The 100 yers old Wurster's salts have long been recognized as compounds with redical cations. Their unusual stabilization derives partly form capto-dative (cd) substitution. This principle is now discussed as one factor of radical stabilization and it is applied to simple methine derivatives. cd-Substitution has synthetically useful applications: cd-substituents on a carbon atom allow its selective dehydrodimerization. Olefines with geminal and thus cross-conjugated cd-substituents are “radicophilic” and permit twofold carbon radical addition. cd-Substituted olefines are useful antioxidants, polymerization inhibitors and are promising agents in the control of biological radical reactions. The inhibition of the model enzyme, haloalkane dehalogenase from Sphingomonas paucimobilis, was investigated by a combination of electrophoretically mediated microanalysis with a partial filling technique, followed by indirect or direct detection. In this setup, part of the capillary is filled with a buffer suitable for the enzymatic reaction (20 mM glycine buffer, pH 8.6) whereas the rest of the capillary is filled with the background electrolyte optimal for separation of substrates and products. Two different background electrolytes and corresponding detection approaches were used to show the versatility of the developed method. The inhibition effect of 1,2-dichloroethane on the dehalogenation of brominated substrate 1-bromobutane was studied by means of 10 mM chromate - 0.1 mM cetyltrimethylammonium bromide (pH 9.2) in combination with indirect detection or 20 mM beta-alanine - hydrochloric acid (pH 3.5) in combination with direct detection. The method was used to estimate the inhibition constant K(I) (0.44 mM by indirect detection and 0.63 mM by of direct detection) and to determine the inhibition type. Compared to spectrophotometric and other discontinuous assays, the method is rapid, can be automated, and requires only small amount of reagents that is especially important in the case of enzymes and inhibitors. 相似文献

The influence of perrhenate concentration, the concentration of the reducing agent and pH of the reaction mixture on the yield of perrhenate reduction were studied to find a possibility to decrease the stannous chloride concentration in the reaction mixture without negative changes on the yield of perrhenate reduction. Britton-Robinson buffer solutions were selected as the background electrolytes because of their buffering capacity in a wide pH interval. The highest degree of perrhenate reduction was obtained at pH 2 at perrhenate concentrations ranging from 10(-4) to 10(-3) mol/L. The stability of reduced rhenium against pH change from 2 to 5.5 and against dilution of rhenium in the reaction mixture to the concentration suitable for the application in radiotherapy were studied as well. Conditions for converting a set of five standard proteins to electrochemically active sodium dodecylsulfate (SDS) complexes were worked out with the aim of using such complexes for conductivity detection with a a chip electrophoresis system. The results obtained were compared with standard capillary electrophoresis (37 cm (effective length 30 cm)×75 μm I.D. capillary, 10 kV, negative polarity at the inlet). The chip separations were run at 500 V per chip (100 V/cm) as compared to the standard capillary arrangement, which was run at 266.6 V/cm. For the capillary set-up the protein complexes were prepared in aqueous solution (Milli-Q water) made 10 mM with respect to SDS. If the SDS concentration was increased to 50 mM, the separation in the capillary was incomplete. On the other hand with the chip system both approaches yielded acceptable results. The chip separations were slightly (but not distinctly) shorter and offered better separations than the standard set-up. The concentration of the surfactant used for the preparation the complexes results in alternations of the elution sequence, which is preserved if the chip separation is used instead of the capillary set-up. Apparently the full capacity of protein–SDS binding is not exploited for the preparation of the adducts. 相似文献

