biosensors are an apparent maximal current Imaxapp of 572–940 nA, an apparent Michaelis constant KMapp of 9.5 mM, a sensitivity of 60–98 nA mM−1 and an improved operational stability represented by a deactivation constant of 1.5–2.0 × 10−4min−1. 相似文献

temperature region at room temperature and are highly responsive to changes in ambient temperature and relative humidity,

光学影像技术是法庭科学物证检验分析的重要技术手段,具有无损、原位、快速等优势。近年来,随着光学影像技术的不断发展,一些新技术、新方法不断被引入法庭科学领域并付诸应用。光学相干层析技术(简称OCT技术)是一种光学断层成像技术,具有无损、高分辨、快速、断层成像的特点,特别是其可以无损地得到材料或生物组织内部的结构信息,突破了传统二维成像只能进行物质表面分析的局限,因此可成为一种很有前景的新型法庭科学光学影像技术。本文首先介绍了自主搭建的一套三维OCT成像系统,并着重介绍了基于该系统应用于法庭科学油漆物证检验、胶带指纹显现和毛囊特征分析等方面的研究。结果表明,OCT技术非常适合用于法庭科学物证检验分析,作为其他检验的先导技术手段,获取样品内部信息,提取新型光学特征参数。由于其采用光纤化技术,还有望实现便携化、小型化,在现场勘查中开展相关应用。 相似文献

Laser-induced breakdown spectroscopy (LIBS) system was developed for determination of toxic metals in wastewater collected from local paint manufacturing plant. The plasma was generated by focusing a pulsed Nd:YAG laser at 1064 nm on the solid residue from wastewater collected from paint industry. The concentration of different elements of environmental significance like, lead, copper, chromium, calcium, sulphur, magnesium, zinc, titanium, strontium, nickel, silicone, iron, aluminum, barium, sodium, potassium and zirconium, in paint wastewater were 6, 3, 4, 301, 72, 200, 20, 42, 4, 1, 35, 120, 133, 119, 173, 28 and 12 mg kg−1, respectively. The evaluation of potential and capabilities of LIBS as a rapid tool for paint industry effluent characterization is discussed in detail. Optimal experimental conditions were evaluated for improving the sensitivity of our LIBS system through parametric dependence study. The laser-induced breakdown spectroscopy (LIBS) results were compared with the results obtained using standard analytical technique such as inductively coupled plasma emission spectroscopy (ICP). The relative accuracy of our LIBS system for various elements as compared with ICP method is in the range of 0.03-0.6 at 2.5% error confidence. Limits of detection (LOD) of our LIBS system were also estimated for the above mentioned elements. 相似文献

Laser paint removal in a water environment does not diffuse ablation pollution products into air. Characteristics of water, such as high specific heat and heat flux, generate different effects of the laser paint removal than in an air environment. In this study, the effects of air and water environments on the mechanism and effect of laser paint removal are analyzed and compared experimentally and theoretically. In air, thermodynamic ablation causes removal of paint, whereas in water, stress coupled with plasma shock waves cause tear and splash removal of paint layers after fracture and damage. Fracture and pressure thresholds of the paint and substrate, respectively, indicate the optimum energy density range for laser paint removal in water, providing a reference for engineering applications. 相似文献

很多轻工业产品需要用到油性油漆,其中的重金属元素的含量是否超标将会直接影响到产品质量及其安全性。因此发展轻工产品中重金属含量的快速检测技术,对促进国民经济的发展和保障消费者生命安全具有重要的意义。我们利用激光诱导击穿光谱技术实现了对油漆中有害重金属含量的快速定性和定量分析。通过观测激光等离子体中重金属原子辐射强度可以定性测量重金属含量的大体水平。然后,在相同检测条件下,将已知浓度的标准样品作为参考,制定出各元素的校正曲线,使用外标法,就能够实现定量分析。对Pb、Cr、Cd三种有害重金属元素,目前检出限分别达到9、3.5和23ppm的水平,分析相对误差小于10%,单次测量时间短于2分钟。汞的结果不太理想,检测灵敏度相对偏低,与其原子辐射寿命较短有关。我们将采取脉冲放电延长汞原子辐射弛豫时间的措施来改进汞元素的分析灵敏度,使其达到较高水平。 相似文献

为建立高分辨率电感耦合等离子体质谱法(HR-ICP-MS)测定涂料中砷化合物形态分布的分析方法,应用离子交换树脂和溶剂萃取相结合的分离技术分离涂料中As(Ⅲ)、As(Ⅴ)、MMA(甲基胂酸)、DMA(二甲基胂酸)等4种砷化合物,试液直接用HR-ICP-MS法同时测定上述4种砷化合物,在高分辨质谱测量模式下避免了大量的质谱干扰,考察了采用内标元素对基体效应的校正,应用标准加入法进行定量分析,确定了实验的最佳测定条件。结果表明,方法的检出限为0.002μg/g,样品的加标回收率为98.2%~104.2%,相对标准偏差为0.72%~2.61%。该法具有简单、快速、准确等优点,应用涂料中砷化合物的4种不同形态砷的测定,结果满意。 相似文献

There is much demand for improvement in the performance of a hard disk drive (HDD) along with recent rapid developments of information technology. While high-speed disk rotation of a HDD is necessary to accommodate such needs, it causes disk flutter induced by pressure fluctuation on disks and degrades reliability of a HDD. In order to understand the mechanism of the fluttering phenomenon, it is important to know pressure field on the rotating disk. However, it is impossible to measure the pressure by ordinary methods such as pressure taps. Pressure-sensitive paint (PSP) is a pressure measurement technique based on the oxygen quenching of luminescence and enables us to measure the pressure non-invasively. In general, however, the temperature sensitivity of PSP makes it difficult to measure the precise pressure on the surface with temperature distribution. We measured the time-averaged pressure on the disk rotating at 10 000-20 000 rpm for the first time by adopting a temperature-insensitive PSP composed of pyrene sulfonic acid (PySO3H) as a luminophore. It was found that the pressure forms a concentric circular distribution and decreases toward the center of the disk. Additionally, we elucidate how disk rotational speed and spacing between co-rotating disks influence on the pressure field. 相似文献
