This paper examines an assemble-to-order environment involving a short-life-cycle product that is sold in two different configurations, each requiring a unique component that must be stocked in advance. Both configurations of the product are assembled on the same equipment which has limited capacity. The focus of the analysis is on the determination of the appropriate stocking quantities for each of the configuration specific components. Because the same assembly capacity is often reused for different instances of the component stocking problem, we treat assembly capacity as exogenous. This represents the fact that capacity decisions are often made less frequently than are procurement/production quantity decisions. We first solve for the first-best stocking policy when the components are produced internally. We then consider the case when the components are procured from external suppliers, and investigate how different forms of contract between the assembler and the component suppliers affect coordination of the supply chain as well as each party’s profit. One particularly interesting finding is that it is possible to coordinate the supply chain with a single-price contract between each supplier and the assembler while awarding all parties positive profit.  相似文献

Intramolecular 13C composition gives access to new information on the (bio) synthetic history of a given molecule. Isotopic 13C NMR spectrometry provides a general tool for measuring the position-specific 13C content. As an emerging technique, some aspects of its performance are not yet fully delineated. This paper reports on (i) the conditions required to obtain satisfactory trueness and precision for the determination of the internal 13C distribution, and (ii) an approach to determining the “absolute” position-specific 13C content. In relation to (i), a precision of <1% can be obtained whatever the molecule on any spectrometer, once quantitative conditions are met, in particular appropriate proton decoupling efficiency. This performance is a prerequisite to the measurement of isotope fractionation either on the transformed or residual compound when a chemical reaction or process is being studied. The study of the trueness has revealed that the response of the spectrometer depends on the 13C frequency range of the studied molecule, i.e. the chemical shift range. The “absolute value” and, therefore, the trueness of the 13C NMR measurements has been assessed on acetic acid and by comparison to the results obtained on the fragments from COOH and CH3 by isotopic mass spectrometry coupled to a pyrolysis device (GC-Py–irm-MS), this technique being the reference method for acetic acid. Of the two NMR spectrometers used in this work, one gave values that corresponded to those obtained by GC-Py–irm-MS (thus, the “true” value) while the other showed a bias, which was dependent to the range covered by the resonance frequencies of the molecule. Therefore, the former can be used directly for studying isotope affiliations, while the latter can only be used directly for comparative data, for example in authenticity studies, but can also be used to obtain the true values by applying appropriate correction factors. The present study assesses several key protocol steps required to enable the determination of position-specific 13C content by isotopic 13C NMR, irrespective of the NMR spectrometer: parameters to be adjusted, performance test using [1,2-13C2]acetic acid, generation of correction factors.  相似文献

An efficient and simple method for the synthesis of 1,2-dihydroquinazolines catalyzed by 4-(N,N-dimethylamino)pyridine (DMAP) from readily available aromatic or heteroaromatic aldehydes, 2-aminobenzophenone, and ammonium acetate under mild conditions is described. The purine riboswitch is a genetic regulatory element found in the 5'-untranslated regions of Gram-positive bacteria that regulates expression of the mRNA specifically in response to either guanine or adenine. We report that the adenine-responsive RNA element is also capable of specifically recognizing pyrimidine compounds bearing modifications at the 6- or 5,6-positions in a fashion similar to that of purine compounds. Using isothermal titration calorimetry and X-ray crystallography, the binding of these compounds is characterized.  相似文献

A flavin-modified gold electrode was developed in order to catalyze the electrochemical oxidoreduction of flavoproteins. Surface modification was carried out by a two-step procedure. In the first step a mixed self-assembled monolayer obtained by adsorption of activated and nonactivated 3,3'-dithiopropionic acid (free acid and N-succinimidyl ester) was formed, followed by the covalent attachment of a N(10)-hexylamino-alkylated flavin derivative via an amide bond in the second step. The electrochemical properties of the flavin-modified electrode are presented and discussed. The redox potential of the attached flavin was measured at various pH values and the electron-transfer rate constant between electrode and flavin was determined as k0 = 5 s(-1) independent of pH. The flavin-modified electrode was successfully applied to the electrochemical and spectroelectrochemical investigation of the flavoprotein WrbA from Escherichia coli that shows some structural similarities to flavodoxins. It is concluded that the electron transfer "electrode --> flavin --> flavoprotein" occurs by a two-step hopping mechanism where the first step is rate determining. Kinetic details are discussed. Furthermore, it turned out that, in contrast to flavodoxins, where the semiquinone state is stabilized, WrbA rapidly takes up two electrons, directly leading to the fully reduced form. The presented electrode surface modification may generally lend itself for spectroelectrochemical investigations of flavoproteins.  相似文献

Vacuum evaporation and molecular beam scattering experiments have been used to monitor the loss of water and dissolution of HCl and HBr in deuterated sulfuric acid at 213 K containing 0 to 100 mM hexanol. The addition of 1-hexanol to the acid creates a surface film of hexyl species. This film becomes more compact with decreasing acidity, ranging from approximately 62% to approximately 68% of maximum packing on 68 to 56 wt % D(2)SO(4), respectively. D(2)O evaporation from 68 wt % acid remains unaltered by the hexyl film, where it is most porous, but is impeded by approximately 20% from 56 and 60 wt % acid. H --> D exchange experiments further indicate that the hexyl film on 68 wt % acid enhances conversion of HCl and HBr into DCl and DBr, which is interpreted as an increase in HCl and HBr entry into the bulk acid. For this permeable hexyl film, the hydroxyl groups of surface hexanol molecules may assist uptake by providing extra sites for HCl and HBr hydrogen bonding and dissociation. In contrast, HCl --> DCl exchange in 60 wt % D(2)SO(4) at first rises with hexyl surface coverage but then drops back to the bare acid value as the hexyl species pack more tightly. HCl entry is actually diminished by the hexyl film on 56 wt % acid, where the film is most compact. These experiments reveal a transition from a porous hexanol film on 68 wt % sulfuric acid that enhances HCl and HBr uptake to one on 56 wt % acid that slightly impedes HCl and D(2)O transport.  相似文献
