The influence of substrate or product level on the initial velocity of cyclodextrin (CD) production by cyclodextringlycosyltransferase from a Brazilian isolate of Bacillus firmus was studied. Our results indicate that the product γ-CD is a stronger inhibitor to the reaction than β-CD. Small saccharides could also inhibit CD production, although to a lesser extent than the products, and maltose was the strongest inhibitor among small saccharides. Increasing substrate concentration resulted in greater reduction on enzyme activity for the formation of β-CD than for γ-CD. We modeled the kinetics of CD production with a set of four reversible reactions including the cyclization/coupling reaction that forms/opens CDs, and three disproportionation reactions. Our model on the initial velocity data explained well the substrate inhibition phenomenon. Kinetic parameters were determined by fitting the initial velocity data into our model.  相似文献

Aqueous-solution complexes of β-cyclodextrin (β-CD) with metoprolol tartrate (MET) have been analysed with 1H NMR and UV–vis spectroscopy. With 1H NMR a [1:1] stoichiometry could be established for the β-CD-MET complex while its stability constant was determined with UV–vis spectroscopy. Powder diffraction data of a polycrystalline sample of the β-CD-MET complex show that a novel product has been formed, likely to be a β-CD-MET [1:1] inclusion complex. Also Hyperchem MM+ molecular-dynamics results suggest an inclusion complex and from 1H NMR data it is inferred that probably the MET is docked in the CD with the formers methoxyethyl-benzene moiety in front. The encapsulation of the acid/base conjugated system salicylic acid(HSA)/salicylate (SA-) by hydroxypropyl--cyclodextrin (HPBCD) has beenstudied through fluorescence emission enhancement measurements in aqueoussolutions at 25 °C. With the aim of analyzing the crucial importance ofa proper and cautious choice of the pH of the medium (i.e. choice of thebuffer), the study has been carried out at pH = 1, 2, 4 and 7. Since the pKa of the HSA/SA- system is 2.95 at 25 °C, the presence of the protonated(HSA) and non-protonated (SA-) forms suitable for inclusion by cyclodextrinvary appreciably within the different pH conditions: 99% HSA,1% SA- at pH = 1, 90% HSA, 10% SA- at pH = 2,10% HSA, 90% SA- at pH = 4, and 0% HSA,100% SA- at pH =7. The association constants K CD : HSA and K CD: SA- have been determined in all cases by using a nonlinear regressionanalysis of the experimental data at three different em. The effectof the pH of the medium on all the equilibria involved as well as in the Kvalues is fully discussed. The 8-anilino-1-naphthalene sulfonate (ANS) +-cyclodextrin (-CD) system, widely reported in literature, hasinitially been studied to check the experimental protocol and the numericalmethod.  相似文献

The detection of phthalates in human biologic fluids remains an important research objective because it provides an important measure of an individual’s exposure to this class of compounds, which have known deleterious health effects. Moreover, the ability to accomplish such detection in fluids that are easy to collect, such as saliva and urine, provides additional practical advantages. Reported herein is the application of cyclodextrin-promoted fluorescence energy transfer and fluorescence modulation to accomplish precisely such detection: the development of sensitive and selective florescence-based detection methods for phthalates in saliva, an easily collectable human biologic fluid. Such saliva-based detection methods occur with high levels of selectivity (100% differentiation) and sensitivity (limits of detection as low as 0.089?µM), and provide significant potential in the development of practical phthalate detection devices.  相似文献

A water‐soluble, cyclodextrin‐supported palladium complex (DACH‐Pd‐β‐CD) catalytic system was designed and synthesized, which can efficiently catalyze Suzuki–Miyaura cross‐coupling reactions between aryl halides and arylboronic acid in water under mild conditions. The catalyst was successfully characterized using the methods of transmission electron microscopy, energy‐dispersive X‐ray spectrometry, X‐ray diffraction, thermogravimetric analysis, and Fourier transform infrared and NMR spectroscopies. Furthermore, the catalyst can be easily separated from the reaction mixture and still maintain high catalytic activity after ten cycles. No leaching of palladium into the reaction solution occurred. The advantages of green solvent (water), short reaction times (2–6 h), low catalyst loading (0.001 mol%), excellent yields (up to 99%) and reusability of the catalyst mean it will have potential applications in green chemical synthesis.  相似文献

Gastric ulcers are one of the most common stomach diseases that often accompanied by inflammation, congestion, edema, scar tissue formation, and pyloric obstruction. Fiberoptic endoscopy and X-ray analysis of the upper GI tract have become the diagnostic procedure of choice for patients. However, conventional diagnosis technology is either invasive or radioactive. Herein, a novel CD-MOF NIR-II fluorophore (GPs-CH1055) was developed. The relative fluorophore intensity was largely consistent at various media and pH buffers, and it can swell into gel particles in solvents and be completely expelled from the gastrointestinal tract without being assimilated. GPs-CH1055 has been further evaluated in vivo, and exhibited strong retention effect on the gastric ulcer sites, bright NIR-II signals with high spatial and temporal resolution. Therefore, GPs-CH1055 shows great promise for realizing real-time gastric ulcer imaging and diagnosis.  相似文献

The main objective of this study is to provide an insight into the interactions involved during adsorption of the alcohols on β-CD composite nanostructured membrane. Interactions between β-cyclodextrin (β-CD) and alcohols (methanol, ethanol and butanol) are studied using the QM/MM method. Magnitude of interaction energies show that the alcohols are adsorbed on the membrane. In addition, the thermochemical analysis suggests that the formation of these host-guest complexes is enthalpy driven.  相似文献

The influence of β-CD concentration on the acid hydrolysis of N-methyl-N-nitroso-p-toluenesulfonamide (MNTS) has been studied in the presence and absence of different alcohol concentrations. The rate of the denitrosation reaction in bulk water decrease as the β-CD concentration increases due to MNTS complexation in the CD cavity and the reaction taking place exclusively outside the cyclodextrin. Changes in this inhibition due to the presence of β-CD allow us to obtain the binding constants of different alcohols to the cyclodextrin. These binding constants are in very good agreement with those determined in the bibliography by other methods.  相似文献
