Polymers were synthesized from substituted phenolic and aromatic amine compounds with hydrogen peroxide as the source of an oxidizing agent and horseradish peroxidase enzyme as the catalyst. The polymerization reaction was carried out in a monophasic organic solvent with small amounts of water at room temperature. Conditions for the synthesis of polymers with respect to reaction time and yield were studied with a number of monomers at different concentrations and in solvents with different buffers with pH range of 5.0–7.5. Physical and chemical properties of these homo-and copolymers were determined with respect to melting point, solubility, elemental analysis, molecular weight distribution, infrared absorption (including FTIR), solid-state 13C nuclear magnetic resonance, thermal gravimetric analysis, and differential scanning calorimetry. The enzyme catalyzed reactions produced polymers of molecular weight greater than 400,000 which were further fractionated by differential solubility in solvent mixtures and the molecular weight distribution of the polymer fractions were determined. In general, the polymers synthesized have low solubilities, high melting points, and some degree of branching.  相似文献

The structural determination of sn-1 and sn-2 hexadecanoic lysophosphatidylcholine (LPC) regioisomers was carried out using fast atom bombardment tandem mass spectrometry (FAB-MS/MS). The collision-induced dissociation (CID) of protonated and sodiated molecules produced diverse product ions due mainly to charge remote fragmentations. Based on the information obtained from the CID spectra of protonated and sodiated molecules, sn-1 and sn-2 hexadecanoic LPC isomers could be discriminated. Especially, the abundance ratio of the diagnostic ion pair [m/z 224/226] in the CID spectra of [M + H](+) ions was shown to be greatly different. Moreover, the CID-MS/MS spectra of sodium-adducted molecules for hexadecanoic LPC isomers showed characteristic product ions such as [M + Na - 103](+), [M + Na - 85](+), and [M + Na - 59](+), by which their regio-specificity can be differentiated.  相似文献

A nonadiabatic quantum dynamic model has been developed to study the process of electron emission from a low-work-function metal surface. The process is initiated by scattering a highly vibrationally excited NO molecule from a surface composed of a Cs layer covering a Ru crystal. The model addresses the increasing quantum yield of the electron emission as a function of the molecular vibrational excitation and incident kinetic energy. The reaction mechanism is identified as a long-range harpooning electron transfer to a molecular ion which is then accelerated toward the surface. Upon impact, the molecular ion emits its excess electron.  相似文献

The mass spectra of two series of aliphatic polynitro compounds are reported and discussed. The fragmentation patterns of aliphatic nitro and polynitro compounds are similar in that no appreciable molecular ion current is observed; however, there are several other features in the fragmentation of aliphatic polynitro compounds which differ from that of nitroalkane spectra. Both series of compounds studied-C(NO2)x(CH3)4?x, where x = 4 to 0 and C2(NO2)x(CH3)6?x, where x = 6,4,2-show a decrease in the number and intensity of alkylions with an increase in the NO+ and NO2+ ion current as x increases. The main ions resulting from the more nitrated compounds are [NO]+, [NO2]+, [CO2]+. and [CH3CO]+, whose noncharged counterparts are the principal species produced in the detonation of these compounds. This similarity of the products of the two processes suggests the use of mass spectroscopy for the investigation of the initial explosive processes. The principal fragmentation pathways of the polynitroalkanes have been elucidated by exact mass measurements and the observation of metastable ion transitions.  相似文献

Single bacterial cells, each expressing a different library variant, were compartmentalized in aqueous droplets of water-in-oil (w/o) emulsions, thus maintaining a linkage between a plasmid-borne gene, the encoded enzyme variant, and the fluorescent product this enzyme may generate. Conversion into a double, water-in-oil-in-water (w/o/w) emulsion enabled the sorting of these compartments by FACS, as well as the isolation of living bacteria cells and their enzyme-coding genes. We demonstrate the directed evolution of new enzyme variants by screening >10(7) serum paraoxonase (PON1) mutants, to yield 100-fold improvements in thiolactonase activity. In vitro compartmentalization (IVC) of single cells, each carrying >10(4) enzyme molecules, in a volume of <10 femtoliter (fl), enabled detection and selection despite the fast, spontaneous hydrolysis of the substrate, the very low initial thiolactonase activity of PON1, and the use of difusable fluorescent products.  相似文献

The micropore structure of four microporous materials (two zeolites, ZSM-5 and Y-82; an activated carbon and an alumina pillared clay) and their binary physical mixtures (50-50 wt%) have been examined by nitrogen adsorption at 77 K. Various micropore sizes have been considered from the stages on the micropore filling mechanism in the microporous materials. The application of the Dubinin-Astakhov (DA) equation to characterize and obtain the adsorption potential distributions of the microporous materials is presented.  相似文献

Solid-phase methodology for the preparation of pyrrolo[3,2-d]pyrimidine-6-carboxylates with diversity at the N3 pyrmidine nitrogen has now been elaborated to allow for the generation of pyrrolopyrimidine libraries with members possessing diversity at the N3, N5, and C6 positions. The diversification of the N5 position was achieved by treating the parent resin-bound pyrrolo[3,2-d]pyrimidines 3 with an alkyl halide in the presence of Cs2CO3 in DMF. Modification of the C6 carboxylate of resin-bound pyrrolopyrimidines 3-5 was first achieved by hydrolysis of the benzyl ester using LiOH in a mixture of THF/H2O/MeOH. Further alteration of the C6 position of resin-bound pyrrolo[3,2-d]pyrimidine-6-carboxylic acids 6-8 was then performed by activation with triphosgene and treatment with an amine to furnish resin-bound pyrrolo[3,2-d]pyrimidine-6-amides. Twenty-two pyrrolo[3,2-d]pyrimidines 1a-v with different substituents at the N3, N5, and C6 positions were obtained in yields of 21-83% and purities of 61-98% after cleavage from the solid support.  相似文献

Abstract— In the non-photoreaclivable bacterial strain E. coli B/phr-/MC2 the photoreversion of four groups of u.v.-induced mutations were investigated. They lead to resistance to Chloramphenicol (2 mg/l; "C"), Penicillin (13 or 16 mg/l; "P13" and "P16") or Streptomycin (3 mg/l; "S"). The u.v.-dose curve is concave for the C-mutations (two to three hits), about linear for P13 and S, and they reach peaks and decrease at high u.v.-doses. Though no photoreactivation of killing (PR) is present there is photoreversion of all four types of mutations (PRM). At u.v.-doses below the peaks in average about 43 per cent mutations are photoreversible. At high u.v.-doses the curves with light-post treatment (L) cross the darkcurves (D). In the photoreactivable strain B/r (by the spontaneous mutation MC2 to Mitomycin-resistance strain B/phr- was made about as u.v.-resistant as B/r is) the photoreversion of the mutation groups C, P13 and P16 (S was not investigated here) was much higher, in average about 77 per cent at low doses. It is assumed that the difference in PRM of about 34 per cent between both strains is due to a PRM-mechanism present in B/r but not in B/phr-/MC2; this mechanism may be the photoreactivating enzyme that opens thymine-dimers. The PRM in B/phr-/MC2 must then be due to a second mechanism which is probably not the dimer opening enzyme. It may be the same mechanism as in the case of mutations of phage kappa which are induced by u.v. and reversed partially by light, both extra cellularly. The premutations giving this second type of PRM may perhaps be cytosine-hydrate in the DNA. Tn average about 23 per cent mutations of B/r are photostable. Since this ratio decreases with low u.v.-doses in the C-mutations and increases in P13 and in P16 probably two types of photostable premutations seem to exist.  相似文献
