A facile method based on high-performance liquid chromatography coupled with electrospray ionization tandem mass spectrometry (HPLC/(+)ESI-MSn) has been established for the analysis of polyoxypregnane glycosides in the stems of Marsdenia tenacissima. The data reveals the ability of MSn in the structural elucidation of polyoxypregnane glycosides including the nature of the polyoxypregnane core, the kinds of the substituents and the types of sugar residues. Offline Fourier transform ion cyclotron resonance mass spectrometry (FTICR-MS) is also performed to assign accurate elemental compositions. In this study, eighteen polyoxypregnane glycosides have been investigated. Among these components, five compounds are unambiguously identified as Marsdenoside K, Tencissoside A, B, C and D; two compounds are established as novel compounds based on mass spectral data; and the other eleven compound's structures are tentatively proposed. Furthermore, breakdown curves are constructed to distinguish five pairs of isomers among these eighteen compounds. As far as our knowledge, this is the first report on identification of polyoxypregnane glycosides in the stems of M. tenacissima by HPLC/ESI-MSn directly, which could save time and material consuming efforts in traditional phytochemistry analyses.  相似文献

Due to the increasing understanding of the health benefits and chemopreventive properties of flavonoids, there continues to be significant effort dedicated to improved analytical methods for characterizing the structures of flavonoids and monitoring their levels in fruits and vegetables, as well as developing new approaches for mapping the interactions of flavonoids with biological molecules. Tandem mass spectrometry (MS/MS), particularly in conjunction with liquid chromatography (LC), is the dominant technique that has been pursued for elucidation of flavonoids. Metal complexation strategies have proven to be especially promising for enhancing the ionization of flavonoids and yielding key diagnostic product ions for differentiation of isomers. Of particular value is the addition of a chromophoric ligand to allow the application of infrared (IR) multiphoton dissociation as an alternative to collision-induced dissociation (CID) for the differentiation of isomers. CID, including energy-resolved methods, and nuclear magnetic resonance (NMR) have also been utilized widely for structural characterization of numerous classes of flavonoids and development of structure/activity relationships.The gas-phase ion chemistry of flavonoids is an active area of research particularly when combined with accurate mass measurement for distinguishing between isobaric ions. Applications of a variety of ab initio and chemical computation methods to the study of flavonoids have been reported, and the results of computations of ion and molecular structures have been shown together with computations of atomic charges and ion fragmentation. Unambiguous ion structures are obtained rarely using MS alone. Thus, it is necessary to combine MS with spectroscopic techniques such as ultraviolet (UV) and NMR to achieve this objective. The application of NMR data to the mass spectrometric examination of flavonoids is discussed.  相似文献

Three new lignan glycosides (1-3) were isolated from the stems of Akebia trifoliata. Their structures were elucidated as (7R,8R,7'R,8'R)3,3',5,5'tetramethoxy-4,4'dihydroxy-7,9':7',9-diepoxylignan-4-O-beta-D-glucopyranoside (1), (7S,8S,8'R)-4,4',9-trihydroxy-3,3',5,5'-tetramethoxy-7,9'-epoxylignan-7'-one 9-O-beta-D-glucopyranoside (2), (7R,8R,8'S)-4,4',9-trihydroxy3,3',5,5'-tetramethoxy-7,9'-epoxylignan-7'-one 9-O-beta-D-glucopyranoside (3) by spectral analyses, primarily NMR, MS and CD. The NMR assignments for the compounds were carried out using 1H, 13C, DEPT, COSY, HSQC, HMBC and ROESY NMR experiments.  相似文献

建立了油茶枯饼中两种主要黄酮苷:山奈酚3-0-[2-O-β-D-木糖-6-0-α-L-鼠李糖]-β-D-葡萄糖苷(Ⅰ)和山奈酚3-0-[2-O-β-D-半乳糖-6-0-α-L-鼠李糖]-β-D-葡萄糖苷(Ⅱ)的高效液相色谱测定方法。采用C18 Column(BDS,Hypersil,250mm×4.6 mm),流动相为V(乙腈)∶V(0.1%H3PO4)=20∶80,流速1 mL/min;检测波长266 nm。结果表明,黄酮苷Ⅰ的线性范围为60-2000 mg/L(R=0.9993,n=6),平均回收率99.5%,RSD为1.6%;黄酮苷Ⅱ的线性范围为30-1200mg/L(R=0.9995,n=6),平均回收率100.7%,RSD为1.2%。该方法可用于检测油茶枯饼及其提取物中两种主要黄酮苷。  相似文献

