The density matrix (DM) obtained from Yang's [Phys. Rev. Lett. 66, 1438 (1991)] divide-and-conquer (DC) Hartree-Fock (HF) calculation is applied to the explicit second-order M?ller-Plesset perturbation (MP2) energy functional of the HF DM, which was firstly mentioned by Ayala and Scuseria [J. Chem. Phys. 110, 3660 (1999)] and was improved by Surján [Chem. Phys. Lett. 406, 318 (2005)] as DM-Laplace MP2. This procedure, termed DC-DM MP2, requires the HF DM of holes, for which we propose two evaluation schemes in DC manner. Numerical studies reveal that the DC-DM MP2 energy deviation from canonical MP2 is the same order of magnitude as DC-HF energy deviation from conventional HF whichever type of hole DM is adopted. It is also confirmed that the central processing unit time of DC-DM MP2 is less than that of DM-Laplace MP2 because the DC-HF DM is sparser than conventional DM.  相似文献

Simultaneous HPLC assay of 1-adamantanamine hydrochloride (amantadine) and its four related compounds [2-adamantanamine hydrochloride (2-ADA), 1-adamantanmethylamine (ADAMA), 1-(1-adamantyl)ethylamine hydrochloride (rimantadine) and 3,5-dimethyl-1-adamantanamine hydrochloride (memantine)] in phosphate-buffered saline (pH 7.4) after pre-column derivatization with 4-fluoro-7-nitro-2,1,3-benzoxadiazole (NBD-F) was developed. Phosphate-buffered saline samples were mixed with borate buffer and NBD-F solution in acetonitrile at 60 degrees C for 5 min and injected into HPLC. Five derivatives were well separated from each other. The lower limits of detection of amantadine, 2-ADA, ADAMA, rimantadine and memantine were 0.008, 0.001, 0.0008, 0.0015 and 0.01 microg/mL, respectively. The coefficients of variation for intra- and inter-day assay were less than 6.4 and 8.2%, respectively. The method presented was applied to a binding study of these compounds to human alpha(1)-acid glycoprotein. While affinity constants and capacities for ADAMA, rimantadine and memantine were calculated by means of Scatchard plots, those for the others were not determined. ADAMA, rimantadine and memantine were bound with different affinities and capacities. These results indicate that NBD-F is a good candidate as a fluorescent reagent to simultaneously determine amantadine and its four related compounds by HPLC after pre-column derivatization. Our method can be applied to binding studies for protein.  相似文献

Monolithic silica columns in semi-micro-format have been synthesized using poly(acrylic acid) as a phase-separation inducer via a sol–gel route. The absence of a thick skin layer accompanied by deformation of the micrometer-sized gelling skeletons on the outermost part of the macroporous silica rod contributed to improve the efficiency of monolithic silica columns as thick as 2.4 mm in diameter. The kinetic plot analysis revealed that monolithic silica columns with macropore diameter of 1 μm and skeleton thickness of 1 μm with decreased macroporosity behave similarly to columns packed with 3 μm particles with slightly lower back pressure.  相似文献

We consider an optimization problem with positively homogeneous functions in its objective and constraint functions. Examples of such positively homogeneous functions include the absolute value function and the p-norm function, where p is a positive real number. The problem, which is not necessarily convex, extends the absolute value optimization proposed in Mangasarian (Comput Optim Appl 36:43–53, 2007). In this work, we propose a dual formulation that, differently from the Lagrangian dual approach, has a closed-form and some interesting properties. In particular, we discuss the relation between the Lagrangian duality and the one proposed here, and give some sufficient conditions under which these dual problems coincide. Finally, we show that some well-known problems, e.g., sum of norms optimization and the group Lasso-type optimization problems, can be reformulated as positively homogeneous optimization problems.  相似文献

The N‐acylsulfonamide group, known as a safety‐catch linker, has been applied to photoaffinity labeling (PAL) using a cinnamate‐type photocrosslinker to improve the efficiency of PAL‐based target identification. A bioorthogonal sulfo‐click reaction was used to stably link a photocrosslinker unit with N‐acylsulfonamide linkage to produce a photoactivatable probe without any protection. In addition, the crosslinked protein was selectively isolated with a small cinnamate tag via linkage disruption upon N‐alkylation. Furthermore, the tag moiety was photochemically converted to a stable coumarin derivative by losing a water molecule, which is a useful property in MS‐based identification.  相似文献

A method of preparing titania-coated monolithic silica stationary phase has been developed to achieve liquid chromatographic separation of phosphorus-containing compounds, which have recently been attracting increasing attention in biochemical research. The titania-coated silica columns exhibited efficient separation with low pressure drop, which is a typical feature of monolithic structures, and also possessed phospho-selectivity, which is a unique property of the titania surface. The material characteristics of titania-coated monolithic silica were examined, and then resin-clad columns were applied to the HPLC analysis of phosphorylated compounds. Highly efficient separation of phosphorylated substances indicated that the novel titania-coated monolithic silica column will find applications as a useful tool in the field of biochemistry, especially in post-genomic analyses.  相似文献

New molecules incorporating a uracil nucleic base and a dihydroindolizine (DHI) unit linked via spacer arms, i.e., uracil-spyrodihydroindolizine (4), were synthesised as models for light sensitive systems for nucleic acid labelling. The uracil-DHI (4) undergoes easy photocoloration to the uracil-betaine (5) generating the UV-detectable species. Preliminary results show rather weak binding of uracil-DHI (4) to calf thymus-DNA.  相似文献

Complete resolution of hydroxyeicosatetraenoic acid (HETE) enantiomers was achieved using hydroxypropyl‐γ‐cyclodextrin (HP‐γ‐CD)‐modified MEKC. The optimum running conditions were determined to be utilizing a 30 mM phosphate–15 mM borate buffer (pH 9.0) containing 30 mM HP‐γ‐CD and 75 mM SDS as the BGE, application of +30 kV as the effective voltage, and carrying out the experiment at 15°C. The eluents were detected at 235 nm. The method was used successfully for the simultaneous separations of (S)‐ and (R)‐enantiomers of regioisomeric 8‐, 11‐, 12‐, and 15‐HETEs. Subsequently, the optimized method was applied to evaluate the stereochemistry of 8‐ and 12‐HETEs from the marine red algae, Gracilaria vermiculophylla and Gracilaria arcuata, respectively. The 8‐HETE was found to be a mixture of 98% (R)‐enantiomer and 2% (S)‐enantiomer, while the 12‐HETE was a mixture of 98% (S)‐enantiomer and 2% (R)‐enantiomer. The present study demonstrates that the HP‐γ‐CD‐modified MEKC method is simple and sensitive and provides unambiguous information on the configuration of natural and synthetic HETEs.  相似文献
