The International and European standards for radiation sterilization require evidence of the effectiveness of a minimum sterilization dose of 25 kGy but do not provide detailed guidance on how this evidence can be generated. An approach, designated VDmax, has recently been described and computer evaluated to provide safe and unambiguous substantiation of a 25 kGy sterilization dose. The approach has been further developed into a practical method, which has been subjected to field evaluations at three manufacturing facilities which produce different types of medical devices. The three facilities each used a different overall evaluation strategy: Facility A used VDmax for quarterly dose audits; Facility B compared VDmax and Method 1 in side-by-side parallel experiments; and Facility C, a new facility at start-up, used VDmax for initial substantiation of 25 kGy and subsequent quarterly dose audits. A common element at all three facilities was the use of 10 product units for irradiation in the verification dose experiment.

The field evaluations of the VDmax method were successful at all three facilities; they included many different types of medical devices/product families with a wide range of average bioburden and sample item portion values used in the verification dose experiments. Overall, around 500 verification dose experiments were performed and no failures were observed. In the side-by-side parallel experiments, the outcomes of the VDmax experiments were consistent with the outcomes observed with Method 1.

The VDmax approach has been extended to sterilization doses >25 and <25 kGy; verification doses have been derived for sterilization doses of 15, 20, 30, and 35 kGy. Widespread application of the VDmax method for doses other than 25 kGy must await controlled field evaluations and the development of appropriate specifications/standards.  相似文献

We used single-pair fluorescence resonance energy transfer (spFRET) measurements to characterize denatured and partially denatured states of the multidomain calcium signaling protein calmodulin (CaM) in both its apo and Ca(2+)-bound forms. The results demonstrate the existence of an unfolding intermediate. A CaM mutant (CaM-T34C-T110C) was doubly labeled with fluorescent probes AlexaFlour 488 and Texas Red at opposing globular domains. Single-molecule distributions of the distance between fluorophores were obtained by spFRET at varying levels of the denaturant urea. Multiple conformational states of CaM were observed, and the amplitude of each conformation was dependent on urea concentration, with the amplitude of an extended conformation increasing upon denaturation. The distributions at intermediate urea concentrations could not be adequately described as a combination of native and denatured conformations, showing that CaM does not denature via a two-state process and demonstrating that at least one intermediate is present. The intermediate conformations formed upon addition of urea were different for Ca(2+)-CaM and apoCaM. An increase in the amplitude of a compact conformation in CaM was observed for apoCaM but not for Ca(2+)-CAM upon the addition of urea. The changes in the single-molecule distributions of CaM upon denaturation can be described by either a range of intermediate structures or by the presence of a single unfolding intermediate that grows in amplitude upon denaturation. A model for stepwise unfolding of CaM is suggested in which the domains of CaM unfold sequentially.  相似文献

13C NMR data for a series of arylthallium trifluoroacetates (ArTlX2, X = OCOCF3) are reported and assigned. The range of carbon—thallium couplings to be expected, the dependence on the disposition of coupled nuclei, and chemical shift effects are discussed. The Tl(OCOCF3)2 group is shown to be a powerful electron withdrawing group, from both the 13C data and 19F substituent chemical shifts of the p-fluorophenyl derivative.  相似文献

The N-heterocyclic carbene (NHC) precursor, 1-methyl-3-(2-pyridinylmethyl)-1H-imidazolium tetrafluoroborate, [HCH3im(CH2py)]BF4, reacted with AgBF4 in the presence of aqueous NaOH to produce the silver complex [Ag(CH3im(CH2py))2]BF4 (1) which was then reacted with Au(tht)Cl to form the corresponding gold(I) complex, [Au(CH3im(CH2py))2]BF4 (2). Complex 2 reacted with 1 equiv of AgBF4 to produce the mixed-metal species [AuAg(CH3im(CH2py))2](BF4)2 (3). The reaction of 2 with 1 equiv of Au(tht)Cl followed by metathesis with NaBF4 produces the dimetallic gold complex [Au2(CH3im(CH2py))2](BF4)2 (4). The reaction of [Ag(CH3im(CH2py))2]BF4 (1) with 1 equiv of AgBF4 produces the trinuclear [Ag3(CH3im(CH2py))3(NCCH3)2](BF4)3 (5) complex, which appears to dissociate into a dimetallic complex in solution. Complexes 1-5 were characterized by 1H NMR, 13C NMR, UV-vis, luminescence spectroscopy, elemental analysis, mass spectrometry, and X-ray crystallography. The CH3im(CH2py) ligands in 3 are arranged in a head-to-head fashion spanning a Au-Ag separation of 3.0318(5) A with the carbene portion of the ligand remaining coordinated to the Au(I) center. In 4, the ligands are arranged in a head-to-tail fashion with an Au-Au separation of 3.1730(5) A. In 5, the ligands bridge the nearly symmetrical Ag3 triangular core with short Ag-Ag separations of 2.7765(8), 2.7832(8), and 2.7598(8) A. All of these complexes, including the ligand precursor, are intensely luminescent in solution and the solid state.  相似文献

