Destabilase-Lysozyme (mlDL) is a multifunctional i-type enzyme that has been found in the secretions from the salivary glands of medicinal leeches. mlDL has been shown to exhibit isopeptidase, muramidase and antibacterial activity. This enzyme attracts interest because it expresses thrombolytic activity through isopeptidolysis of the ε-(γ-Glu)-Lys bonds that cross-link polypeptide chains in stabilised fibrin. To date, three isoforms of mlDL have been identified. The enzymatic properties of pure mlDL isoforms have not yet been described because only destabilase complexes containing other proteins could be isolated from the salivary gland secretion and because low product yield from the generation of recombinant proteins has made comprehensive testing difficult. In the present study, we optimised the procedures related to the expression, isolation and purification of active mlDL isoforms (mlDL-Ds1, mlDL-Ds2, mlDL-Ds3) using an Escherichia coli expression system, and we detected and compared their muramidase, lytic, isopeptidase and antimicrobial activities. After optimisation, the product yield was 30 mg per litre of culture. The data obtained in our study led to the suggestion that the recombinant mlDL isoforms isolated from inclusion bodies form stable oligomeric complexes. Analyses of the tested activities revealed that all isoforms exhibited almost identical patterns of pH and ionic strength effects on the activities. We determined that mlDL-Ds1, 2, 3 possessed non-enzymatic antibacterial activity independent of their muramidase activity. For the first time, we demonstrated the fibrinolytic activity of the recombinant mlDL and showed that only intact proteins possessed this activity, suggesting their enzymatic nature. The recombinant Destabilase-Lysozyme isoforms obtained in our study may be considered potential thrombolytic agents that act through a mechanism different from that of common thrombolytics.  相似文献

Hydrodynamic limit of general k-nary mass exchange processes with discrete mass distribution is described by a system of kinetic equations that generalize classical Smoluchovski's coagulation equations and many other models that are intensively studied in the current mathematical and physical literature. Existence and uniqueness theorems for these equations are proved. At last, for k-nary mass exchange processes with k>2 an alternative nondeterministic measure-valued limit (diffusion approximation) is discussed.  相似文献

The paper is devoted to the well-posedness for nonlinear McKean-Vlasov type diffusions with coefficients depending on the median or, more generally, on the α-quantile of the underlying distribution. The median is not a continuous function on the space of probability measures equipped with the weak convergence. This is one reason why well-posedness of the SDE considered in the paper does not follow by standard arguments.  相似文献

Recently, rejuvenated interest to fuel cells has posed a number of problems regarding the polymer electrolyte membrane properties and their behaviour in different electrolyte solutions. This work was dedicated to study the conductivity of H+-, Fe3+- and mixed H+/Fe3+-forms of cation exchange membranes Neosepta CMS, Nafion 112, 115 and 117 and Selemion HSF under conditions similar to these in the Fe3+/Fe2+–H2/H+ fuel cell in the range of current densities 0–90 mA/cm2. It was found that the conductivities of these membranes in 1.09 M H2SO4 solution decrease in the following order: Selemion HSF › Nafion 117 ≈ Nafion 115 ≈ Neosepta CMS › Nafion 112. Conductivities of perfluorinated membranes were discussed in terms of Hsu and Gierke percolation theory [20]. The Fe3+-forms of Nafion membranes studied displayed a monotonous decline in the resistance when current increased, which is a manifestation of gradual conversion of the Fe3+-form into H+-form of these membranes. Unlike the Nafion membranes, the Fe3+-forms of Neosepta CMS and Selemion HSF membranes exhibited a sharp jump of resistance at relatively high current densities (more than 70 mA/cm2) that is most probably a result of concentration polarization.  相似文献

We consider equations of the form Bf=g, where B is a Galois connection between lattices of functions. This includes the case where B is the Fenchel transform, or more generally a Moreau conjugacy. We characterize the existence and uniqueness of a solution f in terms of generalized subdifferentials, which extends K. Zimmermann's covering theorem for max-plus linear equations.
