Two new syntheses of verrucarinic acid (2S, 3R-dihydroxy-3-methylpentanoic acid) and its derivatives, suitably protected for the further conversion to macrocyclic trichothecenes, are described. The first one makes use of a diastereoselective alkylation of a (?)-(S)-malic acid ester and the regioselective reductin of one carboxyl function toa methyl group. Genetic variation forms the basis for diversity but can as well be harmful and cause diseases, such as tumors. Structural variants (SV) are an example of complex genetic variations that comprise of many nucleotides ranging up to several megabases. Based on recent developments in sequencing technology it has become feasable to elucidate the genetic state of a person’s genes (i.e. the exome) or even the complete genome. Here, a machine learning approach is presented to find small disease-related SVs with the help of sequencing data. The method uses differences in characteristics of mapping patterns between tumor and normal samples at a genomic locus. This way, the method aims to be directly applicable for exome sequencing data to improve detection of SVs since specific SV detection methods are currently lacking. The method has been evaluated based on a simulation study as well as with exome data of patients with acute myeloid leukemia. This paper describes a modified method of preparation of a number of alpha-aryl-alpha-(pyridazin-3-yl)-acetonitriles via the C-arylation reaction of the corresponding carbanionsof phenylacetonitriles using 3-chloropyridazine derivatives. KOH and DMSO were used inthe deprotonation process, which made the reaction very simple and safe to perform.Nitriles were obtained in the hydrolysis reaction to the corresponding alpha-aryl-alpha-(pyridazin-3-yl)-acetamide derivatives, which were next subjected to cyclization to afford the finalproducts. A number of new derivatives of 7H,8H-pyrimido[1,6-b]pyridazin-6,8-dione weresynthesized in the cyclocondensation reaction of respective alpha-aryl-alpha-(pyridazin-3-yl)-acetamides with diethyl carbonate in the presence of EtONa. Giant unilamellar vesicles (GUVs) represent a versatile in vitro system widely used to study properties of lipid membranes and their interaction with biomacromolecules and colloids. Electroformation with indium tin oxide (ITO) coated coverslips as electrodes is a standard approach to GUV production. In the case of cationic GUVs, however, application of this approach leads to notorious difficulties. We discover that this is related to aging of ITO-coated coverslips during their repeated use, which is reflected in their surface topography on the nanoscale. We find that mild annealing of the ITO-coated surface in air reverts the effects of aging and ensures efficient reproducible electroformation of supergiant (diameter > 100 μm) unilamellar vesicles containing cationic lipids. 相似文献

A newly developed spectrometer for energy-dispersive micro X-ray fluorescence spectrometry has been designed for the demands of archaeometry. ArtTAX combines the advantages of non-destructive and sensitive multi-elemental analysis at sub-mm resolution with the possibility of working outside the laboratory. The spectrometer consists of an air-cooled, low-power molybdenum tube, new generation polycapillary X-ray optics, a silicon drift detector without the need for liquid-nitrogen cooling, a CCD camera, and three light diodes for sample positioning. The motor-driven measurement head is fixed on a x,y,z-flexible tripod support which can be assembled and dismantled within minutes. The spot size of the primary X-ray beam was determined to be 94 microm for the Cu(Kalpha) energy, the detection limits are in a range of a few tens of microg g(-1) for the medium energy-range in glass. Additional open helium purging in the excitation and detection paths enables the determination of elements down to sodium, thus avoiding vacuum conditions or a size-limiting sample chamber. A selection of qualitative and quantitative results on pigment, metal, glass, and enamel analyses are presented to show the potential of ArtTAX in the field of art and archaeology. 相似文献

The Least‐Squares Finite Element Method (LSFEM) is an interesting alternative to the standard variational principles, which are used to solve partial differential equations. Advantages of the LSFEM are its robustness and the resulting symmetric positive definite matrices, which allow the use of robust iterative solvers like the CG method. In this paper we consider the application of the LSFEM for Fluid‐Structure Interaction (FSI) problems. Our model uses the LSFEM for the discretisation of the instationary incompressible Navier‐Stokes equations, which is coupled with a standard Galerkin FEM model for a linear elastic structure.
