A new test apparatus was developed for the precise measurement of mechanical properties related to the traveling performance of off-road vehicles. In particular the apparatus is capable of performing the experiment under the condition of constant contact load between the wheel and the soil, which most of the existing apparatus do not fulfill satisfactorily, and of measuring the displacement of soil particles beneath the wheel. The test results obtained by the apparatus for a rigid wheel on sand are discussed with some mechanical interpretations. 相似文献

In this study, the chemical properties of organic acids as DNA polymerase inhibitors were examined. In total, we assayed the inhibitory activities of 23 compounds. We found that the DNA synthesis activity of DNA polymerase was usually reduced to less than 50% in the presence of 100 microM monoprotic acids, which have a Clog P value greater than 7.0 and a pK(a) value less than 5.4. With a minor modification these chemical properties applied to several organic fatty acids previously reported as DNA polymerase inhibitors. Moreover, we also examined the inhibitory activities of perfluorooctadecanoic acid (PFOdA) and perfluorooctanesulfonic acid (PFOS) against DNA polymerase beta in detail. These compounds inhibited the polymerase activity of pol beta competitively with template-primer DNA, and non-competitively with dNTPs. In addition, the 8 kDa domain-defective pol beta was also sensitive to these compounds. Our results suggest that the inhibitory mode of action of PFOdA and PFOS is different from that mediated by the classic fatty acid inhibitors against DNA polymerase beta. 相似文献

A new and reliable procedure for constructing five- to seven-membered carbocycles via an endo-mode ring-closing reaction of 1-phenylsulfonylallenes with a substituent that has a terminal active methine moiety at the C1-position has been developed. Trisubstituted 1-phenylsulfonylallenes underwent a similar endo-mode ring-closing reaction to produce the corresponding five- to seven-membered carbocycles, while the formation of six- and seven-membered carbocycles from the corresponding tetrasubstituted allene was not realized. In addition, the introduction of an aromatic ring to the alkyl side chain of the starting allenes made possible the construction not only of normal-sized carbocycles but also an eight-membered framework. 相似文献

Upon exposure to a catalytic amount of [RhCl(CO)2]2 in 1,4‐dioxane, homopropargylallene‐alkynes underwent a novel cycloisomerization accompanied by the migration of the alkyne moiety of the homopropargyl functional group to produce six/five/five tricyclic compounds in good yields. A plausible mechanism was proposed on the basis of an experiment with 13C‐labeled substrate. Orientation and stretch of entangled polymers under large step shear deformations were investigated through primitive chain network simulations. In the simulations, entangled polymer dynamics is described by 3D motion of entanglements, 1D sliding of monomers along the chain, and creation/destruction of entanglements described by hooking/unhooking with surrounding chains at chain ends. In addition to the conventionally proposed relaxation mechanisms that are reptation, contour length fluctuations, and constraint release (both thermal and convective), the simulations also account for force balance among entanglement strands converging to an entanglement node, and nodes also fluctuate in space. Nonlinear step strain data for monodisperse polystyrene melts (Ferri and Greco, Macromolecules, 37:5931, 2006) were quantitatively reproduced by using the same two molecular-weight-independent parameters already adopted by Masubuchi et al. (J Non-Newt Fluid Mech, 149:87, 2008) to fit linear viscoelastic data of several monodisperse polystyrene melts. Analysis of the orientation tensor and of the chain stretch ratio indicates that the segment orientation and stretch realized in the simulation are quantitatively described by a simple three-chain model (Marrucci et al., Macromol Symp, 158:57, 2000a). 相似文献

Nateglinide is a new quick action/short duration (QRSD) type of oral blood glucose regulator, and nateglinide immediate release tablets are used for patients with mild diabetes under the trade name of Fastic((R)) tablets. In this study, we attempted to determine if it was possible to control both post-prandial blood glucose level (PBG) and fasting blood glucose level (FBG) for moderate or severe diabetes through controlled release of nateglinide. Enteric coated granules were selected for the administration form for controlled release of nateglinide, and three types of enteric coated granules were prepared having dissolution pH values of 5.5, 6.5 and 7.2. The three types of enteric coated granules were each administered separately or the enteric coated granules having an dissolution pH of 6.5 were administered simultaneous to administration of nateglinide immediate release tablets to normal beagle dogs just before feeding followed by measurement of plasma nateglinide concentration, plasma insulin concentration and blood glucose level. In the case of administering enteric coated granules alone (nateglinide: 9 mg/kg), the absorption of nateglinide was confirmed to tend to be delayed as the dissolution pH increased. In the case of an dissolution pH of 5.5, decreases in both PBG and FBG were observed. In the case of dissolution pH values of 6.5 and 7.2, only decrease in FBG was observed. In case of nateglinide immediate release tablets (nateglinide: 9 mg/kg), only decrease in PBG was observed. Decreases in both PBG and FBG were observed in the case of simultaneous administration of dissolution pH 6.5 enteric coated granules and nateglinide immediate release tablets just before feeding (nateglinide: 90 mg/head+60 mg/head). A correlation was observed between plasma nateglinide concentrations and blood glucose levels. On the other hand, there were no correlations observed between changes in plasma insulin concentrations and blood glucose levels. In case of nateglinide immediate release tablets (nateglinide: 150 mg/head), Decreases in both PBG and FBG were observed. However, the nateglinide controlled release formulation is more useful than the nateglinide immediate release tablets from the view point of avoidance of side effect, or of easy control of both PBG and FBG. On the basis of these results, the design of a controlled release formulation that contains nateglinide was suggested to enable control of both PBG and FBG for moderate and severe diabetes patients. 相似文献

Irciniasulfonic acid B (1) was obtained from a marine sponge, Ircinia sp., together with a known multi-drug resistance modulator irciniasulfonic acid (2). Spectroscopic and chemical analyses revealed structure consisting of common unsaturated fatty acids, a novel unsaturated branched fatty acid, and a taurine. Final separation of irciniasulfonic acid B (1) was achieved with its methylated derivatives. Irciniasulfonic acid B (1) reversed the multi-drug resistance in the same way as irciniasulfonic acid. 相似文献
