3,5-Dihalogeno-4-methoxy-N-methylpyridin-2(1H)-ones have been shown to undergo single Suzuki coupling reactions in a site-selective fashion. Monoarylations occur at the C-5 position preferentially, thus leaving the remaining C-3 halide free for further functionalization, to finally access differentially 3,5-disubstituted 2-pyridones. This two-step strategy has been applied to the elaboration of the 3-acyl-5-aryl-4-oxy-2-pyridone subunit that is prevalent in numerous bioactive natural products. [reaction: see text].  相似文献

Patients with cancer produce specific autoantibodies against protein antigens present in limited amount among a large background of immunoglobulins (Igs), nonrelevant as biomarkers, including natural antibodies. Multiple affinity protein profiling (MAPPing) that combines 2-D immunoaffinity chromatography, enzymatic digestion of the isolated proteins, and identification by MS/MS, may facilitate the identification of these so far unknown patient antibodies. The first immunoaffinity chromatography is crucial, as it is used for selectively removing proteins (autoantigens) recognized by natural antibodies. Application of this depletion step to colon cancer cell proteins is specifically described along with the identification of the natural autoantigens, as well as the coupling of this depletion step with the next steps. By enabling to separate antibody-binding proteins recognized by either natural autoantibodies or patient-specific antibodies this approach may contribute significantly towards the definition of autoantibody signatures.  相似文献

The coupling of electron and proton transfers is currently under intense scrutiny. This Communication reports a new kind of proton-coupled electron transfer within a homodinuclear first-row transition-metal complex. The triply-bridged complex [Fe(III)(μ-OPh)(μ(2)-mpdp)Fe(II)(NH(2)Bn)] (1; mpdp(2-) = m-phenylenedipropionate) bearing a terminal aminobenzyl ligand can be reversibly deprotonated to the anilinate complex 2 whose core [Fe(II)(μ-OPh)(μ(2)-mpdp)Fe(III)(NHBn)] features an inversion of the iron valences. This observation is supported by a combination of UV-visible, (1)H NMR, and M?ssbauer spectroscopic studies.  相似文献

The synthesis of β-thiolactone and β-lactam analogs of tetrahydrolipstatin is described from a common late-stage β-lactone derivative. These analogs, and a cis-disubstituted β-lactone analog of tetrahydrolipstatin, were screened for activity against porcine pancreatic lipase and for inhibition of cell growth of a panel of four human cancer lines.  相似文献

Monitoring abundance and population trends of small odontocetes is notoriously difficult and labor intensive. There is a need to develop alternative methods to the traditional visual line transect surveys, especially for low density areas. Here, the prospect of obtaining robust density estimates for porpoises by passive acoustic monitoring (PAM) is demonstrated by combining rigorous application of methods adapted from distance sampling to PAM. Acoustic dataloggers (T-PODs) were deployed in an area where harbor porpoises concurrently were tracked visually. Probability of detection was estimated in a mark-recapture approach, where a visual sighting constituted a "mark" and a simultaneous acoustic detection a "recapture." As a distance could be assigned to each visual observation, a detection function was estimated. Effective detection radius of T-PODs ranged from 22 to 104 m depending on T-POD type, T-POD sensitivity, train classification settings, and snapshot duration. The T-POD density estimates corresponded to the visual densities derived concurrently for the same period. With more dataloggers, located according to a systematic design, density estimates would be obtainable for a larger area. This provides a method suitable for monitoring in areas with densities too low for visual surveys to be practically feasible, e.g., the endangered harbor porpoise population in the Baltic.  相似文献

This paper is devoted to the persistence of periodic orbits under perturbations in dynamical systems generated by evolutionary equations, which are not smoothing in finite time, but only asymptotically smoothing. When the periodic orbit of the unperturbed system is non-degenerate, we show the existence and uniqueness of a periodic orbit (with a minimal period near the minimal period of the unperturbed problem) by using “modified” Poincaré methods. Examples of applications, including the perturbed hyperbolic Navier–Stokes equations, systems of damped wave equations and the system of second grade fluids, are given.  相似文献

We develop a numerical technique to study Anderson localization in interacting electronic systems. The ground state of the disordered system is calculated with quantum Monte Carlo simulations while the localization properties are extracted from the "Thouless conductance" g, i.e., the curvature of the energy with respect to an Aharonov-Bohm flux. We apply our method to polarized electrons in a two-dimensional system of size L. We recover the well-known universal beta(g)=dlogg/dlogL one parameter scaling function without interaction. Upon switching on the interaction, we find that beta(g) is unchanged while the system flows toward the insulating limit. We conclude that polarized electrons in two dimensions stay in an insulating state in the presence of weak to moderate electron-electron correlations.  相似文献
