The reaction of protein‐bound iron–sulfur (Fe‐S) clusters with nitric oxide (NO) plays key roles in NO‐mediated toxicity and signaling. Elucidation of the mechanism of the reaction of NO with DNA regulatory proteins that contain Fe‐S clusters has been hampered by a lack of information about the nature of the iron‐nitrosyl products formed. Herein, we report nuclear resonance vibrational spectroscopy (NRVS) and density functional theory (DFT) calculations that identify NO reaction products in WhiD and NsrR, regulatory proteins that use a [4Fe‐4S] cluster to sense NO. This work reveals that nitrosylation yields multiple products structurally related to Roussin's Red Ester (RRE, [Fe2(NO)4(Cys)2]) and Roussin's Black Salt (RBS, [Fe4(NO)7S3]. In the latter case, the absence of 32S/34S shifts in the Fe?S region of the NRVS spectra suggest that a new species, Roussin's Black Ester (RBE), may be formed, in which one or more of the sulfide ligands is replaced by Cys thiolates.  相似文献

The successful application of dihydropyrido[1,2‐a]indolone (DHPI) substrates in Pd‐catalyzed asymmetric allylic alkylation chemistry facilitates rapid access to multiple alkaloid frameworks in an enantioselective fashion. Strategic bromination at the indole C3 position greatly improved the allylic alkylation chemistry and enabled a highly efficient Negishi cross‐coupling downstream. The first catalytic enantioselective total synthesis of (?)‐goniomitine, along with divergent formal syntheses of (+)‐aspidospermidine and (?)‐quebrachamine, are reported herein.  相似文献

Characterizing paramagnetic complexes in solids is an essential step toward understanding their molecular functions. However, methodologies to characterize chemical and electronic structures of paramagnetic systems at the molecular level have been notably limited, particularly for noncrystalline solids. We present an approach to obtain connectivities of chemical groups and metal-binding structures for unlabeled paramagnetic complexes by 13C and 1H high-resolution solid-state NMR (SSNMR) using very fast magic angle spinning (VFMAS, spinning speed >or=20 kHz). It is experimentally shown for unlabeled Cu(II)(Ala-Thr) that 2D 13C/1H correlation SSNMR under VFMAS provides the connectivity of chemical groups and assignments for the characterization of unlabeled paramagnetic systems in solids. We demonstrate that on the basis of the assignments provided by the VFMAS approach multiple 13C-metal distances can be simultaneously elucidated by a combination of measurements of 13C anisotropic hyperfine shifts and 13C T1 relaxation due to hyperfine interactions for this peptide-Cu(II) complex. It is also shown that an analysis of 1H anisotropic hyperfine shifts allows for the determination of electron-spin states in Fe(III)-chloroprotoporphyin-IX in solid states.  相似文献

Stable and reusable tetraorganosilicon reagents, alkenyl-, aryl-, and silyl[2-(hydroxymethyl)phenyl]dimethylsilanes, undergo 1,4-addition reactions to alpha,beta-unsaturated carbonyl acceptors under mild rhodium-catalysis. The reaction tolerates a diverse range of functional groups and is applicable to gram-scale synthesis. Use of a chiral diene ligand allows the achievement of the corresponding enantioselective transformations using the tetraorganosilicon reagents, providing the silicon-based approach to optically active ketones and substituted piperidones that serve as synthetic intermediates of pharmaceuticals. A rhodium alkoxide species is suggested to be responsible for a transmetalation step on the basis of the observed kinetic resolution of a racemic chiral phenylsilane in the enantioselective 1,4-addition reaction under the rhodium-chiral diene catalysis.  相似文献

