Sulfite is often added to beverages as an antioxidant and antimicrobial agent. In fermented beverages, sulfite is also naturally produced by yeast cells. However, sulfite causes adverse health effects in asthmatic patients and accurate measurement of the sulfite concentration is therefore very important. Current sulfite analysis methods are time- and reagent-consuming and often require costly equipment. Here, we present a system allowing sensitive, ultralow-volume sulfite measurements based on a reusable glass-silicon microdroplet platform on which microdroplet generation, addition of enzymes through chemical-induced emulsion destabilization and pillar-induced droplet merging, emulsion restabilization, droplet incubation, and fluorescence measurements are integrated. In a first step, we developed and verified a fluorescence-based enzymatic assay for sulfite by measuring its analytical performance (LOD, LOQ, the dynamic working range, and the influence of salts, colorant, and sugars) and comparing fluorescent microplate readouts of fermentation samples with standard colorimetric measurements using the 5,5′-dithiobis-(2-nitrobenzoic acid) assay of the standard Gallery Plus Beermaster analysis platform. Next, samples were analyzed on the microdroplet platform, which also showed good correlation with the standard colorimetric analysis. Although the presented platform does not allow stable reinjection of droplets due to the presence of a tight array of micropillars at the fluidics entrances to prevent channel clogging by dust, removing the pillars, and integrating miniaturized pumps and optics in a future design would allow to use this platform for high-throughput, automated, and portable screening of microbes, plant, or mammalian cells.

The 3M Petrifilm Staph Express Count plate method was compared with AOAC Official Method 975.55 for the enumeration of Staphylococcus aureus in selected foods. Four foods--cooked, diced chicken; cured ham; smoked salmon; and pepperoni--were analyzed for S. aureus by 12 collaborating laboratories. For each food tested, the collaborators received 8 blind test samples consisting of a control sample, a low inoculation level, a medium inoculation level, and a medium inoculation level with background flora, each in duplicate. The mean log10 counts for the methods were comparable for all 4 foods. The repeatability and reproducibility variances of the 24 h Petrifilm Staph Express Count plate method were similar to those of the 72 h standard method.  相似文献

Trimesityliridium(III) (mesityl = 2,4,6-trimethylphenyl) reacts with O(2) to form oxotrimesityliridium(V), (mes)(3)Ir=O, in a reaction that is cleanly second order in iridium. In contrast to initial reports by Wilkinson, there is no evidence for substantial accumulation of an intermediate in this reaction. The oxo complex (mes)(3)Ir=O oxidizes triphenylphosphine to triphenylphosphine oxide in a second-order reaction with DeltaH++ = 10.04 +/- 0.16 kcal/mol and DeltaS++ = -21.6 +/- 0.5 cal/(mol.K) in 1,2-dichloroethane. Triphenylarsine is also oxidized, though over an order of magnitude more slowly. Ir(mes)(3) binds PPh(3) reversibly (K(assoc) = 84 +/- 3 M(-1) in toluene at 20 degrees C) to form an unsymmetrical, sawhorse-shaped four-coordinate complex, whose temperature-dependent NMR spectra reveal a variety of dynamic processes. Oxygen atom transfer from (mes)(3)Ir=O and dioxygen activation by (mes)(3)Ir can be combined to allow catalytic aerobic oxidations of triphenylphosphine at room temperature and atmospheric pressure with overall activity (approximately 60 turnovers/h) comparable to the fastest reported catalysts. A kinetic model that uses the rates measured for dioxygen activation, atom transfer, and phosphine binding describes the observed catalytic behavior well. Oxotrimesityliridium does not react with sulfides, sulfoxides, alcohols, or alkenes, apparently for kinetic reasons.  相似文献

The magnetic susceptibilities of cobaltocene and chromocene have been measured between 83 and 293 K; for cobaltocene the results suggest an appreciable orthorhombic splitting of the 2∏(σ2πδ4 ground state, whilst for chromocene the data provide further support for the 3Δ(σδ3) ground state previously deduced.  相似文献

A systematic study was conducted on the Cr catalysed tri- and tetramerisation of ethylene using bis(diphenylphosphino)amine ligands with N-aryl functionality. This study revealed that the oligomerisation reaction product selectivity is primarily dependent on the structure and size of the N-aryl groups.

