Abnormally mild synthesis of bis(dithiolo)pyrroles from 2,5-dimethylpyrroles

[chemical reaction: see text]. Treatment of N-substituted 2,5-dimethylpyrroles 2 with an equilibrated mixture of disulfur dichloride and DABCO in chloroform at 0 degrees C gives pentathiepinopyrroles 3 in moderate yields; further reaction of 3 with the same mixture at room temperature leads, in an extensive reaction cascade, to bis(dithiolo)pyrroles 4 in high yield; 2 can be converted into 4 in a one-pot operation under unusually mild conditions.     

Elucidation of the structure of quindolinone,a minor alkaloid of cryptolepis sanguinolenta: Submilligram 1H-13c and 1H-15N heteronuclear shift correlation experiments using micro inverse-detection

Elucidation of minor natural product structures has been significantly augmented by inverse-detection; further improvement has been afforded by the development of micro inverse-detection probes. We report here the elucidation of the structure of a new alkaloid, quindolinone (5H, 10H-indolo[3,2-b]quinolin-11-one), from the West African plant Cryptolepis sanguinolenta. All nmr data for this minor, preparative hplc-isolated alkaloid, including ¹H-¹⁵N one? bond heteronuclear shift correlation (HMQC) data, were recorded on an 800 μg sample of the alkaloid dissolved in 140 μl of 100% d₆-DMSO using a 400 MHz spectrometer.     

Grignard-Addition an Pyridin-N-oxid

Addition of phenyl-, alkyl-, alkenyl- and alkinyl-Grignard reagents to pyridine-N-oxide in THF leads to 5-substituted (2Z, 4E)-pentadien-aldoximes 6 having (E) (syn)-configuration of the C, N-double bond. The unsaturated oximes are shown to arise through an electrocyclic ring opening reaction from the primary Grignard adducts 5 . These can be trapped by protonation at low temperature.     

Unsymmetrical selenides from selenocyanates and methyl grignard

The addition of methyl Grignard to diethyl acetamido(cyanoselenobenzyl)malonates 3 and 4 at ?78° followed by hydrolysis yields the 3-(4-and 3-methylselenophenyl)alanines 1 and 2.     

The low-lying electronic states of YCu

The spectroscopic constants for the singlet and tripletstates of YCu below about 15 000 cm⁻¹ are determined using an internally contracted multireference configuration-interaction approach. These calculations are calibrated by studies of fewer states using higher levels of correlation treatment and/or basis sets. The computed T_c values and radiative lifetimes are in reasonable agreement with experiment. The calculations confirm the previous experimental assignment for all but one state, where theory helps resolve between two possible assignments.     

A graph-theoretic approach to quantitative structure—activity/reactivity studies

Characterization of molecular species based on the use of suitable graph invariants (graph paths, in particular) can provide a quantitative means of encoding structure; the technique is complementary to commoner approaches to studies of quantitative structure— activity relationships. Graph path encoding is here applied to quantitative studies of relationships between molecular structures and biological activity; the examples are the rates of various substrate reactions with hexoldnase, and the potential opiate-like activity of enkephalin analogs.     

Synthesis of 5-ethynylorotic acid

Condensation of 2,4-dimethoxy-5-iodo-6-carbomethoxypyrimidine ( 10 ) with copper (I) 3-tetrahydropyranyloxyprop-1-ynide ( 4 ) afforded 2,4-dimethoxy-5-(3′-tetrahydropyranyloxyprop-1′-yn)-6-carbomethoxypyrimidine ( 11 ), which was hydrolyzed to produce 2,4-dimethoxy-5-(3′-hydroxyprop-1′-yn)-6-carbomethoxypyrimidine ( 12 ). Oxidation of 12 with dimethyl sulfoxide-oxalyl chloride reagent gave the acetylenic aldehyde ( 13 ), which on treatment with sodium methoxide in dry tetrahydrofuran yielded 2,4-dimethoxy-5-ethynyl-6-carbomethoxypyrimidine ( 14 ). The trimethylsilyl derivative ( 15 ) was deprotected by sequential treatment with iodotrimethylsilane and aqueous sodium hydroxide, leading to the formation of 5-ethynylorotic acid ( 1 ).

The measurement of the cationic transport numbers in MgO single crystals has been performed, using the dilatocoulometric method. In the oxygen partial pressure range 1 to 10^?10 atm at temperatures of 1100 to 1300°C the ionic conduction in MgO is extrinsic.     

ADRIAMYCIN RESISTANCE IN CHINESE HAMSTER FIBROBLASTS FOLLOWING OXIDATIVE STRESS INDUCED BY PHOTODYNAMIC THERAPY

Photodynamic therapy (PDT) generates reactive oxygen species that are responsible for the initial cytotoxic events produced by this treatment. An extended (16 h) porphyrin incubation prior to light irradiation increased expression of the 75, 78 and 94 kDa glucose-regulated stress proteins (GRP), as well as the cognate form of the 70 kDa heat shock protein. However, these stress proteins were not induced following isoeffective PDT doses using a short (1 h) porphyrin incubation protocol. In the current study, Chinese hamster fibroblasts were used to examine sensitivity to adjunctive PDT and adriamycin as previous reports indicate a correlation between stress protein synthesis and a decrease in adriamycin cytotoxicity. Treatments that either induced GRP ( i.e . PDT with an extended porphyrin incubation or exposure to the calcium ionophore A23187) or did not induce GRP ( i.e . PDT with a short porphyrin incubation or UV irradiation) were followed at increasing time intervals with a 1 h adriamycin incubation. A time-dependent decrease in adriamycin cytotoxicity was observed when cells were first exposed to either of the PDT protocols or to A23187. Alterations in intracellular drug levels did not account for the change in adriamycin sensitivity. Likewise, intracellular glutathione concentrations and antioxidant enzyme activities were not significantly altered following PDT or A23187. Parameters associated with altered adriamycin sensitivity included a decrease in the percentage of S phase cells following PDT and A23187 as well as a depletion of intracellular ATP after PDT using the extended porphyrin incubation. These results demonstrate that PDT can be added to the growing list of diverse stresses producing transient resistance to adriamycin and that stress protein induction is not universally associated with all oxidative treatments inducing this resistance.     

Photometrische Antimonbestimmung in Rein- und Elektrolytkupfer

Zusammenfassung Wir zeigen einen Weg zur Bestimmung von Antimon in Rein- bzw. Reinstkupfersorten. Die Erfassungsgrenze liegt bei 0,5 g Metalleinwaage in der Größenordnung zwischen 1 und 30 g Sb/Tonne (0,0001 und 0,0030%).Das Verfahren hat den Vorzug der Schnelligkeit gegenüber allen bisher üblichen Methoden. Die Reproduzierbarkeit der gefundenen Werte kann als befriedigend bezeichnet werden, obgleich die Extinktionskurve nicht ganz geradlinig verläuft.Wenn die oben erwähnte Kupferstammlösung vorliegt, läßt sich die Bestimmung innerhalb von 30 min durchführen.
