A series of twelve anionic, cationic, and neutral nickel(II) complexes have been synthesized and characterized. The interaction of these complexes with bovine serum albumin (BSA), human serum albumin (HSA), lysozyme (Lyso), and tryptophan (Trp) has been studied using steady-state fluorescence spectroscopy. Dynamic and static quenching constants have been calculated, and the role played in quenching by the ligand and complex charge investigated. The nickel complexes showed selectivity towards the different proteins based on the environment surrounding the Trp residue(s). Only small neutral complexes with hydrophobic ligands effectively quenched protein fluorescence via static quenching, with association constants ranging from 10(2) M(-1) (free Trp) to 10(10) M(-1) (lysozyme), indicating a spontaneous and thermodynamically favorable interaction. The number of binding sites, on average, was determined to be one in BSA, HSA and free Trp, and two in lysozyme. 相似文献

The syntheses of several norbornene block copolymers containing oligonucleotide and ferrocenyl side chains and their use in the electrochemical detection of DNA are described. Two kinds of DNA-containing block copolymers with either ferrocenyl or dibromoferrocenyl groups were prepared via ring-opening metathesis polymerization (ROMP). Based on these two distinct ferrocene derivatives, a triblock copolymer labeling strategy was developed. With this strategy, the identity of DNA target can be determined by the E1/2s of the ferrocenyl moieties and the ratio of peak currents. These polymers exhibit predictable and tailorable electrochemical properties, high DNA duplex stability, and unusually sharp melting transitions, which are highly desirable characteristics for DNA detection applications. Significantly, single-base mismatches could be easily detected using two distinct block copolymers as dual-channel detection probes in an electrochemical DNA detection format. 相似文献

We critically review concepts of self‐organization and self‐assembly, and extract from this analysis consistent and workable definitions of both concepts that are applicable across many scientific disciplines. In essence, we distinguish self‐organization from self‐assembly on a thermodynamic basis, where self‐organization implies a nonequilibrium process and self‐assembly is reserved for spontaneous processes tending toward equilibrium. The general structure of violene/cyanine hybrids (see below) is exemplified by tetrakis(4-dimethylaminophenyl)ethene 1(RED) its vinylogue 2(RED) and its diazavinylogue 3(RED). As judged from their cyclic voltammograms and spectroelectrograms, oxidation occurs perfectly reversible by loss of two electrons creating closed shell systems 1-3(OX)+2 with strong bathochromic shifts (Michlers hydrol blue moieties). ESR spectra indicate only minor amounts of radical cations. At much higher potentials by another reversible loss of two electrons (-->1-3(OX)+4) the long wavelengths absorptions are replaced by shorter ones. In system 4, containing two 4-dimethylaminophenyl units only, the violene character is better preserved since oxidation occurs stepwise by single electron transfer up to 4(OX)+4. These results are backed by theoretical calculations for 1-4, demonstrating the strong geometrical differences between the various oxidation levels. Besides, new types of cyclic structures for 1-4(OX)+4 are indicated by these calculations: For systems 1-3 cyclic structures for tetracations have been found to be more stable by 3-20 kcalmol(-1) than acyclic structures, whereas for system 4 the acyclic structure is more stable by about 22 kcalmol(-1). The redox behavior of systems 1-4 is of general importance for electrochromic systems. 相似文献

The development of teacher education programs to better prepare elementary teachers requires an empirical base in which to anchor meaningful change. The research reported herein makes just such an effort. Preservice teachers' mathematics and science knowledge was measured as were their beliefs regarding instruction in these content areas. The results suggest that an increase in the number of college credit hours in science and mathematics content is less likely to effect necessary change than alteration of the methods and curriculum materials. 相似文献
