脂质体由于其特殊的结构和性能,是一种很有发展潜力的药物载体,在医药和化妆品等行业具有广阔的应用前景。超临界逆向蒸发法是用超临界流体代替有机溶剂制备脂质体的方法,具有对水溶性药物脂质体制备过程简单、包覆率高等特点。以葡萄糖为模型药物,利用该方法制备出了葡萄糖脂质体,并详细考察了不同工艺条件对脂质体粒径和包覆率的影响。结果表明:用超临界逆向蒸发法可成功地制备出最小粒径为290nm、包覆率最高可达41.3%的葡萄糖脂质体。压力、温度和平衡时间对粒径和包覆率都有较大的影响。压力在10~30MPa时,随压力的增加,脂质体粒径显著减小至某一值后,基本趋于稳定,而包覆率逐渐增加,达到25.7%~27.5%,随后包覆率逐渐减小;温度在35~65℃时,随温度的增加粒径显著减小至最小值,然后逐渐增大,而包覆率一直增大,最高可达41.3%;平衡时间在15~45min时,随平衡时间的增加,粒径相对稳定,而包覆率显著增加,超过45min之后,粒径显著增加,包覆率显著下降。  相似文献

Thirteen thiomorpholine-bearing compounds were designed and synthesized as dipeptidyl peptidase IV(DPP-IV) inhibitors, with natural and non-natural L-amino acids as the starting materials.Their structures were characterized by 1H NMR,13C NMR and HR-MS.The target compounds were screened for the DPP-IV inhibition,and the preliminary SAR result was obtained.Particularly, compounds 4c,4d and 4f with good DPP-IV inhibition in vitro were further evaluated through a mouse oral glucose tolerance test (OGTT).The preliminary result showed the potential value for further studies on those thiomorpholine-bearing compounds as DPP-IV inhibitors.  相似文献

The 2‐(hydroxymethyl)pyridine modified C60 (PY‐C60) and methoxydiglycol modified C60 (MDG‐C60) are synthesized using Bingel‐Hirsch reaction and characterized by nuclear magnetic resonance (NMR) and mass spectra. PY‐C60 and MDG‐C60 can bind to glucose oxidase (GOx) and quench the fluorescence of tryptophan (Trp) residue in GOx through static mechanism. The conformation of GOx is disturbed after formation of complex with these fullerene derivatives. Kinetic analysis indicates that PY‐C60 and MDG‐C60 may affect the catalytic activity of GOx with a partial mixed‐type inhibition mechanism. In the plasma glucose concentration range (3.6–5.2 mmol·L?1), PY‐C60 may significantly accelerate the catalytic velocity of GOx, however, MDG‐C60 exerts almost no obvious change to the initial velocity of GOx, suggesting that elaborate design of molecular structure of fullerene derivative is very important for regulating the biological activity of fullerene‐enzyme complex.  相似文献

A simple and effective glucose biosensor based on immobilization of glucose oxidase (GOD) in graphene (GR)/Nafion film was constructed. The results indicated that the immobilized GOD can maintain its native structure and bioactivity, and the GR/Nafion film provides a favorable microenvironment for GOD immobilization and promotes the direct electron transfer between the electrode substrate and the redox center of GOD. The electrode reaction of the immobilized GOD shows a reversible and surface‐controlled process with the large electron transfer rate constant (ks) of 3.42±0.08 s?1. Based on the oxygen consumption during the oxidation process of glucose catalyzed by the immobilized GOD, the as‐prepared GOD/GR/Nafion/GCE electrode exhibits a linear range from 0.5 to 14 mmol·L?1 with a detection limit of 0.03 mmol·L?1. Moreover, it displays a good reproducibility and long‐term stability.  相似文献

开发了一种小型化人体血糖浓度无创检测系统,该系统可实现人体血糖浓度的快速、实时、无创检测,主要由嵌入式系统、夹具、光源系统和微型光栅光谱仪四部分组成。提出了一种光源稳压驱动电路方案,可满足多种光谱采集条件对光源的要求。同时提出了一种一体化夹具设计方案,不仅简化了系统的光学结构,而且提高了测量条件的可重复性。分别用小波去噪和核偏最小二乘法对采集的光谱数据进行预处理和数学建模,并对系统进行性能评价,血糖浓度预测值与标准值的相关系数为0.95,预测均方根误差为0.6mmol.L-1,系统的重复性较好。  相似文献