Addition of sufficient steric bulk to the N-phenyl group via ortho-alkyl substitution increased the combined 1-hexene and 1-octene selectivity (overall alpha selectivity) to above 82% at an overall 1-octene selectivity of 56%. The introduction of a single carbon spacer between the N-atom and the aryl-moiety, as well as the addition of branching on this carbon, resulted in further selectivity improvements, achieving an overall 1-octene selectivity of 64% and an overall alpha selectivity of 84%. This was obtained at catalyst productivities in excess of 1,000,000 g/g Cr/h.  相似文献

The VIDAS Immuno-concentration Salmonella (ICS) plus selective plate method (Hektoen enteric, bismuth sulfite, Salmonella identification) method for the detection of Salmonella was compared to the Bacteriological Analytical Manual (BAM)/AOAC culture method in a collaborative study. Thirty-two laboratories participated in the evaluation. Each laboratory tested one or more of the 6 test products: milk chocolate, nonfat dry milk, dried whole egg, soy flour, ground black pepper, and ground raw turkey. The 2 methods were in agreement for 1,283 of the 1,440 test samples. Of the 157 test samples not in agreement, 82 were VIDAS ICS plus selective plate-positive and BAM/AOAC-negative, and 75 were VIDAS ICS plus selective plate-negative and BAM/AOAC-positive.  相似文献

High-resolution NMR spectroscopy has been used to establish the conformational consequences of the introduction of a single 3[prime or minute]-S-phosphorothiolate link in the DNA strand of a DNA : RNA hybrid. These systems are of interest as potential antisense therapeutic agents. Previous studies on similarly modified dinucleotides have shown that the conformation of the sugar to which the sulfur is attached shifts to the north (C(3[prime or minute])-endo/C(2[prime or minute])-exo). Comparisons made between NOESY cross-peak intensities, and coupling constants from PE-COSY spectra, for both non-modified and modified duplexes confirm that this conformational shift is also present in the double helical oligonucleotide system. In addition it is noted that in both the dinucleotides and the modified duplex, the conformation of the sugar ring 3[prime or minute] to the site of modification is also shifted to the north. That this pattern is observed in the small monomeric system as well as the larger double helix is suggestive of some pre-ordering of the sequences. The conclusion is supported by consideration of the (1)H chemical shifts of the heterocyclic bases near the site of the modification. The enhanced stability that these conformational changes should bring was confirmed by UV thermal melting studies. Subsequently a series of singly and doubly 3[prime or minute]-S-phosphorothiolate-modified duplexes were investigated by UV. The results are indicative of an additive effect of the modification with thermodynamic benefit being derived from alternate spacing of two modified linkers.  相似文献

The rate constant for the reaction between the sulphate radical (SO4√−) and the ruthenium (II) tris-bipyridyl dication (Ru(bipy)32+) is (3.3±0.2)×109 mol−1 dm3 s−1 in 1 mol dm−3 H2SO4 and (4.9±0.5)×109 mol−1 dm3 s−1 in 0.1 mol dm−3, pH 4.7 acetate buffer. The SO4√−radical produced by the electron transfer quenching of Ru(bipy)32+* by S2O82− reacts rapidly with both acetate buffer and chloride ions. These side reactions result in a reduction in the overall quantum yield of Ru(bipy)33+ production and reduced reaction selectivity when Ru(bipy)32+* is quenched by persulphate.  相似文献

The preferred site of alkylation of diazine N-oxides by representative hard and soft alkylating agents was established conclusively using the 1H–15N HMBC NMR technique in combination with other NMR spectroscopic methods. Alkylation of pyrazine N-oxides (1 and 2) occurs preferentially on nitrogen regardless of the alkylating agent employed, while O-methylation of pyrimidine N-oxide (3) is favoured in its reaction with MeOTf. As these outcomes cannot be explained in the context of the hard/soft acid/base (HSAB) principle, we have instead turned to Marcus theory to rationalise these results. Marcus intrinsic barriers (ΔG0) and ΔrG° values were calculated at the DLPNO-CCSD(T)/def2-TZVPPD/SMD//M06-2X-D3/6-311+G(d,p)/SMD level of theory for methylation reactions of 1 and 3 by MeI and MeOTf, and used to derive Gibbs energies of activation (ΔG) for the processes of N- and O-methylation, respectively. These values, as well as those derived directly from the DFT calculations, closely reproduce the observed experimental N- vs. O-alkylation selectivities for methylation reactions of 1 and 3, indicating that Marcus theory can be used in a semi-quantitative manner to understand how the activation barriers for these reactions are constructed. It was found that N-alkylation of 1 is favoured due to the dominant contribution of ΔrG° to the activation barrier in this case, while O-alkylation of 3 is favoured due to the dominant contribution of the intrinsic barrier (ΔG0) for this process. These results are of profound significance in understanding the outcomes of reactions of ambident reactants in general.

Marcus theory enables rationalisation and quantification of selectivities in reactions of ambident nucleophiles for which the HSAB principle cannot operate.  相似文献
