High-performance liquid chromatography and thin-layer chromatography assays for Devil's Club (Oplopanax horridus)

Devil's root, Oplopanax horridus, is a widely used folk medicine in Alaska and British Columbia. The inner bark of the root and stem has been used to treat colds, cough, fever, and diabetes. The present study involves the development of high-pressure liquid chromatography (HPLC) and thin-layer chromatography (TLC) methods to detect the presence of trans-nerolidol and sterols in the root bark. The HPLC and TLC analytical methods presented are suitable for the characterization and identification of Oplopanax horridus.     

Production of recombinant bleaching enzymes from thermophilic microorganisms in fungal hosts

Cost-effective production of enzymes for industrial processes makes the appropriate selection of the host-vector expression system critical. We have developed two systems for the bulk production of bleaching enzymes from thermophiles. Kluyveromyces lactis has been developed as a secretion host employing expression vectors based on the 2μ-like plasmid pKD1 of Kluyveromyces drosophilarium. Our second system involves the filamentous fungus Trichoderma reesei. Fusion and nonfusion vectors have been constructed using the strong cellobiohydrolase 1 (cbh1) promoter. The KEX2 protease cleavage site and a 6 × HIS-tag have been incorporated to facilitate both cleavage and purification of the mature foreign proteins.     

Amperometric biosensor for the rapid determination of salicylate in whole blood

A method for the determination of salicylate in whole blood is described. The assay uses salicylate hydroxylase to convert salicylate to catechol in the presence of NADH and molecular oxygen. The formation of catechol is monitored amperometrically by oxidation at +300 mV vs.Ag/AgCl and the size of the oxidation current is related to the concentration of salicylate in the sample. The reagents are incorporated into the working electrode of a disposable strip, allowing measurements to be made on a drop of blood within 1 min. The functional range of the assay can be extended to the equivalent of 7.2 mM plasma salicylate by incorporating benzoate as a component of the reaction system. The method has the advantages of simplicity and speed compared with standard procedures, and should prove especially useful in suspected overdose situations.     

Chemistry and Biology of Cylindrols: Novel Inhibitors of Ras Farnesyl-Protein Transferase from Cylindrocarpon lucidum

Farnesyl-protein transferase (FPTase) is an enzyme responsible for the farnesylation of Ras protein. Farnesylation is required for cell-transforming activity in several tumor-types, and therefore, inhibition of FPTase activity may be a potential target for anticancer drugs. Our continued search for novel inhibitors led to the isolation of a number of bicyclic resorcinaldehyde cyclohexanone derivatives named here cylindrols A(1) to A(4), cylindrols B and B(1), and a number of known compounds, from Cylindrocarpon lucidum. The compounds were isolated by bioassay-guided separation using Sephadex LH-20, silica gel, and reverse phase HPLC. Structures were elucidated by extensive application of 2D NMR and X-ray crystallography. The determination of absolute stereochemistry was accomplished by CD measurements. Chemical transformations of the most abundant compound resulted in a number of key derivatives which were critical for the evaluation of structure activity relationship. These compounds are members of ascochlorin family and showed a wide range of inhibitory activity (0.7 &mgr;M to >140 &mgr;M) against FPTase. The FPTase activity was noncompetitive with respect to both substrates. Isolation, structures, chemical transformations, and FPTase activity are discussed in detail.     

High-performance liquid chromatography systems for the separation of benzodiazepines and their metabolites

The high-performance liquid chromatographic (HPLC) retention characteristics of 21 benzodiazepine drugs and some of their metabolites have been examined on both silica and ODS-silica packing materials. Four HPLC systems have been considered and retention data are presented for the drugs on these systems. The correlation of retention data on the systems is considered with reference to the problem of identifying unknown benzodiazepines.     

The Use of a Strategic Planning Model for Health and Personal Social Services

Experience in using a mathematical model in the strategic planning of Health and Personal Social Services (HPSS) suggests some lessons that may be relevant to strategic modelling in other public services. Two attributes of the HPSS are that its final outputs cannot be measured reliably and that it is organized hierarchically with field authorities having considerable autonomy from the central authority. A model that has proved useful in this situation simulates how the field authorities respond to decisions taken at the centre on aggregate resource availabilities and describes their responses in terms of intermediate or proxy measures of service output. The model is used interactively with planners to examine policy options. It takes account of ideal patterns of service delivery, not merely current practice, and of the priorities and preferences employed by service personnel in the field.     

A mild and efficient one-step synthesis of quinolines

2-Aminoaryl ketones undergo condensation with α-methylene ketones in the presence of a catalytic amount of Y(OTf)₃ at room temperature under mild conditions to afford the corresponding polysubstituted quinolines in excellent yields.
