We describe a general photolithography-based process for the microfabrication of surface-supported Teflon AF structures. Teflon AF patterns primarily benefit from superior optical properties such as very low autofluorescence and a low refractive index. The process ensures that the Teflon AF patterns remain strongly hydrophobic in order to allow rapid lipid monolayer spreading and generates a characteristic edge morphology which assists directed cell growth along the structured surfaces. We provide application examples, demonstrating the well-controlled mixing of lipid films on Teflon AF structures and showing how the patterned surfaces can be used as biocompatible growth-directing substrates for cell culture. Chinese hamster ovary (CHO) cells develop in a guided fashion along the sides of the microstructures, selectively avoiding to grow over the patterned areas. 相似文献

Single-cell electroporation (SCEP) is a relatively new technique that has emerged in the last decade or so for single-cell
studies. When a large enough electric field is applied to a single cell, transient nano-pores form in the cell membrane allowing
molecules to be transported into and out of the cell. Unlike bulk electroporation, in which a homogenous electric field is
applied to a suspension of cells, in SCEP an electric field is created locally near a single cell. Today, single-cell-level
studies are at the frontier of biochemical research, and SCEP is a promising tool in such studies. In this review, we discuss
pore formation based on theoretical and experimental approaches. Current SCEP techniques using microelectrodes, micropipettes,
electrolyte-filled capillaries, and microfabricated devices are all thoroughly discussed for adherent and suspended cells.
SCEP has been applied in in-vivo and in-vitro studies for delivery of cell-impermeant molecules such as drugs, DNA, and siRNA,

We report that GABA(A) receptors in a patch-clamped biological cell form a short-term memory circuit when integrated with a scanning-probe microfluidic device. Laminar patterns of receptor activators (agonists) provided by the microfluidic device define and periodically update the data input which is read and stored by the receptors as state distributions (based on intrinsic multistate kinetics). The memory is discharged over time and lasts for seconds to minutes depending on the input function. The function of the memory can be represented by an equivalent electronic circuit with striking similarity in function to a dynamic random access memory (DRAM) used in electronic computers. Multiplexed biohybrid memories may form the basis of large-scale integrated biocomputational/sensor devices with the curious ability to use chemical signals including odorants, neurotransmitters, chemical and biological warfare agents, and many more as input signals. 相似文献

Radioxenon emissions of the TRIGA Mark II research reactor in Vienna were investigated with respect to a possible impact on the verification of the Comprehensive Nuclear Test-Ban-Treaty. Using the Swedish Automatic Unit for Noble Gas Acquisition (SAUNA II), five radioxenon isotopes 125Xe, 131mXe, 133mXe, 133Xe and 135Xe were detected, of which 125Xe is solely produced by neutron capture in stable atmospheric 124Xe and hence acts as an indicator for neutron activation processes. The other nuclides are produced in both fission and neutron capture reactions. The detected activity concentrations ranged from 0.0010 to 190 Bq/m³. The source of the radioxenon is not yet fully clarified, but it could be micro-cracks in the fuel cladding, fission of 235U contaminations on the outside of the fuel elements or neutron activation of atmospheric Xe. Neutron deficient 125Xe with its highly complex decay scheme was seen for the first time in a SAUNA system. In many experiments the activity ratios of the radioxenon nuclides carry the signature of nuclear explosions, if 131mXe is omitted. Only if 131mXe is included into the calculations of the isotopic activity ratios, the majority of the measurements revealed a “civil” signature (typical for a NPP). A significant contribution of the TRIGA Vienna to the global or European radioxenon inventory can be excluded. New accurate eigenvalue bounds for symmetric matrices of saddle point form are derived and applied for both unpreconditioned and preconditioned versions of the matrices. The estimates enable a better understanding of how preconditioners should be chosen. The preconditioners provide efficient iterative solution of the corresponding linear systems with, for some important applications, an optimal order of computational complexity. There exist two main versions of preconditioners of algebraic multilevel type, the additive and the multiplicative methods.
They correspond to preconditioners in block diagonal and block matrix factorized form, respectively. Both can be defined and
analysed as recursive two-by-two block methods. Although the analytical framework for such methods is simple, for many finite
element approximations it still permits the derivation of the strongest results, such as optimal, or nearly optimal, rate
of convergence and optimal, or nearly optimal order of computational complexity, when proper recursive global orderings of
node points have been used or when they are applied for hierarchical basis function finite element methods for elliptic self-adjoint
equations and stabilized in a certain way. This holds for general elliptic problems of second order, independent of the regularity
of the problem, including independence of discontinuities of coefficients between elements and of anisotropy. Important ingredients
in the methods are a proper balance of the size of the coarse mesh to the finest mesh and a proper solver on the coarse mesh.
This paper presents in a survey form the basic results of such methods and considers in particular additive methods. This
method has excellent parallelization properties.

We present a class of nested iteration schemes for solving large sparse systems of linear equations with a coefficient matrix with a dominant symmetric positive definite part. These new schemes are actually inner/outer iterations, which employ the classical conjugate gradient method as inner iteration to approximate each outer iterate, while each outer iteration is induced by a convergent and symmetric positive definite splitting of the coefficient matrix. Convergence properties of the new schemes are studied in depth, possible choices of the inner iteration steps are discussed in detail, and numerical examples from the finite-difference discretization of a second-order partial differential equation are used to further examine the effectiveness and robustness of the new schemes over GMRES and its preconditioned variant. Also, we show that the new schemes are, at least, comparable to the variable-step generalized conjugate gradient method and its preconditioned variant. 相似文献

Summary. The paper deals with eigenvalue estimates for block incomplete factorization methods for symmetric matrices. First, some
previous results on upper bounds for the maximum eigenvalue of preconditioned matrices are generalized to each eigenvalue.
Second, upper bounds for the maximum eigenvalue of the preconditioned matrix are further estimated, which presents a substantial
improvement of earlier results. Finally, the results are used to estimate bounds for every eigenvalue of the preconditioned
matrices, in particular, for the maximum eigenvalue, when a modified block incomplete factorization is used to solve an elliptic
equation with variable coefficients in two dimensions. The analysis yields a new upper bound of type for the condition number of the preconditioned matrix and shows clearly how the coefficients of the differential equation
influence the positive constant .

Hydrophobic side groups on a stimuli-responsive polymer, encapsulated within a single giant unilamellar vesicle, enable membrane attachment during compartment formation at elevated temperatures. We thermally modulated the vesicle through implementation of an IR laser via an optical fiber, enabling localized directed heating. Polymer-membrane interactions were monitored using confocal imaging techniques as subsequent membrane protrusions occurred and lipid nanotubes formed in response to the polymer hydrogel contraction. These nanotubes, bridging the vesicle membrane to the contracting hydrogel, were retained on the surface of the polymer compartment, where they were transformed into smaller vesicles in a process reminiscent of cellular endocytosis. This development of a synthetic vesicle system containing a stimuli-responsive polymer could lead to a new platform for studying inter/intramembrane transport through lipid nanotubes. 相似文献