Avian eggshell matrix proteins were studied by two analytical approaches. Peptide mapping was done by trypsin and pepsin followed by collagenase cleavage; analyses were carried out by capillary electrophoresis and reversed-phase high-performance liquid chromatography (HPLC). Comparison of peptide maps obtained by both methods revealed a complex mixture of peptides in the insoluble layers of the eggshell; it was concluded that there are at least three different insoluble protein/peptide layers in the avian eggshell (cuticle, palisade, and mammillary layer). Partial characterization of peptides in each layer was made by HPLC-mass spectrometry analysis. There is an evidence that the eggshell insoluble proteins contain species susceptible to collagenase cleavage, however, the sequences split by this enzyme probably are not those typical for the main triple-helical core of collagenous proteins. It is proposed that the action of collagenase upon eggshell proteins is caused by the side effect of collagenase described previously with synthetic peptides. Some of the proteins present are probably glycosylated. Fatty acid content in the insoluble eggshell layers (after decalcification) was in the range of 2-4% (which reflected both lipid and lipoproteins bound fatty acids). The magnetic moments of the 5/2 1 + state in117Te at 274.4 keV and of the 7/2 1 + state in121Te at 443.1 keV have been determined asμ exp(5/2+)=?0.75(5)n.m. andμ exp(7/2+)=+0.63(7) n.m., respectively, using the TDPAD method and the reactions115,119Sn(α,2n)117,121Te. An evaluation method is described which provides, in case of the normalized time differential patternR(t) exhibits less than half of an oscillation period, a unique value of the Larmor frequency. The comparison of the measured magnetic moments with Nilsson-, soft rotor Coriolis- as well as core-particle coupling calculations gives valuable hints on the shape dependence of magnetic moments and, consequently, on the deformation of different states in the odd transitional nuclei117–125Te. In the light of the core-particle coupling model the positive parity states of117Te and121Te are interpreted as the members ofΔJ=1 andΔJ=2 bands built on thes 1/2,d 3/2,d 5/2 and g7/2 single-particle states, respectively. 相似文献

Cyclic voltammetry has been used to study the coupling of ion transfer reactions at a liquid membrane. The liquids are either supported by a porous hydrophobic membrane (polyvinylidene difluoride, PVDF) when the organic solvent is non-volatile (o-nitrophenyloctylether) or are merely a free standing organic solvent layer such as 1,2-dichloroethane comprised between two hydrophilic dialysis membranes supporting the adjacent aqueous phases. The passage of current across the liquid membrane is associated with two ion transfer reactions across the two polarised liquid liquid interfaces in series. It is shown that it is possible to study the transfer of highly hydrophilic ions at one interface by limiting the mass transfer of the other ion transfer reaction at the other interface. Indeed, for systems comprising an ion M in one aqueous phase and a reference ion R partitioned between the membrane and the other aqueous phase, the observed and simulated cyclic voltammograms have a half-wave potential determined by the Gibbs energy of transfer of M transferring at one interface and by the limiting mass transfer of R at the other interface. This new methodology opens a way to measure the Gibbs energy of transfer of highly hydrophilic or hydrophobic ions, which usually limits the potential window at single liquid liquid interfaces (ITIES). 相似文献

Electron Transfer and Ion Pair Formation Single Crystal Structure of Bis(sodium 1,1′-biphenyl-2-thiolate-diglyme): An Intermediate in the Reductive Ring Opening of Dibenzothiophene On Na-metal reduction of dibenzothiophene, the five-membered sulfur ring opens to form a colorless 1,1′-biphenyl-2-thiolate sodium salt, which, according to its single-crystal structure determination, is a dimer containing a four-membered, twice diglyme-solvated ring (diglyme···Na⊕?SR)2. Additional measurements provide the following information: cyclic voltammetry in aprotic MeCN solution shows one quasi-reversible electron transfer at E = ?2.58 V. The dibenzothiophene radical anion can be generated in aprotic THF solution at a K mirror and characterized by an 81-line ESR spectrum and its simulation. This blue species is also the first UV/VIS detectable one before the solution changes via green (due to blue + yellow color mixing) to yellow, yielding across an isosbestic point a second and diamagnetic compound. All of the above results suggest a consecutive two-electron reduction followed by an intersystem protonation, M + (e?) → M.? (blue) + (e?) → (M??, yellow?) + (H⊕) → MH? (colorless), to yield the crystallized and structurally characterized reaction intermediate. The diglyme-solvated sodium-salt dimer provides a basis for a quantum-chemical discussion of some facets of the most likely microscopic reduction pathway. 相似文献
