Cyclosporine, tacrolimus, sirolimus, and everolimus are commonly used immunosuppressants following organ transplantation. Their monitoring is used to determine the optimal dose for therapeutic effectiveness and minimize toxicity. High-performance liquid chromatographic–tandem mass spectrometry with positive electrospray ionization and multiple reaction monitoring mode was validated for the determination of cyclosporine A, tacrolimus, sirolimus, and everolimus in whole blood. A C18 analytical column was employed with a gradient elution of pH 4.0 aqueous 10?mmol/L ammonium acetate and acetonitrile. For the pretreatment of whole blood, simple protein precipitation was used with methanol:zinc sulfate. The calibration curves were linear from 20.0 to 1000?ng/mL for cyclosporine A, 1.0 to 50?ng/mL for tacrolimus and sirolimus, and 1.0 to 30?ng/mL for everolimus. The intra-assay precision and inter-assay precision were less than 15%. The method provides reliable and reproducible results according to the linearity, precision, accuracy, recovery, and matrix effects. The method has been introduced to routine clinical practice in Slovakia for the determination of immunosuppressants in patients after kidney transplantation.  相似文献

Triphenylphosphine-mediated metal-free trifluoromethylthiolation and difluoromethylthiolation of thiols by CF3SO2Cl and CHF2SO2Cl to synthesize trifluoromethyl disulfides and difluoromethyl disulfides, respectively, was achieved at room temperature. Iodine generated in situ from iodide facilitated this reaction via the formation of iodotriphenylphosphonium iodide which could serve as a reducing agent in this transformation. Readily available reagents and mild reaction conditions without transition-metals allow this protocol to be more practical than traditional methods.  相似文献

One palladium dichloride adduct of a phosphine-pyridine ligand N-diphenylphosphanyl-2-aminopyridine (L1) [(L1)PdCl2] (1) has been prepared and structurally characterized. Compound 1 can be used as an effective catalyst for the Suzuki-Miyaura cross-coupling reactions of unreactive aryl chlorides with aryl boronic acids, and worked much better than its mono- or bidentate phosphine ligands. The reactions with a wide scope of substrates proceeded to give desired products in good to excellent yields.  相似文献

A practical copper-catalyzed N-arylation of NH-containing heterocycles with arenediazonium tetrafluoroborates has been developed using CuCl as catalyst and K2CO3 as additive under ligand-free conditions. This reaction system has wide substrate scope including imides, 1H-pyrazole, 1H-tetrazoles, 1,2-benzisothiazol-3(2H)-one, and related sulfonamides and gives moderate to excellent yields (up to 95%) of the desired products. This strategy is very general, simple, environmentally friendly, and tolerant of oxygen.  相似文献

N-Oxysulfonyl formamidine derivatives have been efficiently synthesized under mild conditions through direct condensation of various sulfamates and formamides in the presence of thionyl chloride. The scope of this reaction was investigated, and a plausible mechanism was proposed. The resulting N-oxysulfonyl formamidines can be converted to sulfamates through appropriate deprotection.  相似文献

Dynamic mechanical behaviour of natural rubber-silica composites was studied by a frequency sweep method at different temperatures (40 °C,70 °C and 100 °C) using a dynamic mechanical analyzer and a rotorless rheometer, RPA, in an attempt to establish a correlation between the two. The composites with silica content up to 40 phr were studied. It was found that the dependence of dynamic modulus on the frequency as obtained from both the instruments followed a similar trend. This suggests that the dynamic mechanical properties of rubber compounds can be determined even during curing. A correlation could be arrived at between the two sets of data, making it possible to predict one set knowing the other. The impact of silane coupling agent, bis (3-triethoxysilylpropyl tetrasulphide), TESPT, on viscoelasticity was also investigated. The mechanical properties were improved in the presence of TESPT. Additionally, an increase in thermal stability was also observed in the presence of TESPT. Scanning electron micrographs showed the better filler dispersion in the case of silane-coupled silica composites.  相似文献

New foaming method, enhanced by microwave irradiation, was elaborated and applied to obtain porous poly(vinyl chloride) and its composites with fine cell structure. The so called “thermal runaway” effect was observed during the heating of poly(vinyl chloride) under microwave irradiation. The temperature of this effect decreases as a result of additives incorporation into polymer matrix. Microwave irradiation allowed effective heating of extruded poly(vinyl chloride) and its composites with carbon black (CB) filler, behind the extruder head and decomposing azodicarbonamide (ADC) to obtain porous products. The use of CB additive to poly(vinyl chloride) significantly increased its ability to be heated under microwave irradiation as well as improved the cell structure and decreased the apparent density of final products.Among additionally used fillers (1 wt%) the montmorillonite caused the apparent density decrease of foamed materials ca. 10%, however beneficially influenced on the quality of cells structure, giving the products with isotropic cells and the highest cell density as well as keeping the tensile strength on similar level as in the case of the materials with CB and ADC only.  相似文献

通过软模板法合成了SBA-16分子筛,采用高温氨气氮化的方法使有序介孔硅材料中的氧原子部分被氮原子取代,得到氮化的SBA-16载体(SBA-16-N)。采用满孔浸渍法制备了镍基催化剂,并将制得的Ni/SBA-16和Ni/SBA-16-N催化剂用于甲烷二氧化碳重整反应。通过透射电镜、氮气物理吸附、X射线衍射、X射线光电子能谱和二氧化碳程序升温脱附等手段研究了载体和催化剂的结构,并利用热重分析对反应之后回收催化剂进行了表征。结果表明,高温氮化后的分子筛中掺入了氮元素,增加了载体的碱性,改善了载体对反应气体的吸附活化能力,增强了载体与金属之间的相互作用,从而提高了催化剂的活性和抗积炭性能。  相似文献
