Single-chain variable fragment (scFv) antibodies as therapeutic agents have the potential to reduce the production cost and immunogenicity relative to monoclonal antibodies, but their monovalency and lack of a fragment crystallizable region can lead to reduced function. Multimerization is one strategy for recovering the function; however, their application is limited by the production of multimeric proteins. In our previous study, an anti-lectin-like oxidized low-density lipoprotein receptor-1 (LOX-1) scFv showed potential use in diagnosis and therapy of atherosclerotic diseases, but is limited by its inherent low antigen-binding activity. In this study, to improve the efficacy of the anti-LOX-1 scFv, we constructed the anti-LOX-1 scFv multimers by modifying the linker length between the variable domains of the scFv or by fusing the scFv with self-merization domains and expressed these scFv multimers in Brevibacillus choshinensis hosts. After optimization, all of the scFv multimers obtained efficient secretion expression. Compared with the scFv monomer, the multimers that are successfully fractionated displayed increased neutralization activity and showed elevated antigen-binding avidity, especially the tetramer, which improved the antigen avidity by two orders of magnitude. Moreover, the scFv dimer and the tetramer both displayed better stability and longer half-life in serum, which can be attractive candidates for the next-generation anti-LOX-1 therapeutic antibody.  相似文献

Poly(L-glutamic acid) (PGA)-cystamine-[gadolinium (Gd)-DO3A] was prepared in high yield with a high Gd-DO3A conjugation efficiency. Approximately 55% of the carboxylic groups in PGA were loaded with Gd-DO3A via cystamine as the spacer. Cystamine can be readily cleaved by endogenous thiols to release the Gd(III) chelates from the conjugate facilitating Gd(III) excretion after the magnetic resonance imaging (MRI). The contrast-enhanced MRI with PGA-cystamine-(Gd-DO3A) was investigated in mice bearing MDA-MB-231 breast carcinoma xenografts. PGA-1,6-hexanediamine-(Gd-DO3A), a paramagnetic polymer conjugate of a nondegradable spacer, was used as a control. Both conjugates resulted in similar contrast enhancement in the heart, vasculature, liver and kidneys in the first hour post injection. More substantial signal intensity reduction was observed for PGA-cystamine-(Gd-DO3A) in these organs than PGA-1,6-hexanediamine-(Gd-DO3A) due to release of the Gd chelates from PGA-cystamine-(Gd-DO3A) after the cleavage of the disulfide spacer by the endogenous thiols. Both conjugates resulted in similar tumor enhancement with approximately 70% increased signal intensity in the tumor periphery and 10-40% increased signal intensity in tumor interstitium. No cross-reaction was observed between PGA-cystamine-(Gd-DO3A) and human serum albumin, a plasma protein containing a cysteine residue. PGA-cystamine-(Gd-DO3A) resulted in significantly lower Gd(III) tissue retention than PGA-1,6-hexanediamine-(Gd-DO3A) 10 days after the injection in the mice (P<.05). The conjugation of Gd(III) chelates to biomedical copolymers via the degradable disulfide spacer resulted in significant contrast enhancement in the blood pool and tumor tissue but minimal long-term Gd(III) tissue retention.  相似文献

利用分离式Hopkinson压杆(SHPB),对干燥石英砂进行了被动围压下的动态压缩实验,发现石英砂表现出了明显应变率效应。通过激光粒度分析测量了动静态压缩后的试件的级配曲线,发现同一应力水平下准静态压缩比动态压缩后的试件的颗粒破碎量更大。而通过拟合相对破碎率与外力功之间的关系,发现准静态压缩在颗粒破碎方面能量利用率更高也即破碎效率更高,而这正是脆性颗粒材料应变率效应的本质原因。  相似文献

