Certain contaminants at trace concentrations in surface waters can have dramatic effects on the hormonal system of organisms in the aquatic environment. Therefore, immunoanalytical methods at a very low limit of detection (LOD) and a low limit of quantification (LOQ) are becoming more and more important for environmental analysis and especially for monitoring drinking water quality. Environmental monitoring of antibiotics, hormones, endocrine disrupting chemicals, and pesticides in real water samples (e.g. surface, ground or drinking water) with difficult matrices places high demands on chemical analysis. Biosensors have suitable characteristics such as efficiency in allowing very fast, sensitive, and cost-effective detection. Here we describe an assay optimization process with a fully automated immunoassay for estrone which resulted in a LOD below 0.20 ng L−1 and a LOQ below 1.40 ng L−1. In contrast to common analytical methods such as GC-MS or HPLC-MS, the biosensor used requires no sample pre-treatment and pre-concentration. The very low validation parameters for estrone are the result of the continuous optimization of the immunoassay. The basis of our sensitive assay is the antibody with a high affinity constant towards estrone. During the optimization process, we reduced the amount of antibody per sample and improved the chip surface modification. Finally, this proceeding led to a calibration routine with an amount of antibody of only 3.0 ng per sample (sample volume: 1.0 mL). The reduction of the amount of antibody per sample results in better validation parameters (LOD, LOQ, and IC50), but this reduction leads to the current device-related limitation of the River Analyser (RIANA).For some endocrine disrupting compounds, no effect levels (NOELs) in the lower nanogram per liter range are reported. This defines the challenge, which analytical methods have to compete with and our RIANA instrument with its improved sensitivity for the detection of a single hormone in the lower nanogram per liter range is a powerful tool in aquatic analytics in addition to the common analytical methods.  相似文献

We report the syntheses and structural aspects of cyclodextrin host–guest inclusion compounds containing linear secondary alkylamines (dipropyl, dibutyl, dipentyl, dihexyl, and dioctyl) at 25 °C. Elemental analysis, 13C CP-MAS NMR spectroscopy, and powder X-ray diffraction analysis confirm the inclusion process. The basic host structure of the products is similar to that of typical cyclodextrin inclusion systems. 13C MAS NMR experiments show a different resonance pattern for the confined guest molecules with respect to the amine in the liquid phase. The presence of different resonance signals for the homologous carbon atoms of both dialkylamine branches is evidence for the non-symmetric location of the amine in the cyclodextrin channels.  相似文献

Data have been summarized of several studies on the separation of isomeric pyrrolizidine alcohols by GLC using liquid polar stationary phases. It was shown that the order of emergence of isomers from the chromatographic column is determined to a significant extent by competition of intermolecular hydrogen bonds formed in the sorbate-sorbent systems and intramolecular hydrogen bonds in the molecules of the same pyrrolizidine alcohols. The preference for one or other type of hydrogen bond depends on the stereochemistry of the pyrrolizidine alcohols. Analysis of the geometric conditions for the formation of intramolecular hydrogen bonds in the investigated compounds in conjunction with chromatographic resolution data enables their configurations to be assigned. The anomalously short retention times of highly strained 5-hydroxyalkyl-3-methylpyrrolizidines are explained by the existence in them of a bicyclic conformation predominantly with a trans linkage and with f avora ble geometric conditions for forming intramolecular hydrogen bonds in them.  相似文献

The crystalline one‐dimensional compound, [RhII2(bza)4(pyz)]n ( 1 ) (bza=benzoate, pyz=pyrazine) demonstrates gas adsorbency for N2, NO, NO2, and SO2. These gas‐inclusion crystal structures were characterized by single‐crystal X‐ray crystallography as 1 ?1.5 N2 (298 K), 1 ?2.5 N2 (90 K), and 1 ?1.95 NO (90 K) under forcible adsorption conditions and 1 ?2 NO2 (90 K) and 1 ?3 SO2 (90 K) under ambient pressure. Crystal‐phase transition to the P space group that correlates with gas adsorption was observed under N2, NO, and SO2 conditions. The C2/c space group was observed under NO2 conditions without phase transition. All adsorbed gases were stabilized by the host lattice. In the N2, NO, and SO2 inclusion crystals at 90 K, short interatomic distances within van der Waals contacts were found among the neighboring guest molecules along the channel. The adsorbed NO molecules generated the trans‐NO???NO associated dimer with short intermolecular contacts but without the conventional chemical bond. The magnetic susceptibility of the NO inclusion crystal indicated antiferromagnetic interaction between the NO molecules and paramagnetism arising from the NO monomer. The NO2 inclusion crystal structure revealed that the gas molecules were adsorbed in the crystal in dimeric form, N2O4.  相似文献