Abstract

The alkaline ferricyanide and the Park-Johnson's methods were studied in order to determine glucose oligomers separated by liquid chromatography in nanomole quantities. In the alkaline ferricyanide method, high sensitivity was obtained by measuring ultraviolet absorbance of the reduced reagents. The presence of some kinds of salts or alcohols did not affect the colour development of the reagents. The applicability of the present measuring system to an automatic sugar analyzer has been demonstrated by examining several glucose oligomers.  相似文献

Abstract

Acidification of a solution of glucose oxidase in 35 percent glycerol with 10 percent sulfuric acid and a gel filtration of this acidified glucose oxidase solution yielded apo-glucose oxidase with low residual enzyme activity. Further treatment with charcoal gave apo-glucose oxidase which is devoid of enzyme activity. Using this apo-glucose oxidase, it was possible to measure flavin adenine dinucleotide amperometrically at extremely low concentrations (10?12 M) with ease, rapidity, and convenience.  相似文献

Abstract

A very simple flow injection analysis system for direct determination of glucose in blood serum based on Trinder's reaction is described. The sera samples (15 μl) can be injected directly to the system without the deproteinization or the use of a dialyzer.

Calibration curves are linear in the range 50–400 mg/dl. The sampling frequency is 60 samples per hour. Results obtained by the proposed procedure are compared with those acquired at a local hospital using their routine glucose procedure also based on Trinder's reaction. It is shown that a better mix between sample and reagents is achieved using the single bead string reactor (SBSR).  相似文献

A glucose oxidase (GOx)‐mediated glucose metabolism was in vitro mimicked and employed to regulate the self‐assembly of peptide‐based building blocks. In this new stimuli‐responsive self‐assembly system, two peptide‐based building blocks, respectively, having aspartic acid (gelator 1 ) and lysine (gelator 2 ) residues were designed and prepared. When adding glucose and GOx to the aqueous solution of gelator 1 or the self‐assembled fibrillar hydrogel of gelator 2 to construct glucose metabolism system, the metabolic product (gluconic acid) can trigger the protonation of the peptide molecules and induce the phase transitions of gelators 1 (sol‐gel) and 2 (gel‐sol). Because this glucose metabolism regulated peptide self‐assembly is built on the oxidation of glucose, it can be used as a simple visual biosensor for glucose detection.  相似文献

ABSTRACT|The catalytic activity of various mimetic enzymes instead of the peroxidase have been investigated by 4-aminoantipyrine (4-AAP) and 2, 3, 4-trichlorophenol (TCP) to form a dye utilizing hydrogen peroxide as hydrogen acceptor. The different Chlorophenolic derivatives, which act as a substrate in β-CD-hemin-H2O2-4-AAP catalytic reaction, have been systematically studied.|Meanwhile, the relationship of structure-effect for the β-CD-hemin as catalyst, and chlorphenols as substrate has been respectively discussed. The mechanism of catalytic reaction has been investigated. The results showed that β-CD-hemin was the best mimetic enzyme for peroxidase among those tested and TCP was a good substrate for the determination of hydrogen peroxide with β-CD-hemin. The method for the determination of hydrogen peroxide was proposed using 4-AAP-TCP system with β-CD-hemin as catalyst. A linear calibration graph was obtained over the H2O2 concentration of 4.8×10-?8-7.7×10-?5M, and the relative standard deviation at a H2O2 concentration of 2.8×10-?5M was 2.5%. The apparent molar absorptivity of the chromogenic reaction for H2O2 was 1.54× 104 L.mol-?1.cm?1. Satisfactory results were obtained in the determination of H2O2 in synthetic samples by this method.

Also, the method was coupled with the glucose oxidation reaction to determination glucose in human serum.  相似文献