The effect of dodecanethiolate-protected metallic nanoclusters of gold (Au:SC12, 1), silver (Ag:SC12), palladium (Pd:SC12), and platinum (Pt:SC12) on the catalytic activity of Mn(TPP)Cl (TPP = tetraphenylporphinato) was investigated in styrene oxidation with iodosylbenzene. Among the four metal clusters, only Au:SC12 led to appreciable acceleration of the catalytic reaction. The major role of the Au cluster was to regenerate the active catalytic path involving Mn(III) and Mn(V) from the deactivated Mn(IV) species. The binary 1/Mn(TPP)Cl catalyst system showed an absorption spectrum characteristic of Mn(III)-porphyrin after reaction, whereas a catalytically ineffective Mn(IV) species was observed as the sole porphyrin species in the absence of the Au cluster or in the presence of Pd, Ag, and Pt clusters. Accordingly, the slow oxidation reaction with Mn(TPP)Cl was accelerated by the addition of Au:SC12, and complete conversion of Mn(IV) into Mn(III) was observed in the absorption spectrum. 1H NMR inspection of the reaction of Au:SC12 and iodosylbenzene revealed that the surface dodecyl groups were partially oxidized into dodecanal and eliminated from the cluster surface, thereby producing unprotected gold sites on the surface. A reactivation mechanism involving the reaction of the Mn-porphyrin and the oxidant activated on the gold surface is proposed.  相似文献

Silver nanodimers with a small gap of a few nanometers aligned on glass substrates were used to enhance hyper-Raman scattering of crystal violet dye molecules. When localized surface plasmon of the dimer array was resonantly excited along the interparticle axis, hyper-Raman intensity was significantly enhanced. Moreover, the spectral appearance was slightly different between the two excitation polarizations, suggesting a possibility of two resonance contributions at one-photon and two-photon energies. Since the plasmonic property of dimer arrays can be controlled by the dimer geometry, the dimer arrays are expected to be well-defined substrates for surface-enhanced hyper-Raman spectroscopy.  相似文献

An n-propylpyridinium chloride-modified PDMS elastomeric network, PDMS/Py(+)Cl(-), was prepared from linear PDMS chains containing Si(CH(3))(2)OH end-groups cross-linked by 3-chloropropyltrimethoxysilane and posterior reaction with pyridine. PDMS/Py(+)Cl(-) material was structurally characterized by infrared spectroscopy (IR) and solid state (13)C and (29)Si NMR. Thermogravimetric analysis of the product showed good thermal stability, with the initial temperature of weight loss at 450 K. The ion-exchange capacity of the PDMS/Py(+)Cl(-) was 0.65 mmol g(-1). Metal halides, MCl(z) [M=Fe(3+), Cu(2+), and Co(2+)], were adsorbed by the modified solid from ethanol solutions as neutral species by forming the surface anionic complexes MCl(z+n)(n-). The nature of the anionic complex structure was proposed by UV-vis diffuse reflectance spectra. The species adsorbed were FeCl(-)(4), CuCl(2-)(4), and CoCl(2-)(4). The specific sorption capacities and the heterogeneous stability constants of the immobilized metal complexes were determined with the aid of computational procedures. The trend in affinities of PDMS/Py(+)Cl(-) for the metal halides were found to be FeCl(3)>CuCl(2) approximately CoCl(2).  相似文献

The number of pigments in single light-harvesting complexes (chlorosomes) were calculated by imaging single chlorosomes in a frozen buffer at cryogenic temperature with a confocal laser fluorescence microscope and pigment extraction. Chlorosomes were isolated from two types of green photosynthetic bacteria Chlorobium (Chl.) tepidum and Chloroflexus (Cfl.) aurantiacus and were individually imaged in the frozen medium. Each fluorescence spot observed mainly came from a single chlorosome and was ascribable to self-aggregates of bacteriochlorophyll (BChl) c molecules as core parts of chlorosomes. A three-dimensional distribution of fluorescence of single chlorosomes was analyzed, and the number of chlorosomes in a volume of 54,000 microm(3) was counted directly. On the basis of the results, averaged numbers of the BChl c molecules contained in a single chlorosome of Chl. tepidum and Cfl. aurantiacus were determined to be 1.4 x 10(5) and 9.6 x 10(4), respectively. The present numbers are almost comparable to those estimated by other methods (Martinez-Planells et al., Photosynth. Res. 2002, 71, 83 and Monta?o et al., Biophys. J. 2003, 85, 2560).  相似文献
