采用密度泛函理论(DFT)方法研究了系列含绿色荧光蛋白发色团双自由基分子光学异构体的几何结构、极化率(αs)和第一超极化率(βtot).结果表明,引入电子给受体取代基使分子的极化率增大,而对第一超极化率有不同影响.对于光照前的反式结构,引入电子受体βtot值增加,且βtot值随取代基吸电子能力的增强而增大;引入电子给体βtot值降低,且βtot值随取代基给电子能力的增强而减小.当分子变成相应的顺式结构时,其βtot值变化趋势与反式结构的结果正好相反.光异构化前后分子的βtot值变化不同,引入电子受体使顺式结构的βtot值比反式结构的小,其中―NO2使顺式结构的βtot值减小为反式结构的1/6;引入电子给体使反式结构的βtot值比顺式结构的小,其中―NH2使反式结构的βtot值减小为顺式结构的1/6.从而,光异构化起到调节非线性光学(NLO)响应的作用. 相似文献

建立了定量肽段串联体蛋白质(concatamers of Q peptides, QconCATs)结合18O同位素标记-多反应监测质谱的蛋白质绝对定量新方法。首先对QconCAT重组蛋白质进行了纯度表征,十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)表征结果表明重组蛋白质的纯度在99%以上,相对分子质量约为63.4 kDa。对QconCAT重组蛋白质酶切后的肽段混合物进行质谱分析,并经pFind和pLabel软件处理,验证了目标肽段。还考察了QconCAT重组蛋白质的酶切效率和18O标记效率,并对QconCAT蛋白质结合18O标记-同位素稀释-多反应监测质谱方法进行了评价。实验结果表明,采用该方法对腾冲嗜热厌氧菌(Thermoanaerobacter tengcongensis, TTE)中选定蛋白质的肽段进行绝对含量测定时,相对标准偏差小于20%,准确度较高,说明该方法可用于复杂生物样本中蛋白质的绝对定量。更重要的是所建方法不仅解决了细胞培养氨基酸稳定同位素标记(SILAC)技术的重标试剂价格昂贵的问题,也为定量蛋白质组学提供了一种新的方法。 相似文献

提出了气相色谱法测定木材中五氯酚残留量的方法。样品用乙醇经加速溶剂萃取仪在100℃静态萃取10 min。采用DB-5毛细管柱分离,电子捕获检测器测定五氯酚含量。五氯酚的质量浓度在0.01~50 mg·L-1范围内与峰面积呈线性关系,方法的检出限(3S/N)为0.01 mg·kg-1。以木材样品为基体,进行加标回收试验,回收率在94.2%~98.2%之间,测定值的相对标准偏差(n=6)小于4%。 相似文献

Environmental pollution caused by toxic metals (heavy metals, radioactive metals, etc.) is one of the major global issues, thus removal of toxic metals from contaminated water seems to be particularly important. On the other hand, the recovery and enrichment of metals, especially noble metals, from waste water is also crucial. To address these issues, nanotechnology plays an essential role in environmental monitoring and pollution control. To remove metals from contaminated water, or enrich metals from waste water, carbon nanotubes (CNTs) and their composites have attracted great attention due to their excellent adsorption performance. The removal efficiency for metal ions by CNTs was observed aroud 10–80 %, which could be improved to approach 100 % by selectively functionalizing CNTs with organic ligands. Herein, we review the applications of CNTs in treatment of toxic metal-containing wastewater for environmental monitoring and metals recovery. Due to their higher sensitivity and selectivity towards the enrichment of metals or detection of toxic metal pollution of the environment, and the latest research progress of using CNT composites for metal treatment is also discussed. 相似文献

Tigecycline, a novel intravenously administered glycylcycline antibiotic, currently plays a key role in the management of complicated multiorganism infections. However, current liquid chromatography with tandem mass spectrometry methods briefly describe parameters and the only reported internal standard was sometimes difficult to obtain. In our study, an updated liquid chromatography with tandem mass spectrometry method for the quantitative analysis of tigecycline in human serum was developed. Sample preparation involved precipitation with 20% trichloroacetic acid. Chromatographic separation of tigecycline and tetracycline (internal standard) was achieved on a Hypersil GOLD C18 column using gradient elution. The selected reaction monitoring transitions were performed at m/z 586.1→513.2 for tigecycline and m/z 445.1→410.2 for tetracycline. The assay was linear over the concentration range of 5–2000 ng/mL. The intra‐ and interday precisions at three concentration levels (10, 100, and 1600 ng/mL) were <15% and their accuracies were within the range of 95.1–106.1%. The mean recovery ranged from 94.3 to 105.6% and the matrix effect from 92.1 to 97.6%. Tigecycline was stable under all tested conditions. This validated method was successfully applied to a pharmacokinetic study in critically ill patients. The data demonstrated that our method allows quantification of tigecycline in serum in a quick and reliable manner for widespread application. 相似文献

