A sensitive electrochemical DNA biosensor based on a mixed monolayer structure self-assembled at nanoporous gold (NPG) electrode surface was prepared for Escherichia coli (E. coli) detection. The NPG was fabricated on gold electrode, onto which thiolated oligonucleotides (SH-DNA) and mercaptohexanol (MCH) were covalently linked forming a mixed self-assembled monolayer (SAM). The hybridization between the SH-DNA/MCH modified biosensor and E. coli DNA was monitored with differential pulse voltammetry measurement using methylene blue (MB) as the hybridization indicator. The biosensor can detect 1 × 10?12 M DNA target and 50 cfu/μL E. coli without any nucleic acid amplification steps. The detection limit was lowered to 50 cfu/mL after 5.0 h of incubation.  相似文献

In this paper, a three dimensional ordered macroporous self-doped polyaniline/Prussian blue (3DOM SPAN/PB) bicomponent film was fabricated via the inverted crystal template technique using step-by-step electrodeposition. In this bicomponent film, PB not only acted as a redox mediator, but also presented increased stability in neutral or weak alkaline solution by the protection of SPAN layer on the top. A novel glucose biosensor was fabricated based on the large active surface area and excellent conductivity possessed by the 3DOM SPAN/PB film. The applying experimental conditions of the glucose biosensor have been optimized. Under the optimal conditions, the biosensor showed a wide linear range over three orders of magnitude in glucose concentrations (from 2 to 1600 μM) and a low detection limit of 0.4 μM. Moreover, the biosensor exhibited short response time, high selectivity and excellent operation stability, which can be applied to detect the blood sugar in real samples without any pretreatment.  相似文献

ABSTRACT

A new method is described for the determination of cobalt based on the square wave adsorptive stripping voltammetry of Co(II) complexed with hydroxynaphthol blue (HNB) at the static mercury drop electrode. Optimal conditions were found to be: preconcentration potential, -0.500V vs. Ag/AgCl (KCl 3M); preconcentration time, 30 s (with stirring); pulse height, 50 mV; frequency, 100 Hz; scan increment, 4 mV; step time, 0.010 s; supporting electrolyte, HEPES/HCl (0.1 M, pH 7.0-8.0) or triethanolamine/HCl (0.1 M, pH 7.6); concentration of hydroxynaphthol blue, 5.0 × 10?6 M. The response of the system was found to be linear in a range of Co(II) concentrations from 2.0 to 10.0 μg/L. The limit of detection was found to be 1.8 × 10?9 M with 2 minutes of preconcentration time. The effect of various potential interferences were also studied including a variety of cations, anions and organic surfactants. The interferences by Ni(II), and Cr(VI) may be eliminated by addition of EDTA or CDTA and the of Fe(III) and Ti(IV) by fluoride. The merits of the procedure were demonstrated in the analysis of certified and biological samples.  相似文献

ABSTRACT

The interactions of nile blue sulphate (NBS) with nucleic acids, including calf thymus DNA, fish sperm DNA and yeast RNA, were characterized with resonance light-scattering (RLS) measurements by using a common spectrofluorometer. Accordingly a method for the determination of nucleic acids at nanogram levels was established. At pH's of 7.20~7.60 and ionic strengths lower than 0.012, the interactions of NBS with nucleic acids result in three characteristic RLS peaks at 293.4 nm, 349.4 nm and 560.4 nm. Mechanism study shows that these peaks are ascribed to the long range assembly of NBS on the molecular surface of nucleic acids, which depends on pH, ionic strength and the stranded structure of nucleic acids. A Scatchard plot was constructed by using the RLS data, yielding the assembly number and assembly constant being 6.4 and 7.13x106 mol?1 1 for NBS assembly on the molecular surface of calf thymus DNA. The same parameters are 6.6 and 4.58x106 mol?1 1 for the assembly on that of fish sperm DNA, 3.9 and 1.67x106 mol?1 1 on that of yeast RNA, respectively. Linear relationships were found between the enhanced RLS intensity at 293.4 nm and nucleic acid concentration. If 1.2x10?5 mol I?1 NBS was employed, 0~0.80 μg ml?1 calf thymus DNA and fish sperm DNA, 0.20~0.60 μg ml?1 yeast RNA can be determined with the determination limits being 3.2 ng ml?1 for calf thymus DNA, 11.5 ng ml?1 for fish sperm DNA and 38.3 ng ml?1 for yeast RNA, respectively. Abstract

A methylene blue (MB) poly (vinyl chloride) membrane electrode based on MBtetraphenylborate (TPB) ion-pair complex is described. The linear response covers the range 1x10?2 - 1x10?6 mol dm?3 MB solution, with a slope of 56.5±0.5 mv/decade (pH range 3.0-1 0.0). The detection limit is 7.75x 10?7 mol dm?3. The electrode shows stability, good reproducibility and fast response. Interferences from common inorganic cations and some organic bases are negligible. These characteristics of the electrode enabled it to be used successfully for the determination of MB in injection.  相似文献

