Syntheses and Properties of Two Heterocyclically Substituted Hypericin Derivatives: 10,11-Dibenzothiazolyl-10,11-didemethylhypericin and 10,11-Dibenzoxazolyl-10,11-didemethylhypericin

Summary. The syntheses of the two heterocyclically substituted title hypericin derivatives were achieved starting either from 6-benzothiazolyl-tri-O-methyl-6-demethylemodin or 6-benzoxazolyl-tri-O-methyl-6-demethylemodin. The use of microwave assisted synthesis for the preparation of these anthraquinone synthons and the chemical as well as photochemical properties of the corresponding unique hypericin derivatives, which might constitute new photodynamic therapy agents, are reported. The tautomeric and stereochemical aspects of these hypericin derivatives were investigated by means of semiempirical calculations (AM1).     

Synthesis and mesomorphism of ionic self-assembled complexes of anthraquinones

Anthraquinones are known to exhibit mesomorphism upon appropriate substitutions. In this report, the concept of ionic self-assembly has been applied to design and synthesise new anthraquinone-based liquid crystals. Four different anthraquinones were used as polyelectrolytes which form ionic complexes with double-tail ammonium surfactants. The chemical structures were characterised by spectral techniques and elemental analysis. The thermal behaviour of the complexes was investigated by polarising optical microscopy, differential scanning calorimetry, thermogravimetric analysis and X-ray diffraction studies. All these complexes display lamellar mesomorphism. The amphotropic nature of the complexes has also been studied.     

Polyketide anthraquinone,diphenyl ether,and xanthone derivatives from the soil fungus Penicillium sp. PSU-RSPG99

Three new polyketides including one new diphenyl ether, penicillither (1), one new anthraquinone, penicilliquinone (2), and one new xanthone, penicillixanthone (3), together with twelve known compounds were isolated from the soil fungus Penicillium sp. PSU-RSPG99. Their structures were identified by spectroscopic evidence. In addition, the position of a chlorine atom in 1 was confirmed by the plausible biosynthetic pathway from the isolated chlorobenzophenone. Known GKK1032B displayed mild antimycobacterial and moderate cytotoxic (against human oral carcinoma, KB, human breast cancer, MCF-7, and noncancerous Vero cell lines) activities.     

Application of an efficient strategy based on liquid–liquid extraction,high‐speed counter‐current chromatography,and preparative HPLC for the rapid enrichment,separation, and purification of four anthraquinones from Rheum tanguticum

This study presents an efficient strategy based on liquid–liquid extraction, high‐speed counter‐current chromatography, and preparative HPLC for the rapid enrichment, separation, and purification of four anthraquinones from Rheum tanguticum. A new solvent system composed of petroleum ether/ethyl acetate/water (4:2:1, v/v/v) was developed for the liquid–liquid extraction of the crude extract from R. tanguticum. As a result, emodin, aloe‐emodin, physcion, and chrysophanol were greatly enriched in the organic layer. In addition, an efficient method was successfully established to separate and purify the above anthraquinones by high‐speed counter‐current chromatography and preparative HPLC. This study supplies a new alternative method for the rapid enrichment, separation, and purification of emodin, aloe‐emodin, physcione, and chrysophanol.     

Simultaneous determination of small amounts of carbazole and anthracene in some monosubstituted anthraquinones

Simple separation of carbazole and anthracene from monosubstituted anthraquinones is achieved through the application of the zone-melting technique used biphenyl as a medium. Determination limits for both compounds measured by synchronous fluorimetry are 2 μg g^?1 in 2-methyl-, 2-ethyl-, 1-hydroxy- and 1-chloroanthraquinones, and 10 μg g^?1 in another four derivatives.     

Syntheses,Photochemical Properties,and Tautomerism of Intramolecularly <Emphasis Type="Italic">Friedel-Crafts</Emphasis> Acylated Hypericin Derivatives

Summary. Intramolecularly Friedel-Crafts acylation carried out on the hypericin moiety provided a new class of 9,12-dicarbonyl substituted hypericin derivatives as potential candidates for photodynamic therapy (PDT). Focusing on cyclopentanone and cyclohexanone condensed derivatives, investigations concerning the chemical and photochemical properties as well as the tautomerism of these compounds were performed.     

Determination of 19 rhubarb constituents by high-performance liquid chromatography-ultraviolet-mass spectrometry

Rhubarb (Rhei rhizoma), a commonly used Chinese herb, contains anthraquinones, anthrones, galloylglucoses, stilbenes, and flavan-3-ols compounds, etc. as major constituents. Using 19 of these compounds as markers, an HPLC-UV-MS method was developed to estimate the quality of rhubarb samples within a period of 70 min. Extracts were analyzed with a Cosmosil 5C18-MS column and eluted with a gradient comprising an aqueous solution of acetic acid and methanol at a flow rate of 0.9 mL/min. Peaks were detected by absorbance measurements at 254 nm (6 and 8-19) and 280 nm (1-5 and 7), and the peaks of the marker substances were identified from their UV spectra and MS fragmentation patterns. The proposed method yielded a peak-area ratio RSD value with an intraday SD falling within 0.71-1.78% and an interday SD within 0.78-1.98% at a detection limit of 0.2-3.2 microg/mL. The ESI negative ion mode was used to collect data (molecular weight, CID fragments from MS and MS/MS spectra) for 19 compounds from four types of structure categories: anthraquinones, dianthrone glycosides, stilbenes, and galloylglucosides. The information gathered can be used to identify the structures of various peaks appearing in the LC chromatograms of rhubarb samples.

用荧光光谱法研究了大黄素，黄酮，栎精。柚皮甙，黄芩甙和龙胆苦甙等化合物与人血清白蛋白（ＨＳＡ）的结合反应，发现除龙胆苦甙外，其余化合物对ＨＳＡ的荧光有很强的猝平作用。从荧光猝灭结果求得了它们与ＨＳＡ的结合常数，表明这些化合物与ＨＳＡ的结合能力随其所含极性基团的增多而减弱。根据Ｆｏｒｓｔｅｒ非辐射能量转移机理，求出第一结合部位与ＨＳＡ分子中２１４－色氨酸残基的距离，由此提出了形成复合物的结构模型，并

Summary. Investigating the properties of similar but regioselectively differently substituted hypericin derivatives, 9,12-dibenzothiazolylhypericin was synthesized and compared with the recently prepared 10,11-analogue. A significant difference in the ability to generate singlet oxygen and/or reactive oxygen species and different absorption spectra of these two derivatives were observed.