A family of energetic salts with high thermal stability and low impact sensitivity based on an oxygen‐containing cation, 2,4‐diamino‐1,3,5‐triazine‐6‐one, were synthesized and fully characterized by IR and multinuclear (1H, 13C) NMR spectroscopy, elemental analysis, and differential scanning calorimetry. Insights into their sensitivities towards impact, friction, and electrostatics were gained by submitting the materials to standard tests. The structures of 2,4‐diamino‐1,3,5‐triazine‐6‐one nitrate, 2,4‐diamino‐1,3,5‐triazine‐6‐one sulfate, 2,4‐diamino‐1,3,5‐triazine‐6‐one perchlorate, 2,4‐diamino‐1,3,5‐triazine‐6‐one 5‐nitrotetrazolate were determined by single‐crystal X‐ray diffraction; their densities are 1.691, 1.776, 1.854, and 1.636 g cm?3, respectively. Most of the salts decompose at temperatures over 180 °C; in particular, the salts 2,4‐diamino‐1,3,5‐triazine‐6‐one nitrate and 2,4‐diamino‐1,3,5‐triazine‐6‐one perchlorate, which decompose at 303.3 and 336.4 °C, respectively, are fairly stable. Furthermore, most of the salts exhibit excellent impact sensitivities (>40 J), friction sensitivities (>360 N), and are insensitive to electrostatics. The measured densities of these energetic salts range from 1.64 to 2.01 g cm?3. The detonation pressure values calculated for these salts range from 14.6 to 29.2 GPa, and the detonation velocities range from 6536 to 8275 m s?1; these values make the salts potential candidates for thermally stable and insensitive energetic materials. 相似文献

燃料电池阴极氧还原(ORR)催化剂目前主要以商业Pt/C为主, 其高成本和稀缺性极大地限制了燃料电池的广泛应用. 为了替代Pt/C催化剂, 廉价高效的非贵金属催化剂目前受到了广泛的研究和关注. 利用氧化石墨烯(GO)为诱导模板, 借助表面丰富的含氧官能团, 实现了Co基金属有机框架材料(MOF) (ZIF-67)在GO表面的原位生长, 构筑了ZIF-67/GO层状复合材料. 热解过程中, 石墨烯的存在有效抑制了Co纳米颗粒的团聚, 并且很好地维持了原始的层状结构. 最终获得的Co@N-C/rGO复合催化剂材料实现了活性位的高度分散, 并且具有丰富的孔结构和优异的导电性能. 在电化学性能测试中Co@N-C/rGO表现出优异的ORR性能, 其起始电位为0.96 V, 半波电位0.83 V, 远优于ZIF-67直接热解得到的Co@N-C材料, 且性能与商业Pt/C催化剂相当. 此外, Co@N-C/rGO复合催化剂还表现出良好的催化稳定性和甲醇耐受性, 显示出该材料作为燃料电池氧还原催化剂的重要潜力. 相似文献

Many plants have a high polyphenol content, which causes the matrix effect and makes the analysis of trace pesticide residues highly challenging. A common approach to improving matrix effects is to purify pesticides through the use of sorbents, but this requires a combination of multiple sorbents and extensive use. Zeolitic imidazolate framework-8 is widely used for pesticide analysis due to its high porosity, large specific surface area, and versatility. Here, we established and validated a modified quick, easy, cheap, effective, rugged, and safe method based on a zeolitic imidazolate framework-8 that was used to test the removal ability for polyphenols. To this end, 145 pesticide residues in peppermint, perilla, fennel, and mulberry leaves were analyzed by the modified method coupled with liquid chromatography with tandem mass spectrometry. The mean recoveries of all pesticide residues were in the range of 74.3–103.7%, with mean relative standard deviations ≤9.1% at spiked concentrations of 1, 10, 50, and 100 μg/kg for mulberry leaves. The limits of quantitation of the method ranged from 1 to 50 μg/kg. This study offers a reliable approach for the accurate quantitative analysis of various trace substances in the polyphenol-rich plants. 相似文献