Two new coumarin biosides, tert-O-β-D-apiofuranosyl-(1→6)-O-β-D-glucopyranosyl-byakangelicin (1) and 2′-O-β-D-apiofuranosyl-(1→6)-β-D-glucopyranosyl-peucedanol (2), were isolated from the fresh roots of Angelica dahurica. From the leaves of Globularia alypum, three new phenylethyl glycosides, namely galypumosides A (=2‐(3,4‐dihydroxyphenyl)ethyl Oα‐rhamnopyranosyl‐(1→3)‐4‐O‐[(E)‐caffeoyl]‐6‐O‐[(E)‐p‐coumaroyl]‐β‐glucopyranoside; 1 ), B (=2‐(3,4‐dihydroxyphenyl)ethyl Oα‐rhamnopyranosyl‐(1→3)‐4‐O‐[(E)‐caffeoyl]‐6‐O‐[(E)‐feruloyl]‐β‐glucopyranoside; 2 ), and C (=2‐(3,4‐dihydroxyphenyl)ethyl Oα‐rhamnopyranosyl‐(1→3)‐4‐O‐[(E)‐caffeoyl]‐6‐O‐menthiafoloyl‐β‐glucopyranoside; 3 ), were isolated, together with two known phenylethyl glycosides, calceolarioside A and verbascoside. Eight iridoid glucosides, catalpol, globularicisin, globularin, globularidin, globularinin, globularimin, lytanthosalin, and alpinoside, a flavon glycoside, 6‐hydroxyluteolin 7‐O‐sophoroside, a lignan glycoside, syringaresinol 4′‐Oβ‐glucopyranoside, and a phenylpropanoid glycoside, syringin, were also obtained and characterized. The structures of the isolates were elucidated on the basis of 1D‐ and 2D‐NMR experiments as well as HR‐MALDI‐MS.  相似文献

Phytochemical analyses were carried out on the rhizomes of Clintonia udensis (Liliaceae) with particular attention paid to the steroidal glycoside constituents, resulting in the isolation of three new polyhydroxylated spirostanol glycosides, named clintonioside A ( 1 ), B ( 2 ), and C ( 3 ). On the basis of their spectroscopic data, including 2D‐NMR spectroscopy, in combination with acetylation and hydrolytic cleavage, the structures of 1 – 3 were determined to be (1β,3β,23S,24S,25R)‐1,23,24‐trihydroxyspirost‐5‐en‐3‐yl Oβ‐D ‐glucopyranosyl‐(1→4)‐O‐[α‐L ‐rhamnopyranosyl‐(1→2)]‐β‐D ‐glucopyranoside ( 1 ), (1β,3β,23S,24S)‐3,21,23,24‐tetrahydroxyspirosta‐5,25(27)‐dien‐1‐yl Oα‐L ‐rhamnopyranosyl‐(1→2)‐O‐[β‐D ‐xylopyranosyl‐(1→3)]‐β‐D ‐glucopyranoside ( 2 ), and (1β,3β,23S,24S)‐21‐(acetyloxy)‐24‐[(6‐deoxy‐β‐D ‐gulopyranosyl)oxy]‐3,23‐dihydroxyspirosta‐5,25(27)‐dien‐1‐yl Oα‐L ‐rhamnopyranosyl‐(1→2)‐O‐[β‐D ‐xylopyranosyl‐(1→3)]‐β‐D ‐glucopyranoside ( 3 ).  相似文献

Phytochemical investigation is very valuable for the ethnomedicinally important plants Bauhinia racemosa Lam (BR) and Cordia dichotoma Linn (CD) used for the cure of variety of ailments. This study was thus designed for phytochemical investigation of BR bark and CD leaves. Phytoconstituents were isolated from the methanolic extracts of the plants by column chromatography using silica gel as stationary phase. The structures had been established on the basis of their physicochemical and spectral data, i.e. IR, 1H NMR, 13C NMR and MS. Elution of the columns with different solvents furnished six compounds (16) from the methanolic extract of BR bark and three compounds (79) from the methanolic extract of CD leaves which were structurally elucidated. The present phytochemical investigation reported several new compounds useful in increasing the existing knowledge of phytoconstituents from BR bark and CD leaves which is very valuable, as these drugs are used in the Indian traditional systems of medicine.  相似文献

Two new polyacetylene glycosides, tangshenyne A (1) and tangshenyne B (2), along with six known polyacetylenes were isolated from an 85% MeOH extract of the roots of Codonopsis tangshen Oliv. The chemical structures of the new compounds were determined on the basis of extensive spectroscopic analyses, including UV, IR, 1D and 2D NMR (1H–1H COSY, HSQC and HMBC) and HR-ESI-MS.  相似文献