Abstract

A new nanocomposite was developed by combination of prussian blue (PB) nanoparticles and multiwalled carbon nanotubes (MWNTs) in the matrix of biopolymer chitosan (CHIT). The PB and MWNTs had a synergistic electrocatalytic effect toward the reduction of hydrogen peroxide. The CHIT/MWNTs/PB nanocomposite‐modified glassy carbon (GC) electrode could amplify the reduction current of hydrogen peroxide by ~35 times compared with that of CHIT/MWNTs/GC electrode and reduce the response time from ~60 s for CHIT/PB/GC to 3 s. Besides, the CHIT/MWNTs/PB nanocomposite‐modified GC electrode could reduce hydrogen peroxide at a much lower applied potential and inhibit the responses of interferents such as ascorbic acid (AA) uric acid (UA) and acetaminophen (AC). With glucose oxidase (GOx) as an enzyme model, a new glucose biosensor was fabricated. The biosensor exhibited excellent sensitivity (the detection limit is down to 2.5 µM), fast response time (less than 5 s), wide linear range (from 4 µM to 2 mM), and good selection.  相似文献

Prussian blue has significant application for the construction of electrochemical biosensors. In this work, Prussian blue-reduced graphene oxide modified glass carbon electrodes were successfully fabricated using electrochemical deposition. The high surface area of graphene oxide enhanced the deposition of Prussian blue and the resulting electrocatalytic activity. Infrared spectroscopy and scanning electron microscopy showed that the relatively porous Prussian blue was on the surface of reduced graphene oxide. Cyclic voltammetry showed that Prussian blue-coated reduced graphene oxide composite films improved electron transfer compared to Prussian blue films. The Prussian blue-reduced graphene oxide composite film provided higher response for the reduction of hydrogen peroxide and the oxidation of dopamine compared with the Prussian blue film due to synergistic effects between the reduced graphene oxide and Prussian blue particles. The sensitivity of the electrode was 0.1617 µA µM?1 cm?2. The linear dynamic range extended from 0.5 µM to 0.7 mM dopamine with a limit of detection equal to 125 nM. This work provided a versatile strategy for the design and construction of sensitive amperometric sensors with robust electrocatalytic behavior.  相似文献

Abstract

The coloration reagent - leucomethylene blue, the reduction product of methylene blue, is used to determine the concentration of chlorine dioxide in the presence of Cl2 and anion species such as OCl?, ClO2 ? and ClO3 ?. This simple spectrophotometric method is performed by using the extractant 1,2-dichloroethane at pH 1.3. The linear range of ClO2 measurement extends to 0.95 mgl?1 with a detection limit of 0.02 mgl?1. The presence of chlorine and hypochlorite ion can be masked by oxalic acid. The permissible maximum concentration of ClO2 ? is 2.0 mgl?1, and ClO3 ? anion does not interfere with the measurement.  相似文献

Acid chrome blue K (ACBK) was electropolymerized on the surface of a glassy carbon electrode (GCE) by cyclic voltammetric sweep in the potential range from –0.2 to 0.9 V. The characteristic of poly‐ACBK film was studied by different methods such as electrochemical impedance spectroscopy (EIS) and cyclic voltammetry. This modified electrode showed excellent electrocatalytic response to curcumin with the increase of the electrochemical responses. Under the optimal conditions a good linear voltammetric response could be obtained over the range of 1.0 × 10?7‐7.0 × 10?5 M and the detection limit was got as 4.1 × 10?8 M (S/N = 3). The method was successfully applied for the determination of curcumin in human urinev samples.  相似文献

采用失重法、动电位极化曲线、电化学阻抗谱(EIS)和扫描电子显微镜(SEM)研究了氯化硝基四氮唑蓝(NTBC)在1.0-5.0mo·lL-1HCl溶液中对冷轧钢(CRS)的缓蚀作用.结果表明:NTBC在1.0mo·lL-1HCl溶液中对冷轧钢具有良好的缓蚀作用,且在钢表面的吸附符合Langmuir吸附等温式.缓蚀率随缓蚀剂浓度的增加而增大,但随盐酸浓度和温度的增加而减小.求出了相应的吸附热力学(吸附自由能ΔG0,吸附焓ΔH0,吸附熵ΔS0)和腐蚀动力学参数(腐蚀速率常数k,动力学常数B),并根据这些参数讨论了缓蚀作用机理.动电位极化曲线表明:NTBC为混合抑制型缓蚀剂;EIS谱在高频区呈容抗弧,在低频区出现感抗弧,电荷转移电阻随缓蚀剂浓度的增加而增大.SEM再次表明NTBC对钢在盐酸介质中的腐蚀产生了明显的抑制作用.  相似文献
