We report the chemical design and development of 3-aryl-substituted 7-alkoxy-4-methylcoumarins with enhanced fluorogenic properties. The 3-aryl substituents are installed via an optimized Suzuki–Miyaura cross-coupling (SMC) reaction between a 7-alkoxy-3-bromo-4-methylcoumarin and aryl boronic MIDA esters using Pd(OAc)2/XPhos in a catalytic system with K2CO3 in aqueous THF. Under these conditions, an exocyclic ester functionality is found to be unaffected. Subsequent saponification revealed a carboxylic acid functionality that is suitable for conjugation reactions. Evaluation of their fluorescence properties indicated that the installed 3-heteroaryl substituent, particularly benzofuran-2-yl, resulted in a significant red shift of both the excitation and emission wavelengths.  相似文献

Background

The construction and electrochemical response characteristics of Poly (vinyl chloride) membrane sensors for moxifloxacin HCl (MOX) are described. The sensing membranes incorporate ion association complexes of moxifloxacin cation and sodium tetraphenyl borate (NaTPB) (sensor 1), phosphomolybdic acid (PMA) (sensor 2) or phosphotungstic acid (PTA) (sensor 3) as electroactive materials.

Results

The sensors display a fast, stable and near-Nernstian response over a relative wide moxifloxacin concentration range (1 × 10-2 - 4.0 × 10-6, 1 × 10-2 - 5.0 × 10-6, 1 × 10-2 - 5.0 × 10-6 M), with detection limits of 3 × 10-6, 4 × 10-6 and 4.0 × 10-6 M for sensor 1, 2 and 3, respectively over a pH range of 6.0 - 9.0. The sensors show good discrimination of moxifloxacin from several inorganic and organic compounds. The direct determination of 400 μg/ml of moxifloxacin show an average recovery of 98.5, 99.1 and 98.6% and a mean relative standard deviation of 1.8, 1.6 and 1.8% for sensors 1, 2 and 3 respectively.

Conclusions

The proposed sensors have been applied for direct determination of moxifloxacin in some pharmaceutical preparations. The results obtained by determination of moxifloxacin in tablets using the proposed sensors are comparable favorably with those obtained using the US Pharmacopeia method. The sensors have been used as indicator electrodes for potentiometric titration of moxifloxacin.

Beta glucan (β-glucan) has promising bioactive properties. Consequently, the use of β-glucan as a food additive is favored with the dual-purpose potential of increasing the fiber content of food products and enhancing their health properties. Our aim was to evaluate the biological activity of β-glucan (antimicrobial, antitoxic, immunostimulatory, and anticancer) extracted from Saccharomyces cerevisiae using a modified acid-base extraction method. The results demonstrated that a modified acid-base extraction method gives a higher biological efficacy of β-glucan than in the water extraction method. Using 0.5 mg dry weight of acid-base extracted β-glucan (AB extracted) not only succeeded in removing 100% of aflatoxins, but also had a promising antimicrobial activity against multidrug-resistant bacteria, fungi, and yeast, with minimum inhibitory concentrations (MIC) of 0.39 and 0.19 mg/mL in the case of resistant Staphylococcus aureus (MRSA) and Pseudomonas aeruginosa, respectively. In addition, AB extract exhibited a positive immunomodulatory effect, mediated through the high induction of TNFα, IL-6, IFN-γ, and IL-2. Moreover, AB extract showed a greater anticancer effect against A549, MDA-MB-232, and HepG-2 cells compared to WI-38 cells, at high concentrations. By studying the cell death mechanism using flow-cytometry, AB extract was shown to induce apoptotic cell death at higher concentrations, as in the case of MDA-MB-231 and HePG-2 cells. In conclusion, the use of a modified AB for β-glucan from Saccharomyces cerevisiae exerted a promising antimicrobial, immunomodulatory efficacy, and anti-cancer potential. Future research should focus on evaluating β-glucan in various biological systems and elucidating the underlying mechanism of action.  相似文献

Evidence is presented for the formation of a solid phase based on the smallest fullerene, C20, in thin diamond-like carbon films deposited by ultraviolet laser ablation from diamond onto nickel substrates at room temperature in the presence of 10-4 torr of cyclohexane or benzene. Laser desorption mass spectrometry from the films shows the presence of C20, C21 and C22 species, while micro-Raman spectroscopy and electron diffraction from selected particles together with first principle density-functional calculations, indicate a cubic solid with dodecahedral C20 cages as building blocks. Unlike solid C60 and fully protonated C20, which are bound by van der Waals forces, the proposed structure is stabilized by linking of the C20 dodecahedra with bridging carbon atoms at interstitial tetrahedral sites to form a face-centered-cubic lattice with 22 carbon atoms per unit cell. Spectral studies of Co substituted Ni-Zn ferrites   总被引:1,自引:0,他引:1  
The spinel ferrites Zn0.35Ni0.65−xCoxFe2O4, 0≤x≤1, have been prepared using the standard ceramic technique. Room temperature Mössbauer, X-ray and infrared IR spectra were used for carrying out this study. X-ray patterns reveal that all the samples have single-phase cubic spinel structure. The Mössbauer spectra of the samples show a paramagnetic phase for x=0 and a six-line magnetic pattern and a central paramagnetic phase for x≥0.1. They are analyzed and attributed to two magnetic subpatterns and two quadrupole doublets due to Fe3+ ions at the tetrahedral A-sites and octahedral B-sites. Four absorption bands are observed in IR spectra. They confirm the spinel structure of the samples and existence of Fe3+ ions in the sample sublattices. The deduced hyperfine interactions, lattice parameters, absorption band positions and intensities and force constant are found to be dependent on the substitution factor x, where the cation distribution is estimated. The hyperfine magnetic fields, magnetization and lattice resonant frequency are found to be dependent on the interionic distance.  相似文献
