Enzymatic oxidative decarboxylation is an up-and-coming reaction yet lacking efficient screening methods for the directed evolution of decarboxylases. Here, we describe a simple photoclick assay for the detection of decarboxylation products and its application in a proof-of-principle directed evolution study on the decarboxylase OleT. The assay was compatible with two frequently used OleT operation modes (directly using hydrogen peroxide as the enzyme's co-substrate or using a reductase partner) and the screening of saturation mutagenesis libraries identified two enzyme variants shifting the enzyme's substrate preference from long chain fatty acids toward styrene derivatives. Overall, this photoclick assay holds promise to speed-up the directed evolution of OleT and other decarboxylases. 相似文献

Large, non‐symmetrical, inherently chiral bispyridyl ligand L derived from natural ursodeoxycholic bile acid was used for square–planar coordination of tetravalent PdII, yielding the cationic single enantiomer of superchiral coordination complex 1 Pd3 L 6 containing 60 well‐defined chiral centers in its flower‐like structure. Complex 1 can readily be transformed by addition of chloride into a smaller enantiomerically pure cyclic trimer 2 Pd3 L 3Cl6 containing 30 chiral centers. This transformation is reversible and can be restored by the addition of silver cations. Furthermore, a mixture of two constitutional isomers of trimer, 2 and 2′ , and dimer, 3 and 3′ , can be obtained directly from L by its coordination to trans‐ or cis‐N‐pyridyl‐coordinating PdII. These intriguing, water‐resistant, stable supramolecular assemblies have been thoroughly described by 1H DOSY NMR, mass spectrometry, circular dichroism, molecular modelling, and drift tube ion‐mobility mass spectrometry. 相似文献

Aprotic ionic polymers containing trimethylsilylmethyl-substituted imidazolium structures are synthesized using free radical polymerization of monomers comprising a vinyl group either at the cation or at the anion. Bulk polymerization is used for the room temperature ionic liquid monomer 1-trimethylsilylmethyl-3-vinylimidazolium bis(trifluoromethylsulfonyl)imide. In contrast to this, solution polymerization is applied for 1-trimethylsilylmethyl-3-methylimidazolium p-styrene sulfonate because this monomer undergoes self-polymerization during melting at a higher temperature than selected for bulk polymerization. Glass transition temperature (T g) of the ionic polymers and intrinsic viscosity measurements indicate differences between these polymers, which are composed either of a polycation with a trimethylsilylmethyl substituent at each vinylimidazolium segment of the polymer chain and mobile bis(trifluoromethylsulfonyl)imide (NTf2−) anions or a polyanion containing p-styrene sulfonate segments and mobile 1-trimethylsilylmethyl-3-methylimidazolium cations. The new aprotic ionic polymers containing trimethylsilylmethyl substituents may be interesting for application in adhesive, interlayer and membrane manufacturing. 相似文献

The anticancer drug tamoxifen (TAM) displays two chiral vinyl propeller structures, which interconvert so rapidly that the process is undetectable on the NMR time scale. In the present work, the enantiomerization processes were investigated with molecular modeling techniques. The threshold mechanisms probed at the different rings were shown to be identical, i.e., involving a synchronous three-ring flip, with a correlated rotation of the rings. In order to reveal the pharmacological profiles of the two chiral forms, we performed structural studies on the ligand binding domain of estrogen receptor α (ERα LBD) and associated ligands. The enantiomers, with opposite torsional twist, were found to be discriminated by ERα. For TAM and its main metabolites, the effects of the stereoselectivity of ERα are overcome by the low energy cost for helical inversion between the two torsional enantiomers, estimated to be ~3 kcal/mol. 相似文献

A series of pyrazinoporphyrazine macrocycles carrying externally appended 2-thienyl rings, represented as [Th(8)TPyzPzM], where Th(8)TPyzPz = tetrakis-2,3-[5,6-di(2-thienyl)pyrazino]porphyrazinato anion and M = Mg(II)(H(2)O), Zn(II), Co(II), Cu(II), or 2H(1), were prepared and isolated as solid air-stable hydrated species. All of the compounds, completely insoluble in water, were characterized by their UV-visible spectra and electrochemical behavior in solutions of dimethylformamide (DMF), dimethyl sulfoxide, and pyridine. Molecular aggregation occurs at concentrations of ca. 10(-4) M, but monomers are formed in more dilute solutions of 10(-5) M or less. The examined octathienyl compounds [Th(8)TPyzPzM] behave as electron-deficient macrocycles, and UV-visible spectral measurements provide useful information about how the peripheral thienyl rings influence the electronic distribution over the entire macrocyclic framework. Cyclic voltammetric and spectroelectrochemical data confirm the easier reducibility of the compounds as compared to the related phthalocyanine analogues, and the overall redox behavior and thermodynamic potentials for the four stepwise one-electron reductions of the compounds are similar to those of the earlier examined octapyridinated analogues [Py(8)TPyzPzM]. Quantum yields (Φ(Δ)) for the generation of singlet oxygen, (1)O(2), the cytotoxic agent active in photodynamic therapy (PDT), and fluorescence quantum yields (Φ(F)) were measured for the Zn(II) and Mg(II) complexes, [Th(8)TPyzPzZn] and [Th(8)TPyzPzMg(H(2)O)], and the data were compared to those of corresponding octapyridino macrocycles [Py(8)TPyzPzZn] and [Py(8)TPyzPzMg(H(2)O)] and their related octacations [(2-Mepy)(8)TPyzPzZn](8+) and [(2-Mepy)(8)TPyzPzMg(H(2)O)](8+). These measurements were carried out in DMF and in DMF preacidified with HCl (ca. 10(-4) M). All of the examined Zn(II) compounds behave as excellent photosensitizers (Φ(Δ) = 0.4-0.6) both in DMF and DMF/HCl solutions, whereas noticeable fluorescence activity (Φ(F) = 0.36-0.43) in DMF/HCl solutions is shown by the Mg(II) derivatives; these data might provide perspectives for applications in PDT (Zn(II)) and imaging response and diagnosis (Mg(II)). 相似文献