The ability of 4-methylpyrimidinium ylides (as 1,3-dipoles) to react with activated non-symmetrical substituted dipolarophiles (alkenes and alkynes) is presented. 4-Methylpyrimidinium ylides did not react with alkenes. With alkynes the reactions are regiospecific, a single regioisomer being obtained. A possible mechanism for the reaction pathway is proposed. For the first time in the pyrimidinium ylides series both isomers resulting from bonding to the 2-and 6-positions of the heterocycle ring were obtained. The appropriate conditions in order to increase the selectivity of one of the isomers were determined.  相似文献

The palladium(0)‐catalyzed polyaddition of bifunctional vinyloxiranes [1,4‐bis(2‐vinylepoxyethyl)benzene ( 1a ) and 1,4‐bis(1‐methyl‐2‐vinylepoxyethyl)benzene ( 1b )] with 1,3‐dicarbonyl compounds [methyl acetoacetate ( 4 ), dimethyl malonate ( 6 ), and Meldrum's acid ( 8 )] was investigated under various conditions. The polyaddition of 1 with 4 was carried out in tetrahydrofuran with phosphine ligands such as PPh3 and 1,2‐bis(diphenylphosphino)ethane (dppe). Polymers having hydroxy, ketone, and ester groups in the side groups ( 5 ) were obtained in good yields despite the kinds of ligands employed. The number‐average molecular weight value of 5b was higher than that of 5a . The polyaddition of 1b and 6 was affected by the kinds of ligands employed. The corresponding polymer 7b was not obtained when PPh3 and 1,2‐bis(diphenylphosphino)ferrocene were used. The polyaddition was carried out with dppe as the ligand and gave polymer 7b in a good yield. The molecular weight of the polymer obtained from 1b and 8 was much higher than those of polymers 5b and 7b . The polyaddition with Pd2(dba)3 · CHCl3/dppe as a catalyst (where dba is dibenzylideneacetone) produced polymer 9b in a 92% yield (number‐average molecular weight = 45,600). The stereochemistries of all the obtained polymers were confirmed as an E configuration by the coupling constant of the vinyl proton. © 2002 Wiley Periodicals, Inc. A method for the quantitative determination of the major anionic constituents of fountain solutions, typically mono-, di- and hydroxycarboxylates, alkylbenzenesulfonates, and inorganic anions, including orthophosphate and polyphosphates, is presented here for the first time. The analytical problems arising from extensive co-elution of many of these analytes on an ion-exchange column have been resolved through a combination of (i) careful selection of the concentration gradient of the sodium hydroxide eluent; (ii) parallel analysis by ion-exclusion chromatography; and (iii) determination of total phosphorus by inductively coupled plasma atomic emission spectrometry.  相似文献

The reaction of the cluster salts [Cp(2*) Nb(CO)(2)](n)[Co(11)Te(7)(CO)(10)] (Cp*=C(5)Me(5); n=1, 2) with excess PMe(2)Ph gave the neutral, dark brown clusters [Co(11)Te(7)(CO)(6)(PMe(2)Ph)(4)] (5) and [Co(11)Te(7)(CO)(5)(PMe(2)Ph)(5)] (6) with 147 metal valence electrons. The new compounds were characterized by IR spectroscopy, elemental analyses, and mass spectrometry. The molecular structure of 6 was determined by X-ray crystallography. Like its precursor anion, it consists of a pentagonal-prismatic [Co(11)Te(7)] core, but with a ligand sphere composed of five CO and five PMe(2)Ph ligands. Detailed electrochemical studies of both reactions reveal that a stepwise substitution of CO ligands in the initial cluster anions takes place leading to intermediate [Co(11)Te(7)(CO)(10-m)(PMe(2)Ph)(m)](n-) ions (m=1-5; n=1, 2). Each of these intermediates is distinguished by at least one oxidation and two reduction waves, giving rise to a total of 21 redox couples and 27 electroactive species. The electron sponge character of the new compounds is particularly pronounced in 5, which exhibits charges n between +1 and -4 corresponding to metal valence electron counts of between 146 and 151.  相似文献

A method has been developed for species-selective analysis of organotin compounds in solid, biological samples. The procedure is based on accelerated solvent extraction (ASE) of analytes and includes extraction of the tin species with a methanol–water (90% methanol) solution of acetic acid/sodium acetate containing tropolone (0.03% w/v), their ethylation with NaBEt4, and separation and detection by GC–FPD. The analytical procedure was optimized with an unspiked sample of harbor porpoise (Phocoena phocoena) liver. Effects of ASE operational variables (extraction temperature and pressure, solvent composition, number of static extraction steps) are discussed. Method detection limits (MDL) were in the range 6–10 ng(Sn) g–1 dry weight and 7–17 ng(Sn) g–1 dry weight for butyl- and phenyltin compounds, respectively. Recoveries were comparable with or better than those obtained by use of other procedures reported in the literature. The analytical procedure was validated by analysis of NIES No. 11 (fish tissue) certified reference material.  相似文献
