This study presents a novel method for rapid prototyping of polymer microsystems. The method is based on excimer laser ablation of a thermally and mechanically stable polymer, such as PEEK (poly-ether-ether-ketone). A negative of the desired microsystem is laser machined in PEEK, which can then be used directly for hot embossing or injection moulding of a series of prototypes. This approach is very rapid and considerably cheaper than more traditional approaches to toolmaking, while still performing well in terms of reproduction of tool dimensions. The reduction in time and cost for a master tool using this method opens up new possibilities for testing small series in the R&D phase of a microsystem. Finally, two particular applications of the technique are presented.  相似文献

A series of six new dyads consisting of a zinc or magnesium porphyrin appended to a platinum terpyridine acetylide complex via a para-phenylene bisacetylene spacer are described. Different substituents on the 4' position of the terpyridinyl ligand were explored (OC7H15, PO3Et2, and H). The ground-state electronic properties of the dyads are studied by electronic absorption spectroscopy and electrochemistry, and they indicate some electronic interactions between the porphyrin subunit and the platinum complex. The photophysical properties of these dyads were investigated by steady-state, time-resolved, and femtosecond transient absorption spectroscopy in N,N-dimethylformamide solution. Excitation of the porphyrin unit leads to a very rapid electron transfer (2-20 ps) to the nearby platinum complex followed by an ultrafast charge recombination, thus preventing any observation of the charge separated state. The variation in the rate of the photoinduced electron transfer in the series of dyads is consistent with Marcus theory. The results underscore the potential of the para-phenylene bisacetylene bridge to mediate a rapid electron transfer over a long donor-acceptor distance.  相似文献

During the last few years there has been a rapid increase in the use of electrochemical reactions in lab-on-a-chip devices. This development, which has so far mainly focussed on electrochemical detection in chip-based capillary electrophoresis, can be explained by the fact that electrochemical techniques and devices are particularly well-suited for inclusion in lab-on-a-chip systems. The most important reason for this is that the required electrodes can readily be manufactured and miniaturised without loss of analytical performance using conventional microfabrication methods. In this Research Highlight article, the developments during the last three years concerning electrochemical techniques for lab on-a-chip applications are discussed, with particular focus on emerging electrochemical methods for sample clean-up and preconcentration, electrochemical derivatisation and electrochemical detection in chip-based capillary electrophoresis.  相似文献

DNA-protein cross-linking is one of the many DNA lesions mediated by hydroxyl radicals, the most damaging among the reactive oxygen species in biological systems. Density functional theory methods are employed to investigate the complex reaction mechanisms of the formation of cytosine-tyrosine cross-links as observed in gamma-irradiated aqueous solutions of cytosine and tyrosine, as well as in gamma-irradiated nucleohistone. The majority of the radical addition mechanisms considered are found to have significant barriers and therefore to be thermodynamically unfavorable for the formation of the initial cross-linked product. Our calculated reaction potential energy surfaces suggest that a feasible complete mechanism consists of radical combination forming the initial cross-linked product, a hydrogen shuffle within the initial cross-linked product, and an acid-catalyzed dehydration reaction. Water and hydrogen-bonding interactions are suggested to play a key role in catalyzing the hydrogen-transfer step of the reaction.  相似文献

The preparation of donor (D)-photosensitizer (S) arrays, consisting of a manganese complex as D and a ruthenium tris(bipyridyl) complex as S has been pursued. Two new ruthenium complexes containing coordinating sites for one (2a) and two manganese ions (3a) were prepared in order to provide models for the donor side of photosystem II in green plants. The manganese coordinating site consists of bridging and terminal phenolate as well as terminal pyridyl ligands. The corresponding ruthenium-manganese complexes, a manganese monomer 2b and dimer 3b, were obtained. For the dimer 3b, our data suggest that intramolecular electron transfer from manganese to photogenerated ruthenium(III) is fast, k(ET) > 5 x 10(7) s(-)(1).  相似文献