We exploited the ability of photocatalytic titanium dioxide (TiO(2)) as an agent for the biofilm control. Two photocatalytic systems were investigated: a 3 g L(-1) suspension of TiO(2) nanopowder in demineralized water and glass slides coated with a TiO(2) thin film, achieved by sol-gel deposition. A running protocol for the photoactivation of TiO(2) was set up using the dye rhodamine B. The microorganisms studied were Pseudomonas stutzeri, Pseudomonas aeruginosa and a Bacillus cereus-group as planktonic cells. P. aeruginosa biofilms were also studied at both the solid-liquid and the solid-air interface. The TiO(2) nanopowder produced 1-log reduction of Bacillus sp. planktonic cells in 24 h, 2-log reduction of P. stutzeri planktonic cells in 30 min and 1-log reduction of P. aeruginosa planktonic cells in 2 h compared with non-photo-activated TiO(2). TiO(2) thin film produced almost a complete eradication of P. aeruginosa planktonic cells (initial concentration 10(8) cells mL(-1)) in 24 h compared to a 3-log reduction caused by UV-A light alone. In contrast, neither the photocatalytic treatment with TiO(2) film nor that with TiO(2) nanopowder had any effect on P. aeruginosa biofilms at all the interfaces investigated. Possible explanations for these findings, and for the discrepancy between this work and literature data, are discussed. 相似文献

This paper presents an in situ spectroelectrochemical characterization of polyazulene (PAz) and PAz-C(60) composite films using Fourier Transform Infrared Attenuated Total Reflection (FTIR-ATR) spectroscopy. In situ FTIR-ATR spectra were recorded simultaneously as the films were charged and discharged electrochemically. The aim was to clarify how the use of ILs and the addition of C(60) affected the electronic transport and structural changes occurring in PAz during electrochemical charging. We found that electrosynthesis of PAz in an IL lowered the oxidation potential of the film and improved its electroactivity. The FTIR-ATR data also suggest that PAz with a longer effective conjugation length is obtained during electrosynthesis when using ILs. With in situ FTIR-ATR it is possible to quite accurately determine the onset potential for oxidation/reduction. These values are important since they determine the suitability of the polymer for a specific application. Our experiments indicate that two types of charge carriers are formed during electrochemical oxidation of PAz in an IL. Furthermore, their formation is strongly affected by the addition of C(60) into the film. The type of charge carrier formed affects the electronic and possibly also ionic transport within the film. The inclusion of C(60) into PAz influenced the optical and structural properties considerably. In situ FTIR-ATR is also an extremely useful method for studying the potential stability of an IL during electrochemical cycling. We showed that cathodic decomposition of N,N-butyl-methyl-pyrrolidinium bis(trifluoromethylsulfonyl)imide ([BMP][Tf(2)N]) occurs at less negative potentials than those determined electrochemically. 相似文献

Bacterial contamination of implanted devices is a common cause of their failure. The aim of the present study was to assess the capability of electrochemical procedures to: (a) promote the formation of anatase on the surface of commercially pure Grade 2 Ti and Ti Grade 5 (Ti6Al4V) alloy; (b) inhibit in vitro biofilm formation of Staphylococcus aureus, Staphylococcus epidermidis, Streptococcus mutans and Porphyromonas gingivalis and oral plaque in vivo, (c) preserve favorable response of osteoblasts and fibroblasts to materials surfaces. Ti Grade 2 and Ti Grade 5 were respectively anodized at two different voltages: 90 and 130V for pure titanium; 100 and 120V for Ti6Al4V alloy. Surface characterization was performed by scanning electron microscopy (SEM) equipped with EDS probe, laser profilometry and X-ray diffractometry. Bacterial adhesion characterization was performed either in vitro and in vivo in patients. Osteoblast and fibroblast response was evaluated by metabolic activity assessment. The higher voltage applied in the anodization treatment of pure titanium (130V) and Ti6Al4V alloy (120V) surfaces, compared to the untreated pure titanium and Ti6Al4V and to lower voltage treatments, resulted in a greater decrease in bacterial attachment and biofilm formation in both in vitro and in vivo experiments. In contrast, the high voltage treatments were found to promote osteoblasts and fibroblasts proliferation. The observations indicated that the experimented high voltage anodization treatments may contribute to preserve the tissue integration and reduce bacteria colonization of titanium and titanium alloy for implantable applications. 相似文献