采用CVD法合成GaSb纳米线,并分析生长时间对其长度的影响,随后对其进行光学表征.实验过程中,分别采用喷金法和滴金法对硅片进行预处理,再置于相同条件下制备GaSb纳米线;之后对其进行表征分析,根据扫描电子显微镜(SEM)表征结果证实,两种方法制备的纳米线都满足VLS生长机制.且发现GaSb纳米线的生长长度,可以通过改变其生长时间来进行控制.通过该纳米线的透射电子显微镜图(TEM)、X射线衍射图(XRD)等结构表征,表明该纳米线为结晶品质优良的立方闪锌矿结构;同时,从GaSb纳米线的拉曼光谱(Raman)及光致发光谱(PL)可以反映该纳米线具有优良的光学性质.由此证明,采用CVD法制得的纳米线光学性质优异,且可以实现可控制备.  相似文献

针对超高周疲劳试件疲劳区长度设计的随意性大、要求不明确等问题,本文通过建立超高周疲劳试件的理论模型,定义了应力放大系数、疲劳区尺寸等关键设计指标参数,探索了各指标参数随试件尺寸的变化规律。结果表明,随着试件疲劳区长度的增加,试件应力放大系数反而减小,应力均匀度降低。这成为制约试件尺寸设计的主要矛盾。为了确定超高周疲劳试件的合理疲劳区长度,本文提出了超高周疲劳试件设计的约束条件和优化指标,实现了超高周疲劳试件的尺寸优化设计,得到疲劳区的合适范围为5mm~10mm。通过本文提出的尺寸优化指标求解出6063铝合金超高周试件疲劳区半长度应取6mm。  相似文献

Heterogeneous Au-Pt nanostructures have been synthesized using a sacrificial template-based approach. Typically, monodispersed Au nanoparticles are prepared first, followed by Ag coating to form core-shell Au-Ag nanoparticles. Next, the galvanic replacement reaction between Ag shells and an aqueous H(2)PtCl(6) solution, whose chemical reaction can be described as 4Ag + PtCl(6)(2-)→ Pt + 4AgCl + 2Cl(-), is carried out at room temperature. Pure Ag shell is transformed into a shell made of Ag/Pt alloy by galvanic replacement. The AgCl formed simultaneously roughens the surface of alloy Ag-Pt shells, which can be manipulated to create a porous Pt surface for oxygen reduction reaction. Finally, Ag and AgCl are removed from core-shell Au-Ag/Pt nanoparticles using bis(p-sulfonatophenyl)phenylphosphane dihydrate dipotassium salt to produce heterogeneous Au-Pt nanostructures. The heterogeneous Au-Pt nanostructures have displayed superior catalytic activity towards oxygen reduction in direct methanol fuel cells because of the electronic coupling effect between the inner-placed Au core and the Pt shell.  相似文献

In this study, a facile one-step redox polymerization method for the preparation of highly dispersed palladium (Pd)/polypyrrole (PPy) nanocapsules has been demonstrated. During the polymerizaion process, the formation of RB-PdCl(4)(2-) complex via an electrostatic interaction plays a key role for the preparation of Pd/PPy composite nanocapsules. The well-dispersed Pd nanocrystals with small sizes of 2-4 nm embedded in PPy nanocapsules exhibited a good catalytic activity during the catalytic reduction of p-nitrophenol into p-aminophenol by NaBH(4) in aqueous solution. The kinetic apparent rate constant (k(app)) was about 8.87×10(-3) s(-1). Moreover, the as-prepared Pd/PPy composite nanocapsules exhibited a good reusability, which could be repeatedly used for the reduction of p-nitrophenol with a high catalytic activity for at least 10 successive cycles.  相似文献

给出KdV-Burgers方程的有界行波解的精确级数解,采用Adomian算子分解法分别求行二个区域ζ〈0和ζ〉0的级数解,然后利用对接连续条件构成整体级数解。所得级数解能精确满足对接连续条件,并由此得到确定级数的系数递推公式,无需解非红性高阶代数方程组。与某些精确解及其它方法比较,计算简捷具在对接点处是收剑的。对某些非线性波动现象的研究,可作为计算和分析的数学依据。  相似文献