Reported in this contribution are the synthesis, characterization, and charge transport properties of wire-like Ru2(ap)4(OPEn), where ap is 2-anilinopyridinate and OPE is -(CCC6H4)nSCH2CH2SiMe3 with n = 1 (1) and 2 (2). Scanning tunneling microscopy (STM) measurements of compound 2 inserted into a SAM of C11 thiol reveal that molecule 2 exhibits (i) the stochastic switching characteristic of wire molecules embedded in insulating SAMs and (ii) higher conductivity than the C11 thiol SAM. More importantly, analysis of the molecular electronic decay constant (beta) exhibits a decrease of at least 15% as compared to purely organic molecular analogues. Hence, the transport characteristics of molecules can be significantly improved for nanoscale electronics through the incorporation of a Ru2 fragment into conjugated backbone.  相似文献

Irradiation with UVA light of the anti-inflammatory drug diclofenac [2-(2,6-dichloroanilino)phenylacetic acid] in aqueous buffer or methanol solution leads to sequential loss of both chlorine substituents and ring closure to carbazole-1-acetic acid as the major product. Minor products result from substitution by the solvent. The photosensitizing properties of diclofenac and its major photoproduct were tested with singlet oxygen substrates and in the free radical polymerization of acrylamide. Although the major carbazole product is a weakly phototoxic agent, able to generate singlet oxygen more efficiently than diclofenac, the free radical photodechlorination process is postulated as the probable initiation step of in vivo photosensitivity responses.  相似文献

Measurements of pigment triplet-triplet absorption, pigment phosphorescence and photosensitized singlet oxygen luminescence were carried out on solutions containing monomeric bacteriochlorophylls (Bchl) c and d, isolated from green photosynthetic bacteria, and their magnesium-free and farnesyl-free analogs. The energies of the pigment triplet states fell in the range 1.29-1.34 eV. The triplet lifetimes in aerobic solutions were 200-250 ns; they increased to 280 +/- 70 microseconds after nitrogen purging in liquid solutions and to 0.7-2.1 ms in a solid matrix at ambient or liquid nitrogen temperatures. Rate constants for quenching of the pigment triplet state by oxygen were (2.0-2.5) x 10(9) M-1 s-1, which is close to 1/9 of the rate constant for diffusion-controlled reactions. This quenching was accompanied by singlet oxygen formation. The quantum yields for the triplet state formation and singlet oxygen production were 55-75% in air-saturated solutions. Singlet oxygen quenching by ground-state pigment molecules was observed. Quenching was the most efficient for magnesium-containing pigments, kq = (0.31-1.2) x 10(9) M-1 s-1. It is caused mainly by a physical process of singlet oxygen (1O2) deactivation. Thus, Bchl c and d and their derivatives, as well as chlorophyll and Bchl a, combine a high efficiency of singlet oxygen production with the ability to protect photochemical and photobiological systems against damage by singlet oxygen.  相似文献

A method based on Ge(Li) γ-ray spectrométry is applied to the determination of 224Ra (t12= 3.64 days) in natural waters. The 224Ra is first removed from several hundred liters of water by preconcentration onto manganese dioxide-impregnated acrylic fibers. The fibers are leached, radium is coprecipitated with barium sulfate, and the γ-ray activity is counted so that activity ratios among 224Ra, 225Ra and 226Ra can be calculated. Concentrations are determined by using the 226Ra concentration determined on a small separate sample. Results from samples collected from ground water, estuarine, and continental shelf environments are presented.  相似文献

Abstract. The ability of UV-irradiated African green monkey kidney cells (CV-1) to support the growth of unirradiated herpes simplex virus type 1 as measured by plaque forming ability has been investigated. The lowering of plaque formation by the virus when the host cell was irradiated was examined at thirteen different wavelengths. An action spectrum for this cellular parameter (capacity) was obtained in the wavelength region of 235–302 nm. This action spectrum points to nucleic acid as the critical target molecule for this effect.  相似文献

The reactions of the allylic peroxides ascaridole, cyclohexadiene endoperoxide, and α-phellandrene peroxide with triphenylphosphine follow an SN i'-type mechanism. In contrast, the acyclic allylic compounds, allyl tert-butyl peroxide, α-cumyl cyclohexenyl peroxide, and tert-butyl cyclohexenyl peroxide apparently react with triphenylphosphine by a free-radical mechanism. The saturated cyclic peroxide dihydroascaridole, in which there is no possibility of an allylic rearrangement, gives with triphenylphosphine a mixture of olefinic alcohols. Di-n-butyl peroxide is readily cleaved by sodium dialkyl phosphites, but sterically hindered peroxides do not react under similar conditions. Reaction can, however, take place at the oxygen adjacent to a large group if a smaller substituent is present on the other oxygen atom.  相似文献