Analysing the general dispersion relation numerically, the unstable regions of the relativistic electron-beam plasma system were determined. The external parameters were varied to ensure more effective instability excitations. The full charge and current compensation presumptions lead to new synchronism predictions. Spin exchange between different chemical environments is an important observable for characterizing chemical exchange kinetics in various contexts, including protein folding, chelation chemistry, and host–guest interactions. Such spins experience effective spin–spin relaxation rate, R2,eff, that typically shows a dispersive behavior which requires detailed analysis. Here, we describe a class of highly simplified R2,eff behavior by relying on hyperpolarized 129Xe as a freely exchanging ligand reporter. It provides large chemical shift separations that yield reduced expressions of both the Swift–Connick and the Carver–Richards treatment of exchange-induced relaxation. Despite observing a diamagnetic system, R2,eff is dominated by large Larmor frequency jumps and thus allows detection of otherwise inaccessible analyte concentrations with a single spin echo train (only 0.01% of the overall hyperpolarized spins need to be transiently bound to the molecule). The two Xe hosts cryptophane-A monoacid (CrA-ma) and cucurbit[6]uril (CB6) represent two exemplary families of container molecules (the latter one also serving as drug delivery vehicles) that act as highly efficient phase shifters for which we observed unprecedented exchange-induced relaxivity r2 (up to 866 s−1 mM−1). By including methods of spatial encoding, multiple data points can be collected simultaneously to isolate the exchange contribution and determine the effective exchange rate in partially occupied binding sites with a single delivery of hyperpolarized nuclei. The relaxivity is directly related to the guest turnover in these systems and temperature-dependent measurements yield an activation energy of EA = 41 kJ mol−1 for Xe@CrA-ma from simple relaxometry analysis. The concept is transferable to many applications where Xe is known to exhibit large chemical shifts.

Localized detection of hyperpolarized, exchanging Xe spins enables quantitative insights at unprecedented sensitivity for characterizing chemical exchange kinetics in various contexts such as host–guest interactions and displacement assays.  相似文献

The extracellular β-xylosidase (EC 3.2.1.37) excreted by the thermophilic eubacteriumRhodothermus marinus when grown on xylan has been investigated. The enzyme has been partially purified by ultrafiltration and gel filtration, and some of its characteristics are presented.Rhodothermus marinus grew on xylan with μmax= 0.4 h? and the α-xylosidase activity was 50 nkat/mL after 24 h in a batch fermentation. The α-xylosidase activity had a half-life of more than 1 h at 90°C and of 14 h at 85 °C. At 80°C, 80% of the initial activity remained after 24 h. The initial activity increased with increasing temperature, showing maximal activity at 90°C. The β-xylosidase had a pH-optimum of 6 and was stable in the range between pH 5 and 9. At pH 10 and 11, 82 and 66%, respectively, of the initial activity remained after 24 h when incubated at 65°C. The molecular weight was estimated to be 169,000 dalton by gelfiltration.  相似文献

The polyamino ligand 1,4,7-tris(2-aminoethyl)-1,4,7-triazacyclononane (1) has been used to synthesise two new ligands by Schiff-base condensation with methyl sodium acetyl phosphonate to give ligand L and methyl sodium 4-methoxybenzoyl phosphonate to give ligand L1 in the presence of lanthanide ion as templating agent to form the complexes [Ln(L)] and [Ln(L1)](Ln = Y, La, Gd, Yb). Both ligands L and L1 have nine donor atoms comprising three amine and three imine N-donors and three phosphonate O-donors and form Ln(III) complexes in which the three pendant arms of the ligands wrap around the nine-coordinate Ln(III) centres. Complexes with Y(III), La(III), Gd(III) and Yb(III) have been synthesised and the complexes [Y(L)], [Gd(L)] and [Gd(L1)] have been structurally characterised. In all the complexes the coordination polyhedron about the lanthanide centre is slightly distorted tricapped trigonal prismatic with the two triangular faces of the prism formed by the macrocyclic N-donors and the phosphonate O-donors. Interestingly, given the three chiral phosphorus centres present in [Ln(L)] and [Ln(L1)] complexes, the three crystal structures reported show the presence of only one diastereomer of the four possible. 1H, 13C and 31P NMR spectroscopic studies on diamagnetic [Y(L)] and [La(L)] and on paramagnetic [Yb(L)] complexes indicate the presence in solution of all the four different diastereomers in varying proportions. The stability of complexes [Y(L)] and [Y(L1)] in D2O in both neutral and acidic media, and the relaxivity of the Gd(III) complexes, have also been investigated.  相似文献
